Homogenization of geographical variants at the nontranscribed spacer of rDNA in Drosophila mercatorum 总被引:2,自引:0,他引:2
rDNA nontranscribed spacer (NTS) lengths of Drosophila mercatorum have been
measured in individuals from several geographic regions. Individuals from
the different geographic subpopulations share some length fragments but are
in general distinct. The length differences, both within and between
individuals, arise from different copy numbers of a 250-bp repeating unit
that is localized to one part of the NTS. In addition to the length
differences caused by the 250-bp repeat, there is a Y chromosome
(male)-specific length variant elsewhere in the NTS that is approximately
70 bp shorter than the NTS fragment from the X chromosome. Sexual
dimorphism seems to be present in all Drosophila. Also, D. mercatorum has
fewer NTS length variants per individual than does D. melanogaster while
possessing comparable levels of restriction- site polymorphism. The
mechanisms that may cause this pattern of variation are selection, gene
conversion, and unequal recombination.

Resorufin acetate is shown to be an attractive substrate to use with chymotrypsin since the absorbance of the product is several times more intense than that formed by the widely usedp-nitrophenyl acetate. Furthermore, under the right conditions, resorufin acetate allows convenient observation of the burst reaction by conventional spectrophotometry. The steady-statekcatvalues for chymotrypsin-catalyzed hydrolysis of resorufin acetate andp-nitrophenyl acetate are virtually the same, as expected for a rate-limiting deacylation step involving an identical intermediate from both substrates. Stopped-flow studies show that the maximal bursts of product from both substrates are again (in molar terms) about the same. When chymotrypsin is presented with a mixture of both substrates, the monitoring of reaction with resorufin acetate (at 571 nm) is not interfered with by simultaneous hydrolysis ofp-nitrophenyl acetate. Under these conditions,p-nitrophenyl acetate is shown to increase the burst rate constant for acylation of the enzyme by resorufin acetate, demonstrating unequivocally thatp-nitrophenyl acetate can bind to chymotrypsin elsewhere than in the active site. 相似文献

The computational method of constrained constructive optimization was used to generate complex arterial model trees by optimization with respect to a target function. Changing the target function also changes the tree structure obtained. For a parameterized family of target functions a series of trees was created, showing visually striking differences in structure that can also be quantified by appropriately chosen numerical indexes. Blood transport path length, pressure profile, and an index for relative segment orientation show clear dependencies on the optimization target, and the nature of changes can be explained on theoretical grounds. We report the nucleotide sequence of a cloned cDNA, pMTS-3, that contains a
1-kb insert corresponding to mouse thymidylate synthase (E.C. 2.1.1.45).
The open reading frame of 921 nucleotides from the first AUG to the
termination codon specifies a protein with a molecular mass of 34,962
daltons. The predicted amino acid sequence is 90% identical with that of
the human enzyme. The mouse sequence also has an extremely high degree of
similarity (as much as 55% identity) with prokaryotic thymidylate synthase
sequences, indicating that thymidylate synthase is among the most highly
conserved proteins studied to date. The similarity is especially pronounced
(as much as 80% identity) in the 44-amino-acid region encompassing the
binding site for deoxyuridylic acid. The cDNA sequence also suggests that
mouse thymidylate synthase mRNA lacks a 3' untranslated region, since the
termination codon, UAA, is followed immediately by a poly(A) segment.

Bispecific monoclonal antibodies (BsMAbs) prepared by somatic cell fusion bind monovalently to their targets and yet are extremely potent enhancers of target cell lysis by relevant effector cells. The mechanisms underlying this efficiency are not known. To investigate this property, we studied the ability of selected antibodies to modulate potentiation of tumor lysis by a bispecific antibody (CL158) which targets Fc gamma RIII-expressing cells, via the 3G8 epitope, to malignant cells expressing CA19-9 antigen. Antibodies directed against the 3G8 and B73.1 epitopes of Fc gamma RIII efficiently inhibited BsMAb-mediated SW948 tumor cell lysis by interleukin-2 (IL-2)-activated lymphocytes (PBLs). Unexpectedly, Leu 19 antibody reversed antibody-dependent but not antibody-independent lysis of 51Cr-labeled SW948 cells by IL-2-activated PBLs in a concentration-dependent fashion. Leu 19 binds to CD56, a neural cell adhesion molecule (N-CAM) isoform expressed by large granular lymphocytes (LGLs). The effects of Leu 19 on bispecific antibody promotion of lysis were due to competition for binding to the 3G8 epitope of Fc gamma RIII and led to inhibition of binding between LGLs and SW948 cells. Leu 19 did not inhibit antibody-dependent lysis by the monospecific, bivalent IgG2a variant of CA19-9 antibody. These studies show that competition assays can be useful in dissecting the relevant mechanisms underlying BsMAb-promoted lysis. Steric constraints between effector cell trigger molecules (i.e., Fc gamma RIII) and CAM such as N-CAM may regulate the function of these molecules. Understanding the roles of diverse CAM in this phenomenon will facilitate efforts to expand and use defined effector cell populations with maximal lytic potential and to identify potentially responsive tumor phenotypes. 相似文献
