Computerized tomography (CT) was used to monitor the exact anatomical location and dimensions of the cryosurgical iceball within the brain. The gross and microscopic appearance of the tissue iceball was examined in both acute and chronic animals. Iceball formation was monitored in the brain of four dogs under a general anesthesia. The radiographic image of the iceball was that of a well-demarcated radiolucent sphere that disappeared upon thawing. The post-thaw contrast-enhanced CT scan revealed a zone of blood-brain barrier breakdown extending no more than 1 mm beyond the maximum diameter that the iceball had attained. Histological examination demonstrated a sharp transition from frankly necrotic brain within the iceball to the normal cytoarchitecture of the surrounding neuropil. The safety and efficacy of a neurosurgical ablative procedure depends on the precision with which it can be generated. The use of CT imaging to monitor the formation of the cryosurgical iceball offers the neurosurgeon a means to precisely control the size of the ablative lesion. Small deeply situated brain tumors can be incorporated into the iceball under direct CT observation, thereby ensuring the completeness of the cryoablation while minimizing damage to the surrounding brain.  相似文献

Hybridization has many and varied impacts on the process of speciation. Hybridization may slow or reverse differentiation by allowing gene flow and recombination. It may accelerate speciation via adaptive introgression or cause near‐instantaneous speciation by allopolyploidization. It may have multiple effects at different stages and in different spatial contexts within a single speciation event. We offer a perspective on the context and evolutionary significance of hybridization during speciation, highlighting issues of current interest and debate. In secondary contact zones, it is uncertain if barriers to gene flow will be strengthened or broken down due to recombination and gene flow. Theory and empirical evidence suggest the latter is more likely, except within and around strongly selected genomic regions. Hybridization may contribute to speciation through the formation of new hybrid taxa, whereas introgression of a few loci may promote adaptive divergence and so facilitate speciation. Gene regulatory networks, epigenetic effects and the evolution of selfish genetic material in the genome suggest that the Dobzhansky–Muller model of hybrid incompatibilities requires a broader interpretation. Finally, although the incidence of reinforcement remains uncertain, this and other interactions in areas of sympatry may have knock‐on effects on speciation both within and outside regions of hybridization.  相似文献

Background: Range expansion often results in colonisation bottlenecks that should both deplete genetic diversity and increase genetic differentiation towards the margins of a species' geographic distribution.

Aims: We tested whether genetic differentiation increased among populations of the annual plant Mercurialis annua after its colonisation of the Iberian Peninsula from Morocco. Previous work showed that this colonisation resulted in a decrease of phenotypic and genetic diversity from the core in North Africa towards the distribution margins of M. annua in north-eastern and north-western Spain.

Methods: Seeds were sampled from 20 populations located across the hexaploid range of M. annua. Patterns of phenotypic and genetic differentiation among experimentally grown populations were analysed and compared between the Iberian Peninsula and North Africa.

Results: The level of phenotypic and genetic differentiation among populations in the expanded range of the Iberian Peninsula was similar to that in the core range in North Africa.

Conclusions: Our findings imply that the observed effects of range expansion on genetic differentiation may be independent of the effects on genetic diversity. They point to the importance of taking both historic and contemporary processes of migration into account when predicting the results of range expansion.  相似文献

The objectives of this study were to optimize a sensitive high-performance liquid chromatography (HPLC) method for fatty acid (FA) analysis for the quantification of polyunsaturated FAs (PUFAs) in cell lipid extracts and to analyze the lipid and FA patterns of three cell lines used in blood-brain barrier (BBB) models: RBE4, ECV304, and C6. Thin-layer chromatographic analysis revealed differences in the phosphatidylcholine-phosphatidylethanolamine (PC:PE) ratios and the triglyceride (TG) content. The PC:PE ratio was <1 for RBE4 cells but >1 for ECV304 and C6 cells. ECV304 cells displayed up to 9% TG depending on culture time, whereas the other cell lines contained about 1% TG. The percentages of docosahexaenoic acid were 9.4 +/- 1.7% of the unsaturated FAs in RBE4 cells (n = 5; 4 d in culture; 9.9% after 10 d), 8.1 +/- 2.0% in ECV304 cells (n = 11; 10 to 14 d), and 6.7 +/- 0.6% in C6 cells (n = 6; 10 to 14 d) and were close to the published values for rat brain microvascular endothelium. The percentage of arachidonic acid (C20:4) was about half that in vivo. ECV304 cells contained the highest fraction of C20:4, 17.8 +/- 2.2%; RBE4 cells contained 11.6 +/- 2.4%; and C6 cells 15.8 +/- 1.9%. It is concluded that a sensitive HPLC method for FAs is now optimized for the analysis of long-chain PUFAs. The results provide a useful framework for studies on the effects of lipid modulation and give reference information for the development of further BBB models.  相似文献

Homoploid hybrid speciation occurs through stabilization of a hybrid segregate (or segregates) isolated by premating and/or postmating barriers from parent taxa. Theory predicts that ecological and spatial isolation are of critical importance during homoploid hybrid speciation, and all confirmed homoploid hybrid species are ecologically isolated from their parents. Until recently, such species have been identified long after they originated, and consequently it has not been possible to determine the relative importance of spatial and ecological isolation during their origin. Here we present evidence for the recent origin (within the past 300 years) of a new homoploid hybrid species, Senecio squalidus (Asteraceae), in the British Isles, following long-distance dispersal of hybrid material from a hybrid zone between S. aethnensis and S. chrysanthemifolius on Mount Etna, Sicily, Italy. Historical records show that such hybrid material from Sicily was introduced to the Oxford Botanic Garden in Britain in the early part of the 18th century and that S. squalidus began to spread from there after approximately 90 years. A survey of randomly amplified polymorphic DNA/intersimple sequence repeats (RAPD/ISSR) marker variation demonstrated that S. squalidus is a diploid hybrid derivative of S. aethnensis and S. chrysanthemifolius that grow at high and low altitudes, respectively, on Mount Etna and that form a hybrid zone at intermediate altitudes. Senecio squalidus contained 11 of 13 RAPD/ISSR markers that were recorded at high frequency in S. chrysanthemifolius but were absent or occurred at low frequency in S. aethnensis, and 10 of 13 markers for which the reverse was true. Bayesian admixture analysis showed that all individuals of S. squalidus surveyed were of mixed ancestry with relatively high mean proportions of ancestry derived from both S. chrysanthemifolius and S. aethnensis (0.644 and 0.356, respectively). We argue that long-distance isolation of hybrid material from its parents on Mount Etna would have helped favor the origin and establishment of S. squalidus in the British Isles, regardless of whether the initial hybrid material introduced to Britain was preadapted to local conditions.  相似文献

Plague impacts prairie dogs (Cynomys spp.), the endangered black-footed ferret (Mustela nigripes) and other sensitive wildlife species. We compared efficacy of prophylactic treatments (burrow dusting with deltamethrin or oral vaccination with recombinant “sylvatic plague vaccine” [RCN-F1/V307]) to placebo treatment in black-tailed prairie dog (C. ludovicianus) colonies. Between 2013 and 2015, we measured prairie dog apparent survival, burrow activity and flea abundance on triplicate plots (“blocks”) receiving dust, vaccine or placebo treatment. Epizootic plague affected all three blocks but emerged asynchronously. Dust plots had fewer fleas per burrow (P < 0.0001), and prairie dogs captured on dust plots had fewer fleas (P < 0.0001) than those on vaccine or placebo plots. Burrow activity and prairie dog density declined sharply in placebo plots when epizootic plague emerged. Patterns in corresponding dust and vaccine plots were less consistent and appeared strongly influenced by timing of treatment applications relative to plague emergence. Deltamethrin or oral vaccination enhanced apparent survival within two blocks. Applying insecticide or vaccine prior to epizootic emergence blunted effects of plague on prairie dog survival and abundance, thereby preventing colony collapse. Successful plague mitigation will likely entail strategic combined uses of burrow dusting and oral vaccination within large colonies or colony complexes.  相似文献

Glomalean mycorrhizal fungi from tropical Australia   总被引:6,自引:0,他引:6  
 A comparison of different methods for isolation of vesicular-arbuscular mycorrhizal (VAM) fungi into open-pot cultures was undertaken as part of a study of the diversity of these fungi. Four different isolation techniques using spores separated from soil, soil trap cultures, root samples, or transplanted seedlings grown in intact soil cores were used to obtain as many fungi as possible from each site. Isolation methods were compared using paired samples from the same locations within natural (savanna, rocky hill, wetland, rainforest) and disturbed (minesite) habitats in a seasonally dry tropical region in the Northern Territory of Australia. There were large differences in (i) the efficiency (rate of increase in mycorrhizal colonisation), (ii) the proportion of successful cultures, (iii) fungal diversity (number of fungal species in each culture) and (iv) specificity (identity of species isolated) between these four procedures. However, the less-efficient procedures generally resulted in a higher proportion of cultures of one fungus, which could be used without further isolation steps. Most species of Scutellospora, Acaulospora and Gigaspora were obtained primarily from field-collected spores, but only 50% of these culture attempts were successful. Spores from these initial cultures produced mycorrhizas much more rapidly and successfully when used to start second-generation cultures. Several species of fungi, rarely recovered as living spores from field soils, were dominant in many trap cultures started from soil or roots. Most of these fungi were Glomus species, that were first distinguished by colonisation patterns in roots and eventually identified after sporulation in second- or third-generation trap cultures. These experiments demonstrated that glomalean fungi in the habitats sampled belonged to two functional categories, based on whether or not spores were important propagules. The "non-sporulating" fungi were dominant in many trap cultures, which suggests that these fungi had higher total inoculum levels in soils than other fungi. Pot-culturing methods provided additional information on fungal diversity which complemented spore occurrence data obtained using the same soil samples and provided valuable new information about the biology of these fungi.
