According to the World Health Organization, cataracts account for half of the blindness in the world, with the majority occurring in developing countries. A cataract is a clouding of the lens of the eye due to light scattering of precipitated lens proteins or aberrant cellular debris. The major proteins in the lens are crystallins, and they are extensively deamidated during aging and cataracts. Deamidation has been detected at the domain and monomer interfaces of several crystallins during aging. The purpose of this study was to determine the effects of two potential deamidation sites at the predicted interface of the betaA3-crystallin dimer on its structure and stability. The glutamine residues at the reported in vivo deamidation sites of Q180 in the C-terminal domain and at the homologous site Q85 in the N-terminal domain were substituted with glutamic acid residues by site-directed mutagenesis. Far-UV and near-UV circular dichroism spectroscopy indicated that there were subtle differences in the secondary structure and more notable differences in the tertiary structure of the mutant proteins compared to that of the wild type betaA3-crystallin. The Q85E/Q180E mutant also was more susceptible to enzymatic digestion, suggesting increased solvent accessibility. These structural changes in the deamidated mutants led to decreased stability during unfolding in urea and increased precipitation during heat denaturation. When simulating deamidation at both residues, there was a further decrease in stability and loss of cooperativity. However, multiangle-light scattering and quasi-elastic light scattering experiments showed that dimer formation was not disrupted, nor did higher-order oligomers form. These results suggest that introducing charges at the predicted domain interface in the betaA3 homodimer may contribute to the insolubilization of lens crystallins or favor other, more stable, crystallin subunit interactions. 相似文献

A unique family of ribonucleases was identified by exhaustive screening of genomic and cDNA libraries using a probe derived
from a gene encoding a ribonuclease stored in the mouse eosinophil secondary granule. This family contains at least 13 genes,
which encode ribonucleases, and two potential pseudogenes. The conserved sequence identity among these genes (∼70%), as well
as the isolation/purification of these ribonucleases from eosinophil secondary granules, has led us to conclude that these
genes form a unique clade in the mouse that we have identified as the Ear (Eosinophil-associated ribonuclease) gene family. Analyses of the nucleotide substitutions that have occurred among these ribonuclease genes reveal
that duplication events within this family have been episodic, occurring within three unique periods during the past 18 ×
106 years. Moreover, comparisons of non-synonymous (Ka) vs. synonymous (Ks) rates of nucleotide substitution show that although these genes conserve residues necessary for RNase activity, selective
evolutionary pressure(s) exist such that acquired amino acid changes appear to be advantageous. The selective advantage of
these amino acid changes is currently unclear, but the occurrence of this phenomenon in both the mouse and the human highlights
the importance of these changes for Ear and, therefore, eosinophil effector function(s).

This study of population structure of two intertidal monodontine species: Diloma nigerrima and Diloma subrostrata, revealed the level of genetic connectedness among populations. Despite their markedly different geographic ranges (D. nigerrima is a geographically widespread species, inhabiting both Chile and New Zealand, including its subantarctic islands, whereas D. subrostrata is endemic to New Zealand), both species are believed to possess short-lived lecithotrophic larval stages. Polymorphic DNA microsatellite sequences were used to reveal the level of genetic connectedness among populations, thus inferring the two species' relative effective dispersal abilities. For each species F statistics, AMOVA values and the strength of the relationship between geographic and genetic distance were calculated. We observed a higher within-species level of genetic variation (ΦST = 0.099 vs. ΦST = 0.016) and a higher proportion of variance (11.15% vs. 0%) among populations of D. nigerrima than of D. subrostrata. A larger fraction of significant FST values was observed among D. nigerrima population pairs (65%) than among D. subrostrata population pairs (33%). Significant correlation between genetic and geographic distance was observed for D. nigerrima but not for D. subrostrata, but this relationship was not consistent among pairwise D. nigerrima population comparisons and PCA analysis confirms that, for each species, population structure does not follow a consistent pattern of increasing with geographic distance. The lack of population structure among D. subrostrata populations is probably due to its ubiquitous distribution, meaning little opportunity exists for genetic structure. D. nigerrima, by contrast has a patchier distribution, which allows for greater opportunities for genetic differentiation to occur. We argue that, despite the probable short larval stage in this species, the lack of geographical pattern in the genetic structure found in D. nigerrima is best explained by occasional dispersal over relatively short distances around the coast of New Zealand, over longer distances from New Zealand to the subantarctic islands and even across the Pacific Ocean from New Zealand to Chile. 相似文献

The aim of the study was to examine the effects of an elevated dietary cobalt supply to dairy cows on rumen fermentation parameters and microbial vitamin B12 synthesis in the rumen. Five lactating dairy cows fitted with a ruminal and a duodenal cannula were subsequently fed either a ration containing only the native cobalt content (0.17 mg Co/ kg DM) or a ration supplemented with cobalt sulphate (0.29 mg Co/kg DM). The pH-value, the ammonia concentration as well as the concentration and the molar proportions of short chain fatty acids in the rumen were not significantly influenced by feeding the ration with the higher cobalt content. While there was no difference in microbial protein flow, the cobalamin flow at the duodenum was significantly elevated in supplemented animals (3.67 +/- 0.69 vs. 8.63 +/- 2.22 mg B12/d). The efficiency of cobalt utilisation for ruminal vitamin B12 synthesis was calculated to be 7.1 +/- 1.3% for the unsupplemented and 9.5 +/- 2.4% for the supplemented ration. Further investigation has to prove if there are any benefits for cows resulting from the elevated cobalamin synthesis measured, caused by feeding higher amounts of dietary cobalt. 相似文献

Different assemblages of intertidal biota may be associated with the hardness of the rock type that comprises the seashore. However, very few published studies have investigated mobile and sessile assemblage differences between hardness classes. To remedy this, we investigated the physical attributes and biotic assemblages of 12 rock platforms across two bioregions, encompassing seven rock types categorised into either the soft (≤4 using Moh’s scale of scratch hardness) or hard (>4 on Moh’s scale) classes. Rock types from the soft versus hard classes differed physically, but the biotic assemblages showed few general differences between hardness classes. Instead, most biotic differences associated with rock hardness were specific to just one of the marine bioregions sampled, not both. Hardness-related assemblage differences were only weakly-to-moderately correlated with hardness differences in mineralogy or microhabitat density. The detection of bioregion-specific hardness differences for intertidal assemblages, rather than general hardness class trends, indicate that the type of rock comprising the seashore in each bioregion may be more strongly associated with the biotic patterns identified than hardness per se. 相似文献
