BACKGROUND: Poor transduction of the ciliated airway epithelium and inefficient airway delivery of viral vectors are common difficulties encountered in lung gene therapy trials with large animals and humans. METHODS: We delivered a helper-dependent adenovirus vector, incorporating a human epithelial cell-specific expression cassette, to rabbit lung. An intratracheal device was used to aerosolize a moderate dose of virus (5 x 10(11) particles), mixed with the enhancing agent LPC (L-alpha-lysophosphatidylcholine), directly into the airways. Lung mechanics, body weight and temperature, transgene expression and histopathology were studied at day 5. RESULTS: Transgene expression was seen in the epithelium of large and small airways, from trachea to terminal bronchioles, with a strong tendency toward the right lung. All cell types of the surface epithelium were transduced. Extensive transduction of the epithelium (66% of cells in trachea) was obtained using virus formulated in isotonic 0.1% LPC, while virus formulated in 0.01% LPC transduced fewer cells (24% in trachea). A transient decrease in dynamic lung compliance was observed immediately following aerosol delivery. Fever and mild-to-moderate patchy pneumonia without edema were also observed. CONCLUSION: These data demonstrate a strategy for efficient and effective transduction of airway epithelium in a large animal. 相似文献

PURPOSE OF REVIEW: Rupture of an atherosclerotic plaque is the predominant underlying event in the pathogenesis of acute coronary syndromes and stroke. While ruptured plaques are morphologically well described, the precise molecular mechanisms involved in plaque rupture are still incompletely understood. Over the last few years, techniques like microarray, suppression subtractive hybridization and differential display enabled us to study complex gene expression profiles that occur during the process of atherogenesis. In this review we focus on recent large-scale gene expression profiles performed on whole mount vascular specimens. RECENT FINDINGS: The gene expression profiles on whole mount vascular tissue confirmed that at least three mechanisms are involved in plaque rupture: (1) a disturbed balance in extracellular matrix turnover, (2) disturbed regulation of cell turnover and (3) processes involved in lipid metabolism. Animal models exhibiting features of plaque rupture reflect the involvement of these three mechanisms. The most dramatic mouse phenotypes were observed after interventions in at least two of these mechanisms. SUMMARY: The observation of plaque rupture in recent mice models is indicative of the multifactorial process of plaque rupture. This multifactorial character of plaque rupture suggests that interventions may be most effective when they influence more than one mechanisms at a time. 相似文献

Detritus derived from the filamentous cyanobacterium ,Oscillatorialimnetica was fed to Daphnia galeataDetritus supported growthand reproduction comparable to that on the green alga Scenedesmusobliquus. The live filaments of O.limnetica were, however, oflower food quality. Biochemical parameters of these food types,thought to be important in Daphnia nutrition, were also determined.It is concluded that detritus can be a good quality food forDaphnia, making eutrophic lakes better food environments thangenerally assumed.

In this paper we describe the life history consequences of feeding Daphnia galeata with different food types in different concentrations. We fed the animals with four concentrations of two green algae Scenedesmus obliquus and Chlamydomonas globosa, given separately as well as in a 1:1 mixture. Growth, reproduction and condition of the animals and consequently the intrinsic rate of population increase, r, increased with the concentration of the food for all three experimental series. The life history parameters measured were lowest in the animals raised on C. globosa. Animals fed on S. obliquus showed higher values for r. We observed a distinct synergistic effect of the two algal species: growth, reproduction, condition and r were significantly higher for the animals raised on the mixed media as compared to the animals raised on the mono-algal media. Summary Superfusion with Pb2+ induces a slow, noninactivating and reversible inward current in voltage-clamped N1E-115 neuroblastoma cells. The amplitude of this inward current increases in the range of 1–200 m Pb2+. Single-channel patch-clamp experiments have revealed that this inward current is mediated by discrete ion channels. Reversal potentials from linearI–V relationships are close to 0 mV for whole-cell and single-channel currents and the single-channel conductance amounts to 24 pS. The Pb2+-induced membrane current is not mediated by various known types of ion channels, since it is not blocked by external tetrodotoxin, tetraethylammonium,d-tubocurarine, atropine, ICS 205-930 and by internal EGTA. In Na+-free solutions superfusion with Pb2+ neither evokes a whole-cell inward current, nor single-channel openings. At –80 mV the open-time distribution of the single channels activated by 1m Pb2+ is dual exponential with time constants of 17 and 194 msec. When the Pb2+ concentration is increased from 1 to 20 m these time constants decrease to 2 and 13 msec, but the amplitude of single-channel currents remains –1.9 nA. Cd2+ and Al3+ induce inward currents and single-channel openings similar to Pb2+. Time constants fitted to the open-time distribution of single channels are 14 and 135 msec in the presence of 1 m Cd2+ and 15 and 99 msec in the presence of 50 m Al3+. Conversely, Cu2+ induces an irreversible inward current in neuroblastoma cells. Single-channel openings are undetected in the presence of Cu2+ and in Na+-free solutions Cu2+ is still able to induce an inward current. It is concluded that Pb2+, Cd2+ and possibly Al3+ activate a novel type of metal ionactivated (MIA) channel in N1E-115 cells. 相似文献

Effects of Cd2+, Co2+, Pb2+, Fe2+ and Mg2+ (1-100 microM) on single-channel properties of the intermediate conductance Ca(2+)-activated K+ (CaK) channels were investigated in inside-out patches of human erythrocytes in a physiological K+ gradient. Cd2+, Co2+ and Pb2+, but not Fe2+ and Mg2+, were able to induce CaK channel openings. The potency of the metals to open CaK channels in human erythrocytes follows the sequence Pb2+, Cd2+ > Ca2+ > or = Co2+ > Mg2+, Fe2+. At higher concentrations Pb2+, Cd2+ and Co2+ block the CaK channel by reducing the opening frequency and the single-channel current amplitude. The potency of the metals to reduce CaK channel opening frequency follows the sequence Pb2+ > Cd2+, Co2+ > Ca2+, which differs from the potency sequence Cd2+ > Pb2+, Co2+ > Ca2+ to reduce the unitary single-channel current amplitude. Fe2+ reduced the channel opening frequency and enhanced the two open times of CaK channels activated by Ca2+, whereas up to 100 microM Mg2+ had no effect on any of the measured single-channel parameters. It is concluded that the activation of CaK channels of human erythrocytes by various metal ions occurs through an interaction with the same regulatory site at which Ca2+ activates these channels. The different potency orders for the activating and blocking effects suggest the presence of at least one activation and two blocking sites. A modulatory binding site for Fe2+ exists as well. In addition, the CaK channels in human erythrocytes are distinct from other subtypes of Ca(2+)-activated K+ channels in their sensitivity to the metal ions. 相似文献

Between November 1970 and November 1972 nine sampling trips were carried out in the Friesian Lake District. Fourteen lakes were sampled. The chemical composition of the lake water, chlorophyll-a content and the species composition and population densities of zooplankton and benthos were determined simultaneously. Results are discussed in relation to the hydrology of the Lakes system and the morphometry of the individual lakes. The lakes showed a high degree of similarity. 相似文献

In this study, the effect of the food concentration on severallife history parameters of Daphnia galeata, Daphnia cucullataand the hybrid between these two species was examined. The foodconcentration had a significant effect on both growth and reproductionof all three taxa. Mortality was highly species specific, andto a lesser extent dependent on the food concentration. Changesin life history parameters resulted in changes in the intrinsicrate of population increase (r). The food dependence of thedifferent species was found to be different. Daphnia cucullatahad a relatively constant and low r. The r values for the hybridvaried most, with low values compared with the parental specieswhen grown at low food levels and relatively high values athigher food concentrations. Our results suggest that when foodconcentrations are high the hybrid will have a selective advantageover both parental species, but that at lower food levels D.galeatawill have an advantage over both D.cucullata and the hybrid. 相似文献
