Electron microscopy of negatively stained preparations of phosphorylase a (tetrameric form) from solutions with and without added protamine revealed three characteristic types of particle images. In preparations with added protamine several types of crystalline formation were observed: three types of plane monolayers, tubes and small three-dimensional crystals. The tubes have been studied by optical diffraction and filtering. In the main class of tubes the particles form three-start helices (it may well be that narrow tubes with different parameters exist). Now and again, one can observe tubes twice as large in diameter, and tubes with two-layer walls. Analysis of the images of particles in solution and in crystalline formations showed that their structure can be characterized in terms of one model consisting of four elongated bent subunits arranged with the point-group symmetry 222 at the vertices of a tetrahedron. The structure of phosphorylase b particles, previously studied (Kiselev et al., 1971), can also be characterized satisfactorily by the same model. The difference in the structure of these two forms expresses itself in a different character of mutual aggregation of the molecules in plane layers, and in the parameters of the helical packing of the molecules in tubes. 相似文献

Experiments were conducted on anesthetized cats with microelectrode recording to study the synaptic responses that develop in the lumbar motoneurons on stimulation of the afferent fibers of groups II and III in the nerves of the ipsilateral and contralateral forelegs. Stimulation of these afferents evoked predominantly inhibitory postsynaptic potentials (IPSP) in the extensor motoneurons and excitatory postsynaptic potentials (EPSP) in the flexor motoneurons. A basically inhibitory change in the rhythmic background activity developed under the influence of descending impulsation. The duration of the total inhibition of "spontaneous" motoneuron activity corresponded to the duration of the inhibitory influences exerted by the forelimb flexor-reflex afferents (FRA) on the interneurons. The interaction of the descending and segmental PSP resulted in inhibition and facilitation of the segmental responses in the motoneurons. The ultimate result of this interaction was determined by the shifts in the membrane potential of the motoneuron and by the effects created in the interneurons.I. P. The cos-site of lambda phage from pHC79 cosmide is transferred to DNA from M13 mp18 phage. The recombinant DNA thus obtained (MC18) is efficiently packaged into lambda proteins in vitro. The BamHI-HindIII fragment of pGP588 (a pBR322 derivatives containing fragment of human DNA) is subcloned into MC18. Although this pGP588 fragment contains numerous Alu repeats, no essential rearrangements of the insert were revealed. The efficiency infection by recombinant DNA packaged with lambda proteins is about 1 X 10(5) pfu/microgram DNA, whereas in the similar conditions the efficiency of lambda EMBL3A was 1 X 10(6) pfu/microgram. It is assumed that the MC vectors might be suitable for cloning and sequencing large fragments either with cohesive or blunt ends. By means of small angle X-ray scattering, an aggregation of beef pancreas Trp-tRNA synthetase (EC 6.1.1.2) was observed at physiological temperatures. A Trp-tRNA synthetase preparation which is homogeneous after PAGE in beta-ME-SDS was found to be heterogeneous in particle sizes even at low (4-8 degrees C) temperature. At heating up to 30-45 degrees C, the oligomer sizes increased as well as its proportion depending on the incubation time and temperature; very large aggregates were observed 10 times exceeding the sizes of initial particles. Cooling to 20 degrees C caused no disaggregation due to disulphide bond formation between associated subunits of Trp-tRNA synthetase. A hypothesis is proposed that the aggregation of bovine Trp-tRNA synthetase evaluated in vitro and not observed earlier with any aminoacyl-tRNA synthetases of unicellular organisms might serve as one of the mechanisms of its compartmentation in pancreas. 相似文献

A method for suppression of virus reproduction in cells using fatty acylated antiviral antibodies, which in contrast to non-modified antibodies are capable of intracellular penetration, has been suggested. The addition of stearoylated antiviral antibodies to influenza A/Chili virus-infected cells causes a 100-fold suppression of virus reproduction.
