Grain-mould is a major problem in grain sorghum utilization as mouldy grain has a reduced quality due to the deterioration of the endosperm and reduced embryo viability. Here, our objective was to use genome mapping to improve knowledge of genetic variation and co-variation for grain-mould incidence and other inter-related agronomic traits. Grain-mould incidence, kernel-milling hardness, grain density, plant height, panicle peduncle length, foliar-disease incidence, and plant color were measured on 125 F5 genotypes derived from a cross of Sureño and RTx430. Quantitative trait loci were detected by means of 130 mapped markers (44 microsatellites, 85 AFLPs, one morphological-trait locus) distributed among ten linkage groups covering 970 cM. One to five QTLs affected each trait, with the exception of grain density for which no QTLs were detected. Grain-mould incidence was affected by five QTLs each accounting for between 10 and 23% of the phenotypic variance. The effects and relative positions of QTLs for grain-mould incidence were in accordance with the QTL distribution of several inter-related agronomic traits (e.g., plant height, peduncle length) and with the correlation between these phenotypic traits and grain-mould incidence. The detection of QTLs for grain-mould incidence was dependent on the environment, which is consistent with heritibility estimates that show strong environmental and genotype × environment effects. Several genomic regions affected multiple traits including one region that affected grain-mould incidence, plant height, panicle peduncle length, and grain-milling hardness, and a second region that influenced grain-mould (in four environments) and plant height. One genomic region, which harbors loci for plant color, influenced the severity of foliar disease symptoms and the incidence of grain-mould in one environment. Collectively QTLs detected in the present population explained between 10% and 55% of the phenotypic variance observed for a given trait.  相似文献

We investigated whether primers able to specifically amplify a 0.7-kb DNA fragment from the conserved cpx genes could be applied to analyze A. pleuropneumoniae field isolates. The specific cpx primers were tested on 120 strains of A. pleuropneumoniae and other NAD-dependent field isolates from healthy and diseased animals to analyze A. pleuropneumoniae isolates from pigs in Brazil. We found that PCR and hybridization were able to discriminate between isolates of A. pleuropneumoniae and other bacteria. The 0.7-kb cpx DNA fragments were amplified from all 63 A. pleuropneumoniae isolates from herds with clinical symptoms and were isolated from lesions of acute cases of swine pleuropneumonia, both serotypable and nonserotypable. The PCR was also applied to 57 field isolates obtained from animals of apparently healthy herds, and the amplified cpx product was present in four serotypable and only two out of eleven A. pleuropneumoniae nonserotypable isolates. All nonserotypable A. pleuropneumoniae isolates revealed the apxA amplification pattern compatible with previously known serotypes. Some nonserotypable isolates might represent a population of isolates that originally were serotypable but lost the ability to react with serotype-specific antisera or might belong to novel serotypes. The PCR method applied is highly sensitive for serotypable A. pleuropneumoniae strains and for nonserotypable strains isolated from acute cases of swine pleuropneumoniae in Brazil. Most soils inhibit fungal germination and growth to a certain extent, a phenomenon known as soil fungistasis. Previous observations have implicated microorganisms as the causal agents of fungistasis, with their action mediated either by available carbon limitation (nutrient deprivation hypothesis) or production of antifungal compounds (antibiosis hypothesis). To obtain evidence for either of these hypotheses, we measured soil respiration and microbial numbers (as indicators of nutrient stress) and bacterial community composition (as an indicator of potential differences in the composition of antifungal components) during the development of fungistasis. This was done for two fungistatic dune soils in which fungistasis was initially fully or partly relieved by partial sterilization treatment or nutrient addition. Fungistasis development was measured as restriction of the ability of the fungi Chaetomium globosum, Fusarium culmorum, Fusarium oxysporum, and Trichoderma harzianum to colonize soils. Fungistasis did not always reappear after soil treatments despite intense competition for carbon, suggesting that microbial community composition is important in the development of fungistasis. Both microbial community analysis and in vitro antagonism tests indicated that the presence of pseudomonads might be essential for the development of fungistasis. Overall, the results lend support to the antibiosis hypothesis.  相似文献

The monoclonal antibody (MAb) Ki-67 is routinely used in clinical studies to estimate the growth fraction of tumors. However, the role of pKi-67, the protein detected by the Ki-67 MAb, remains elusive, although some biochemical data strongly suggest that it might organize chromatin. To better understand the functional organization of pKi-67, we studied its three-dimensional distribution in interphase cells by confocal microscopy and electron tomography. FluoroNanogold, a single probe combining a dense marker with a fluorescent dye, was used to investigate pKi-67 organization at the optical and ultrastructural levels. Observation by confocal microscopy followed by 3D reconstruction showed that pKi-67 forms a shell around the nucleoli. Double labeling experiments revealed that pKi-67 co-localizes with perinucleolar heterochromatin. Electron microscopy studies confirmed this close association and demonstrated that pKi-67 is located neither in the fibrillar nor in the granular components of the nucleolus. Finally, spatial analyses by electron tomography showed that pKi-67 forms cords 250-300 nm in diameter, which are themselves composed of 30-50-nm-thick fibers. These detailed comparative in situ analyses strongly suggest the involvement of pKi-67 in the higher-order organization of perinucleolar chromatin.  相似文献

Recent studies combining biochemical, molecular, and traditional morphological and ecological traits have shown that some currently recognized species of the red algal genus Porphyra are actually "form species" or "complexes" comprising several morphologically similar but genetically distinct taxa. Conflicting reports of chromosome numbers and differences in DNA sequences reported for Porphyra purpurea (Roth) C. Agardh have raised suspicion that more than one taxon has been confused under this name in the Northwest Atlantic. We have identified one of these cryptic taxa and have recently described it as a new species, Porphyra birdiae . Like P. purpurea (Roth) C. Agardh, it has an ovate to broadly elongate, foliose blade with reproductive areas segregated by a distinct line into male and female sectors. While reproductive specimens have historically been confused with P. purpurea , non-reproductive specimens of P. birdiae have been incorrectly identified as P. umbilicalis Kützing. Although P. birdiae is morphologically similar to both of these species, sequences of SSU (nuclear small subunit rRNA gene) and rbc L (plastid ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit gene) indicate that it is not closely related to either one. Based on rbc L sequences, P. birdiae is closely related to P. aestivalis Lindstrom et Fredericq, a proposed new species from Alaska.  相似文献

During aerobic degradation of naphthalene-2-sulfonate (2NS), Sphingomonas xenophaga strain BN6 produces redox mediators which significantly increase the ability of the strain to reduce azo dyes under anaerobic conditions. It was previously suggested that 1,2-dihydroxynaphthalene (1,2-DHN), which is an intermediate in the degradative pathway of 2NS, is the precursor of these redox mediators. In order to analyze the importance of the formation of 1,2-DHN, the dihydroxynaphthalene dioxygenase gene (nsaC) was disrupted by gene replacement. The resulting strain, strain AKE1, did not degrade 2NS to salicylate. After aerobic preincubation with 2NS, strain AKE1 exhibited much higher reduction capacities for azo dyes under anaerobic conditions than the wild-type strain exhibited. Several compounds were present in the culture supernatants which enhanced the ability of S. xenophaga BN6 to reduce azo dyes under anaerobic conditions. Two major redox mediators were purified from the culture supernatants, and they were identified by high-performance liquid chromatography-mass spectrometry and comparison with chemically synthesized standards as 4-amino-1,2-naphthoquinone and 4-ethanolamino-1,2-naphthoquinone.  相似文献

The Pharmacogenetics Knowledge Base (PharmGKB; http://www.pharmgkb.org/) contains genomic, phenotype and clinical information collected from ongoing pharmacogenetic studies. Tools to browse, query, download, submit, edit and process the information are available to registered research network members. A subset of the tools is publicly available. PharmGKB currently contains over 150 genes under study, 14 Coriell populations and a large ontology of pharmacogenetics concepts. The pharmacogenetic concepts and the experimental data are interconnected by a set of relations to form a knowledge base of information for pharmacogenetic researchers. The information in PharmGKB, and its associated tools for processing that information, are tailored for leading-edge pharmacogenetics research. The PharmGKB project was initiated in April 2000 and the first version of the knowledge base went online in February 2001.  相似文献

Invasive, non-native, white grubs (Coleoptera: Scarabaeidae) cause significant damage in urban landscapes. Although the lack of natural enemies in their new home is often suggested as an important factor in the establishment and spread of invasive species, the potential of incumbent generalist parasites and pathogens to delay their establishment and spread has not been explored. We compared the susceptibility of the introduced Popillia japonica and the native Cyclocephala borealis to 16 species and strains of entomopathogenic nematodes isolated from within or outside the geographic ranges of the two scarabs. We found large variation in the virulence of the species/strains of nematodes with over 50% mortality of P. japonica produced by Heterorhabditis zealandica strain X1 and H. bacteriophora strain GPS11 and of C. borealis by H. zealandica and H. bacteriophora strains KMD10 and NC1. Heterorhabditis indica and H. marelatus caused less than 20% mortality of both scarab species. When considered as a group the nematode species and strains from within and outside the geographic ranges of either P. japonica or C. borealis did not differ in virulence towards either scarab species. Dose response studies with selected nematode species and strains against P. japonica and two additional non-native species Anomala (Exomala) orientalis and Rhizotrogus majalis and the native C. borealis indicated that R. majalis was the least susceptible and P. japonica and A. orientalis were as susceptible as the native C. borealis. Heterorhabditis zealandica was significantly more virulent than any other species or strain against P. japonica with a LC50 of 272 IJs/grub. The LC30 and LC50 values for H. zealandica were also the lowest among the four nematode species/strains tested against A. orientalis and C. borealis. The LC50 values for H. zealandica and H. megidis (UK strain) were significantly lower for the native C. borealis than the introduced A. orientalis. H. zealandica also showed the highest penetration efficiency and the lowest encapsulation in P. japonica and C. borealis grubs. Results suggest that the introduction of the exotic H. zealandica into the front-line states with respect to the movement of P. japonica and A. orientalis should be explored as a tactic to delay their establishment and spread. The results also suggest that the manipulation of the indigenous H. bacteriophora populations may help in delaying spread and mitigating losses caused by the invasive grub species.  相似文献

Tropical landscapes are dominated by agroecosystems, but the potential value of agroecosystems for the survival of species is often overlooked. In agroecosystems, species conservation is especially important when functional groups such as predators are affected. In Central Sulawesi, we sampled arthropods on cocoa in a gradient of land-use intensity from extensively used forest gardens to intensively used agroforestry systems. The abundance and diversity of all arthropods did not correlate with land-use intensity, so human impact was not followed by high species losses. However, the number of species and abundance of the phytophagous arthropods increased and that of the entomophagous arthropods decreased with land-use intensity. The reduced predator–prey ratio in intensified systems can be related to their reduced species richness of shade trees and the changed microclimate (increased temperature, decreased humidity and canopy cover). In conclusion, transformation of traditional into intensified agroforestry systems had a great impact on arthropod community structure on cocoa. Since predator–prey ratios decreased with increasing land-use intensity, local farmers should have least pest problems in the traditionally diversified agroforestry systems.  相似文献
