Conformational Properties of Purine-Like C-Nucleosides

Abstract

The multiplicities and chemical shifts of the 5′-hydroxyl resonances in the NMR spectra of a series of purine-like C-nucleosides reflect the presence of a hydrogen bond to N(1), and hence afford a method for assessing solution syn/anti conformational preferences.

     

Acute and chronic vitamin C deficiency in guinea-pigs: its effect on ceruloplasmin and cytochrome P 450 and b5 levels

Serum and liver ceruloplasmin levels rose markedly in guinea pigs with acute scurvy and with chronic latent scurvy. Their increase in the former condition can be attributed to the general stress reaction, but the increase in ceruloplasmin levels in concentration may have a stimulant effect on the ceruloplasmin, when the oxidation of Fe2+ to Fe3+ is potentiated, may obstruct the binding of iron to protoporphyrin and prevent formation of the haeme of cytochrome P 450 and b5.     

Epstein-Barr virus (EBV) receptors, complement receptors, and EBV infectibility of different lymphocyte fractions of human peripheral blood. II. Epstein-Barr virus studies.

In B-cell fractions isolated from human peripheral blood, the frequency of surface immunoglobulin-positive and of complement receptor-positive cells showed a good correlation with the frequency of EBV-binding cells, as detected by membrane fluorescence or by a quantitative bioassay for infectious virus in the absorbed supernatant fluid. There was a close relationship between all three parameters mentioned, the frequency of EBNA-positive cells 2 or 3 days after the infection, and the stimulation of cellular DNA synthesis. So-called O-cell fractions remaining after the removal of nylon adherent and E-rosetting cells contained a certain frequency of complement receptor-positive cells and absorbed EBV to a limited extent, but did not respond to EBV infection with EBNA induction or stimulation of DNA synthesis. None of the T-cell fractions absorbed EBV to a detectable extent. This includes the T_ea+ fraction that contained a certain proportion of complement receptor-positive cells. It is concluded that the previously demonstrated relationship between EBV receptors and complement receptors on B-lymphoblastoid lines also holds for peripheral B lymphocytes. In these cells, virus absorption is followed by an intracellular infectious process, signaled by the appearance of EBNA and cellular DNA synthesis. O cells carry complement receptors and absorb EBV to a certain extent, but do not respond with EBNA synthesis or DNA stimulation, presumably due to intracellular restrictions. T cells do not bind EBV, and the complement receptors present on some cells of the T_ea+ fraction do not function as EBV receptors.     

Whole-body protein metabolism in lactating and nonlactating women

The adaptive responses of body protein metabolism to lactation were characterized in women at 1, 5, and 12 mo postpartum and in nulliparous controls during a controlled diet of measured protein and energy intakes by nitrogen balance, a constant infusion of [13C]bicarbonate, and a primed constant infusion of [1-13C]leucine and [alpha-15N]-lysine. Dietary energy intakes in the lactating women were 27% greater than those in the nulliparous controls. Despite these differences, lactating women had significantly lower nitrogen balances compared with the nonlactating women (-4.0 +/- 37.8 vs. +44.7 +/- 30.8 mg.kg-1.day-1). No significant differences in amino acid flux, oxidation, or incorporation into protein were detected during fasting conditions in the two groups of women. However, significantly positive associations were noted between dietary intakes and the variables of protein metabolism in the lactating women. A more complete understanding of the mechanisms that regulate the disposition of dietary nutrients into maternal body stores or milk production will enhance the determination of nutrient requirements in lactating women.     

A simple method for preparing thin (10 microM) histological sections of undecalcified plastic embedded bone with implants

A simple method for preparing undecalcified thin sections of bone with implants has been developed. After exposing a surface of bone and implant in a plastic block by sawing thick sections, the surface is stained prior to making a thin section. A glass coverslip is affixed with a thin layer of cement to the stained surface to stabilize the tissue and implant during sectioning. A mixture of glycerine and water is used as a coolant and lubricant. The orientation in situ is preserved allowing demonstration of bone architecture and cells, and the tissue-implant interface.     

Der mikrochemische Nachweis der Alkaloide in der Pflanze

Zusammenfassung Es wurde eine eindeutige sichere Methode ausgearbeitet, um das Kokain in der Pflanze mikrochemisch zu fassen und zu verfolgen. Der sicherste Nachweis läßt sich erbringen durch Mikrosublimation des Kokains und Nachweis im Sublimate mit Au Cl₃ + K Br oder Extraktion mit CHCl₃ + NH₃ und Reaktion mit Au Cl₃ + K Br. Daneben lassen sich noch verwenden die Benzoesäuresublimation und die direkte Reaktion im Schnitt, die jedoch nur bei Anwesenheit von größeren Mengen Kokain gute Resultate geben.     

A simplified screening test for the diagnosis of allergy.

The Quidel allergy screen is a relatively rapid (less than 2 hours) multiallergen dipstick method for detecting specific immunoglobin E antibodies in serum. It was developed to answer the need of primary physician nonspecialists in allergy for a convenient in-office screening test for diagnosing allergy. The new test was evaluated against the benchmark diagnostic skin tests and the radioallergosorbent serologic tests for sensitivity, specificity, accuracy, and technical feasibility in an office setting. It was found that while the Quidel allergy screen lacks the specificity of the standard tests, its overall sensitivity, as defined by the percentage of patients with positive skin reactions who also tested positive with the Quidel screen (68%), its ease of use, and its rapidity warrant its consideration as a screening tool for confirming a possible case of allergy.
