A simple method for electron microscopic preparation of plant protoplasts is described. The main problems in preparing these fragile protoplasts for electron microscopy have been cell collapse due to steep gradients between protoplasts and fixatives and unacceptable loss of material during the many steps of the procedure. These problems may be solved by immobilization of the protoplasts in calcium alginate beads. The free diffusion properties of this gel prevent steep gradients. The beads also simplify handling and prevent loss of material. Protoplasts isolated from hypocotyls of rape, Brassica napus (var. Niklas), have been used as a model system. Transmission electron microscopy of the immobilized protoplasts osmotically stabilized with glucose demonstrated adequate structural and ultrastructural preservation. 相似文献

Isolation in a 55% yield of the low molecular weight pancreatic secretory trypsin inhibitor was achieved by gel filtration of an acid extract of whole inactive rat pancreas juice on Sephadex G-50 at pH 2.5 followed by desalting and ion-exchange chromatography on SP Sephadex C-50 at pH 4.5. Two distinct chromatographic fractions were obtained, labeled fraction 1 and 2. Fractions 1 and 2 showed three, respectively two, distinct closely migrating cationic bands on gel electrophoresis in barbital buffer, pH 8.6. Each fraction demonstrated one band on polyacrylamide disc electrophoresis at pH 4.6. The inhibitor is homogenous on gel filtration and on the basis of its stoichiometry with active site titrated rat anionic trypsin. Its molecular weight is approx. 6024. The amino acid composition is included. Rat pancreatic secretory trypsin inhibitor is trypsin-specific and interacts on a 1:1 molar basis with rat trypsin. It is good inhibitor of bovine trypsin but a poor inhibitor of human cationic trypsin and its binding to trypsin is reversible by acidification. Like other inhibitors of this sort, it is present in about 0.1–0.2% of the total protein content of the juice, and normally exists in its free form. A simple procedure for the production of antiserum to the inhibitor which is a poor antigen is also described. 相似文献

Exposure of mouse epidermal cell cultures to β-retinyl acetate (RA) affects a number of parameters presumed to be important in chemical carcinogenesis. (1) RA alters the course of differentiation of the epidermal cells in culture resulting in a reduced rate of cell death which normally follows cellular maturation during the first two weeks in culture. The extended life span of the cultures appeared due to prolonged survival of cells and not to increased growth rate since RA inhibited the rate of cellular proliferation. This inhibition took place only after completion of a full cell cycle in the presence of RA. (2) DNA repair in response to physical and chemical agents was quantitatively unaffected in the presence of RA. (3) The activity of constitutive aryl hydrocarbon hydroxylase (AHH) was slightly decreased after exposure to RA but the level of enzyme induced by benz[a]anthracene was strongly reduced to 20% of the controls. (4) In the presence of RA, binding of 7,12-dimethylbenz[a]anthracene to epidermal cell DNA was markedly decreased. In a previous paper [(1976) Anal. Biochem.74, 282] we reported a procedure for the analysis of 25-hydroxycholecalciferol (25-OH-D3) in cow plasma or serum by high performance liquid chromatography. Since then, we obtained synthetic 25-hydroxyergocalciferol (25-OH-D3) and found that it had the same retention time as 25-OH-D3 and interfered with the 25-OH-D3 analysis. We also encountered a relatively small number of samples that did not give sufficiently clean chromatograms by our procedure. In long-lived species, any negative effect of pollution on adult survival may pose serious hazards to breeding populations. In this study, we measured concentrations of various organochlorines (OCs) (polychlorinated biphenyl and OC pesticides) in the blood of a large number of adult glaucous gulls (Larus hyperboreus) breeding on Bjørnøya (Bear Island) in the Norwegian Arctic, and modelled their local survival using capture–recapture analysis. Survival was negatively associated with concentrations of OCs in the blood. The effect of OCs was nonlinear and evident only among birds with the highest concentrations (the uppermost deciles of contamination). The threshold for depressed survival differed between the sexes, with females being more sensitive to contamination. For birds with lower OC concentration, survival was very high, i.e. at the upper range of survival rates reported from glaucous and other large gull species in other, presumably less contaminated populations. We propose two non-exclusive explanations. First, at some threshold of OC concentration, parents (especially males) may abandon reproduction to maximize their own survival. Second, high contamination of OC may eliminate the most sensitive individuals from the population (especially among females), inducing a strong selection towards high-quality and less sensitive phenotypes. 相似文献

The ontogeny of the skeleton of the nassellarian radiolarian Amphimelissa setosa begins with the formation of three radial bars of silica; these correspond to the medial bar and spines L1 and L2 in the adult. By a precise sequence of the addition of various siliceous arcs and spines, this basic structure develops into the eucephalus. A new growth mechanism, in which existing skeletal elements serve as templates for the formation of new skeletal members, is hypothesized to explain the observed developmental patterns. The early ontogeny of nassellarian skeletons can be used to determine which skeletal features develop earliest. Among the structural characteristics used by radiolarian systematists, these conservative features are most suitable for use in interpretation of the taxonomic relationships and phylogeny of the group. 相似文献
