Pantoea stewartii subsp. stewartii is the causative agent of Stewart''s wilt, a bacterial disease transmitted by the corn flea beetle mainly to sweet corn (Zea mays). In many countries, it is classified as a quarantine organism and must be differentiated from other yellow enteric bacteria frequently occurring with corn. We have created novel primers from the pstS-glmS region of P. stewartii for use in conventional PCR (cPCR) and quantitative PCR (qPCR). To facilitate rapid diagnosis, we applied matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) analysis. Using whole-cell protein extracts, profiles were generated with a Bruker microflex machine, and the bacteria classified. P. stewartii strains were clearly distinguished from strains of Pantoea agglomerans, Pantoea dispersa, and Pantoea ananatis. Dendrogram analysis of the protein profiles confirmed the score values and showed the formation of separate clades for each species. The identification achieved by MALDI-TOF MS analysis agrees with the diagnosis by specific PCR primers. The combination of both methods allows a rapid and simple identification of the corn pathogen. P. stewartii subsp. stewartii and P. stewartii subsp. indologenes are highly related and can be distinguished not only by virulence assays and indole tests but also by a characteristic pattern in the nucleotide sequence of recA.Stewart''s wilt, caused by Pantoea stewartii subsp. stewartii (synonym Erwinia stewartii) is a serious disease of sweet corn (Zea mays) that was originally described in the United States (17, 18). Its transmission depends on the corn flea beetle (Chaetocnema pulicaria), which ingests the pathogen from infected tissue and transfers the bacteria to healthy plants. The beetle is also the main niche for overwintering of P. stewartii. Direct distribution by seed transmission is also possible (3, 11) but is not considered a major source. Stewart''s wilt is also a problem on certain elite inbred maize lines used for producing hybrid field corn seed in the mideastern United States (2). According to data from the European and Mediterranean Plant Protection Organization (EPPO) about its occurrence in Europe, Stewart''s wilt was reported from but not established in Austria, Greece, Poland, Romania, and Russia. More than 60 countries place import regulations on maize seed imports from affected areas, and surveillance of traded plant material is required to prevent further distribution of the pathogen (14).Several detection methods have been described for P. stewartii, including monoclonal antibodies for enzyme-linked immunosorbent assay (ELISA) (8). For the detection of P. stewartii by PCR analysis, primer pairs derived from rRNA genes and chromosomal markers, such as regions coding for the Hrp type III secretion system (hrp) and capsular exopolysaccharide (EPS) synthesis (cps), have been published (4). These primers were derived from chromosomal regions which are also common to other bacteria. A unique DNA area of P. stewartii might therefore be better suited for the design of specific primers. Another approach, the ligase chain reaction, requires radioactively labeled primers (21). Primers complementary to cpsD (wceL) were applied for quantitative PCR (qPCR) (19). A fingerprinting analysis based on miniprimer PCR and utilizing 10-mer short oligonucleotides combined with modified Taq polymerase has been reported (22). The signal intensity of PCRs is often affected by inhibitory plant components in the extracts. Thus, low levels of P. stewartii may not be detected. A collective drawback of PCR-based identification approaches is the detection of DNA from nonviable cells and traces of residual nucleic acids. This could lead to the rejection of safe seed lots. A method involving culturing of bacteria extracted from plants, lysis, and subsequent PCR analysis and named bio-PCR was established to ensure the detection of only viable bacterial populations (16). Screening of individual colonies from a plate with mixed cultures by PCR to verify reisolation of the pathogen is tedious and needs another fast and reliable method. Strains of Pantoea stewartii subsp. indologenes cause leaf spot on foxtail millet (Setaria italica) and pearl millet (Pennisetum americanum) or rot of Ananas comosus, and one strain was isolated from a diseased cluster bean (Cyamopsis tetragonolobus) (10). It is important to distinguish between P. stewartii subsp. stewartii and P. stewartii subsp. indologenes, since only the stewartii subspecies causes Stewart''s wilt.Furthermore, some bacterial isolates might not be unambiguously identified with PCR and with phytopathological methods. The recent successful identification of Erwinia isolates with matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) analysis profiling of protein patterns from whole cells (15) induced us to apply this method for the detection of P. stewartii and its differentiation from Pantoea agglomerans and other Pantoea species.  相似文献

Complex I (NADH:ubiquinone oxidoreductase) has a central function in oxidative phosphorylation and hence for efficient ATP production in most prokaryotic and eukaryotic cells. This huge membrane protein complex transfers electrons from NADH to ubiquinone and couples this exergonic redox reaction to endergonic proton pumping across bioenergetic membranes. Although quinone reduction seems to be critical for energy conversion, this part of the reaction is least understood. Here we summarize and discuss experimental evidence indicating that complex I contains an extended ubiquinone binding pocket at the interface of the 49-kDa and PSST subunits. Close to iron–sulfur cluster N2, the proposed immediate electron donor for ubiquinone, a highly conserved tyrosine constitutes a critical element of the quinone reduction site. A possible quinone exchange path leads from cluster N2 to the N-terminal β-sheet of the 49-kDa subunit. We discuss the possible functions of a highly conserved HRGXE motif and a redox–Bohr group associated with cluster N2. Resistance patterns observed with a large number of point mutations suggest that all types of hydrophobic complex I inhibitors also act at the interface of the 49-kDa and the PSST subunit. Finally, current controversies regarding the number of ubiquinone binding sites and the position of the site of ubiquinone reduction are discussed.  相似文献

The present study shows that small admixtures of one chlorophyll a (Chla) molecule per several hundred lipid molecules have strong destabilizing effect on lipid bilayers. This effect is clearly displayed in the properties of the Lα-HII transformations and results from a Chla preference for the HII relative to the Lα phase. Chla disfavors the lamellar liquid crystalline phase Lα and induces its replacement with inverted hexagonal phase HII, as is consistently demonstrated by DSC and X-ray diffraction measurements on phosphatidylethanolamine (PE) dispersions. Chla lowers the Lα-HII transition temperature (42 °C) of the fully hydrated dipalmitoleoyl PE (DPoPE) by ∼ 8 °C and ∼ 17 °C at Chla/DPoPE molar ratios of 1:500 and 1:100, respectively. Similar Chla effect was recorded also for dielaidoyl PE dispersions. The lowering of the transition temperature and the accompanying significant loss of transition cooperativity reflect the Chla repartitioning and preference for the HII phase. The reduction of the HII phase lattice constant in the presence of Chla is an indication that Chla favors HII phase formation by decreasing the radius of spontaneous monolayer curvature, and not by filling up the interstitial spaces between the HII phase cylinders. The observed Chla preference for HII phase and the substantial bilayer destabilization in the vicinity of a bilayer-to-nonbilayer phase transformation caused by low Chla concentrations can be of interest as a potential regulatory or membrane-damaging factor.  相似文献

Selected biological effects of 1,4-naphthoquinone, menadione (2-methyl-1,4-naphthoquinone) and structurally related quinones from natural sources - the 5-hydroxy-naphthoquinones juglone, plumbagin and the 2-hydroxy-naphthoquinones lawsone and lapachol - were studied in human keratinocytes (HaCaT). 1,4-naphthoquinone and menadione as well as juglone and plumbagin were highly cytotoxic, strongly induced reactive oxygen species (ROS) formation and depleted cellular glutathione. Moreover, they induced oxidative DNA base damage and accumulation of DNA strand breaks, as demonstrated in an alkaline DNA unwinding assay. Neither lawsone nor lapachol (up to 100 μM) were active in any of these assays. Cytotoxic and oxidative action was paralleled by stimulation of stress signaling: all tested quinones except lawsone and lapachol strongly induced phosphorylation of the epidermal growth factor receptor (EGFR) and the related ErbB2 receptor tyrosine kinase. EGFR activation by plumbagin, juglone and menadione was attenuated by a superoxide dismutase mimetic, indicating that ROS-related mechanisms contribute to EGFR activation by these naphthoquinones.  相似文献

Altered Bone Morphogenetic Protein (BMP) signaling leads to multiple developmental defects, including brachydactyly and deafness. Here we identify chondroitin synthase 1 (CHSY1) as a potential mediator of BMP effects. We show that loss of human CHSY1 function causes autosomal-recessive Temtamy preaxial brachydactyly syndrome (TPBS), mainly characterized by limb malformations, short stature, and hearing loss. After mapping the TPBS locus to chromosome 15q26-qterm, we identified causative mutations in five consanguineous TPBS families. In zebrafish, antisense-mediated chsy1 knockdown causes defects in multiple developmental processes, some of which are likely to also be causative in the etiology of TPBS. In the inner ears of zebrafish larvae, chsy1 is expressed similarly to the BMP inhibitor dan and in a complementary fashion to bmp2b. Furthermore, unrestricted Bmp2b signaling or loss of Dan activity leads to reduced chsy1 expression and, during epithelial morphogenesis, defects similar to those that occur upon Chsy1 inactivation, indicating that Bmp signaling affects inner-ear development by repressing chsy1. In addition, we obtained strikingly similar zebrafish phenotypes after chsy1 overexpression, which might explain why, in humans, brachydactyly can be caused by mutations leading either to loss or to gain of BMP signaling.  相似文献

Summary This article proposes saddlepoint approximations to the expectation and variance–covariance function of multitype age‐dependent branching processes. The proposed approximations are found accurate, easy to implement, and much faster to compute than by simulating the process. Multiple applications are presented, including the analyses of clonal data on the generation of oligodendrocytes from their immediate progenitor cells, and on the proliferation of Hela cells. New estimators are also constructed to analyze clonal data. The proposed methods are finally used to approximate the distribution of the generation, which has recently found several applications in cell biology.  相似文献

Alpha-synuclein (αS) is a 140-amino-acid protein that is involved in a number of neurodegenerative diseases. In Parkinson's disease, the protein is typically encountered in intracellular, high-molecular-weight aggregates. Although αS is abundant in the presynaptic terminals of the central nervous system, its physiological function is still unknown. There is strong evidence for the membrane affinity of the protein. One hypothesis is that lipid-induced binding and helix folding may modulate the fusion of synaptic vesicles with the presynaptic membrane and the ensuing transmitter release. Here we show that membrane recognition of the N-terminus is essential for the cooperative formation of helical domains in the protein. We used circular dichroism spectroscopy and isothermal titration calorimetry to investigate synthetic peptide fragments from different domains of the full-length αS protein. Site-specific truncation and partial cleavage of the full-length protein were employed to further characterize the structural motifs responsible for helix formation and lipid-protein interaction. Unilamellar vesicles of varying net charge and lipid compositions undergoing lateral phase separation or chain melting phase transitions in the vicinity of physiological temperatures served as model membranes. The results suggest that the membrane-induced helical folding of the first 25 residues may be driven simultaneously by electrostatic attraction and by a change in lipid ordering. Our findings highlight the significance of the αS N-terminus for folding nucleation, and provide a framework for elucidating the role of lipid-induced conformational transitions of the protein within its intracellular milieu.  相似文献
