Ecology of muskoxen in Jameson Land, northeast Greenland

Muskoxen Ovibos moschatus in Jameson Land exist at a density of somewhat more than 1 km⁻² of useable habitat and select moist meadows and snow bed vegetation for summer grazing and wind-exposed, dry dwarf shrub heath vegetation in winter. Graminoids dominate the winter diet and willows are the main component of the summer diet. Quality of the winter diet, as measured by the protein to fiber ratio is about one fourth that of the summer diet. During summer muskoxen supplement dietary sodium by using mineral licks. Muskoxen, especially females, retain considerable unused fat reserves through the winter and these are drawn upon during the post-calving period of lactation. Alternate year breeding is a common occurrence. Calves are frequently not weaned before the end of their first winter. Mean calf mortality is relatively low in the absence of significant predation and annual removal by hunting Inuits approaches the annual increment.     

Population Heterogeneity of Agrobacterium tumefaciens in Galls of Populus L. from a Single Nursery

This study focused on the natural crown gall infections occurring in a Leuce poplar nursery. Soil effects on crown gall frequency were detected, indicating that contamination was due to a resident Agrobacterium tumefaciens population, which was present before seedling plantation. The crown gall frequency on poplar progenies varied from 3 to 67%, indicating the feasibility of improvement in crown gall resistance. Of 129 tumor isolates, 128 were pathogenic. These isolates were of biotype 1 or 2. Biochemical, serological, and antibiotic resistance typing results concurred, indicating the presence of four biotype 1 and two biotype 2 resident subpopulations. No significant change was noticed in the relative proportions of subpopulations from one year to another. Pathogenic subpopulations both in vitro and in planta were susceptible to Kerr K84 (P. B. New and A. Kerr, J. Appl. Bacteriol. 90:172-179, 1972). In addition, no serological cross-reactions were found to occur between K84 and the pathogenic subpopulations.     

23Na NMR study of intracellular sodium ions in Dictyostelium discoideum amoeba

The intracellular sodium concentration in the amoebae from the slime mold Dictyostelium discoideum has been studied using 23Na NMR. The 23Na resonances from intracellular and extracellular compartments could be observed separately in the presence of the anionic shift reagent Dy(PPPi)7-2 which does not enter into the amoebae and thus selectively affects Na+ in the extracellular space. 31P NMR was used to control the absence of cellular toxicity of the shift reagent. The intracellular Na+ content was calculated by comparison of the intensities of the two distinct peaks arising from the intra- and extracellular spaces. It remained low (0.6 to 3 mM) in the presence of external Na+ (20 to 70 mM), and a large Na+ gradient (20- to 40-fold) was maintained. A rapid reloading of cells previously depleted of Na+ was readily measured by 23Na NMR. Nystatin, an antibiotic known to perturb the ion permeability of membranes, increased the intracellular Na+ concentration. The time dependence of the 23Na and 31P NMR spectra showed a rapid degradation of Dy(PPPi)7-2 which may be catalyzed by an acid phosphatase.     

Pertussis and cholera toxin ADP-ribosylation in Dictyostelium discoideum membranes

We report a 39 kDa substrate for cholera and pertussis toxins is present in D. discoideum membranes. This protein did not co-migrate with alpha subunits of either Gs (45 kDa and 52 kDa) or Gi (41 kDa) from control mammalian cells. The presence of GTP or its non-hydrolyzable analogs enhanced the ADP-ribosylation in response to cholera toxin, but did not significantly alter ADP-ribosylation by pertussis toxin. Divalent cations inhibited the ADP-ribosylation by both toxins. The possible association of this novel G-protein with D. discoideum adenylate cyclase may underlie some of the unique regulatory features of this enzyme. Alternatively, this G-protein may regulate one of several other cellular responses mediated by the cAMP receptor.     

Ethanol inhibits dual receptor stimulation of pineal cAMP and cGMP by vasoactive intestinal peptide and phenylephrine

Concurrent activation of vasoactive intestinal peptide and alpha 1-adrenergic receptor resulted in greater than 20-fold increases in pineal cAMP and cGMP accumulation. We now find that an intoxicating level of ethanol (0.2%, 34 mM) inhibits greater than 50% the large increases in pineal cAMP and cGMP produced by concurrent treatment with vasoactive intestinal peptide and phenylephrine. The potency of the various alcohols tested was directly related to their chain length. This inhibition appears to be specific since a five-fold higher concentration of ethanol does not inhibit the stimulation of cAMP and cGMP accumulation produced by concurrent treatment with isoproterenol and phenylephrine. Accordingly, it seems that one mechanism of action of ethanol on neural function may be its ability to selectively inhibit ethanol-sensitive integrative mechanisms which regulate cyclic nucleotides.     

Cardiac glycosides stimulate phospholipase C activity in rat pinealocytes

Ouabain and related cardiac glycosides stimulate phospholipase C activity 5-fold in rat pinealocytes. The combined treatment of ouabain and norepinephrine, which also stimulates phospholipase C, produces an additive effect. The effects of either ouabain or norepinephrine are blocked by EGTA. However, there are notable differences. The stimulatory effect of ouabain is lost when extracellular Na+ is reduced to 20 mM and is not blocked by prazosin. In contrast, the stimulatory effect of norepinephrine is not blocked when extracellular Na+ is reduced to 20 mM but is blocked by prazosin. Ouabain appears to increase phospholipase C activity through a mechanism involving inhibition of Na+,K+-ATPase, and an accumulation of intracellular Na+ and Ca2+, not involving alpha 1-adrenoceptors. These findings raise the possibility that activation of phospholipase C might be a more general effect of cardiac glycosides.     

Continuous production of L-phenylalanine by transamination

L-Phenylalanine was produced continuously from L-as-partate and phenylpyruvate by transaminase from a newly screened Pseudomonas putida strain. The process was carried out with an isolated enzyme in homogeneous phase in an enzyme membrane reactor and with immobilized whole cells in a stirred tank reactor, respectively. Due to the difference in transport resistance, the productivity of the free enzyme in homogeneous phase (72 mmol/L h) was about 3 times higher than the productivity achieved using immobilized cells. However, a better stability of the biocatalyst was observed with immobilized cells.

Pour rechercher son partenaire sexuel Zabrotes subfasciatus doit utiliser les signaux olfactifs émis soit par les mâles ou par les femelles. Le pouvoir attractif de l'odeur des mâles et des femelles sont donc étudiés et comparés par olfactométrie. Le comportement de mâles et de femelles vierges et inséminées, mis en contact des graines de la plante hôte ou non, est observé en fonction de l'âge des imagos. En aucun cas, les odeurs des mâles n'exercent une attraction sur les femelles. Les femelles vierges en absence de graines de haricot (Phaseolus vulgaris) n'exercent pas d'attraction sur les mâles vierges. Par contre les odeurs des femelles vierges mises en contact avec les graines de haricot sont très attractives pour les mâles vierges et varie en fonction de l'âge: il est plus affirmé chez les femelles plus âgées. L'émission de la phéromone sexuelle est interrompue après l'insémination des femelles mais reprend au bout d'un certain temps après celle-ci.     

The rat MIS1/Pvt-1 locus is syntenic with MYC on chromosome 7

Mouse Pvt-1 and rat MIS1 are frequent proviral integration sites in retrovirally induced lymphomas. The Pvt-1 locus is also involved in mouse plasmacytoma (6;15) and in the variant Burkitt lymphoma (2;8) translocations. We show that the Pvt-1/MIS1 locus is syntenic with MYC on rat chromosome 7. This is consistent with a postulate of close linkage and, possibly, a functional relationship between the MYC protooncogene and the MIS1/Pvt-1 locus.
