The use of small molecule drugs in cancer chemotherapy has mostly been limited by dose-dependent toxicity and development of drug resistance resulting from repeated administrations. To overcome such problems, efforts have been made to develop drug delivery systems that can bear multiple therapeutic agents in one system. The purpose of this study is to deliver human tumor necrosis factor (TNF)-related apoptosis-inducing ligand (Apo2L/TRAIL) and doxorubicin (Dox, an anti-cancer drug) with micellar nanoparticles self-assembled from a biodegradable cationic copolymer P(MDS-co-CES) to achieve synergistic cytotoxic effects in cancer cells. Exogenously expressed TRAIL using recombinant methods shows great potential in cancer therapy as it induces cell death selectively in cancer cells with limited toxicity to normal tissues. Dox-loaded nanoparticles and TRAIL formed stable nanocomplexes with a size of ~ 225 nm and zeta potential of ~ 70 mV. Effects of nanocomplexes on both wild type and TRAIL-resistant SW480 colorectal carcinoma cells were investigated. The assemblies of Dox and TRAIL with P(MDS-co-CES) nanoparticles were efficiently delivered to cancer cells. Receptor-blocking studies showed that the nanocomplexes entered cells via death receptor-mediated endocytosis. Synergism in cell death induction was analysed by the isobologram method to study drug interactions. Cytotoxicity of the nanocomplexes to non-cancerous cells was significantly lower than cancerous cells. Anti-proliferative effects of nanocomplexes were retained in remaining cancer cells in long-term cultures after treatment with the nanocomplexes. In summary, this Dox and TRAIL co-delivery system can be a promising candidate for cancer treatment. 相似文献

We investigated a possible association between interleukin (IL)-10 single nucleotide polymorphisms (SNPs) and susceptibility to and severity of lumbar disc degeneration (LDD) in a Chinese cohort of 320 patients with LDD and 269 gender- and age-matched controls. The degree of disc degeneration was determined by magnetic resonance imaging using Schneiderman's classification. Genetic analysis of IL-10 promoter polymorphisms (at -1082 A/G, -819 T/C, and -592 A/C) was carried out by PCR-RFLP. A total of 134 herniated lumbar intervertebral discs were collected during surgery for IL-10 mRNA detection. For SNPs at -592, the A allele and AA genotype frequencies were significantly higher in LDD patients than in controls. Similarly, the AA genotype and A allele frequencies at -1082 were significantly higher in cases than in controls. Among the LDD subjects, carriers of AA at -592 and GG at -1082 had significantly lower mean IL-10 mRNA expression than the other two genotypes. The SNPs at each locus were not significantly associated with severity grade in the LDD patients. Logistic regression analyses showed that the AA at -1082, AA at -592, and IL-10 mRNA expression level were independent risk factors for LDD. We conclude that the IL-10 SNPs at -1082 A/G and -592 A/C as well as IL-10 mRNA in the herniated lumbar intervertebral discs are associated with susceptibility to LDD in this Chinese cohort, but not with disease severity. 相似文献

An ideal expression algorithm should be able to tell truly different expression levels with small false positive errors and be robust to assay changes. We propose two algorithms. PQN is the non-central trimmed mean of perfect match intensities with quantile normalization. DQN is the non-central trimmed mean of differences between perfect match and mismatch intensities with quantile normalization. The quantiles for normalization can be either empirical or theoretical. When array types and/or assay change in a study, the normalization to common quantiles at the probe set level is essential. We compared DQN, PQN, RMA, GCRMA, DCHIP, PLIER and MAS5 for the Affymetrix Latin square data and our data of two sets of experiments using the same bone marrow but different types of microarrays and different assay. The objective of this paper was the investigation of a suitable Sepabeads? support and method for immobilization of lipase from Candida rugosa. Three different supports were used, two with amino groups, (Sepabeads? EC-EA and Sepabeads? EC-HA), differing in spacer length (two and six carbons, respectively) and one with epoxy group (Sepabeads? EC-EP). Lipase immobilization was carried out by two conventional methods (via epoxy groups and via glutaraldehyde), and
with periodate method for modification of lipase. The results of activity assays showed that lipase retained 94.8% or 87.6%
of activity after immobilization via epoxy groups or with periodate method, respectively, while glutaraldehyde method was
inferior with only 12.7% of retention. The immobilization of lipase, previously modified by periodate oxidation, via amino
groups has proven to be more efficient than direct immobilization of lipase via epoxy groups. In such a way immobilized enzyme

Studies have been made on the permeability of the inner membrane of the liver mitochondria from hibernating and active ground squirrels for succinate, glutamate, hydroxybutyrate and inorganic phosphate. The permeability was calculated from the rate of mitochondrial swelling in 100 mM ammonium salts of the substrates and phosphate. It was shown that the rate of mitochondrial swelling in hibernating animals is 2--3 times lower than in active ones, being essentially identical in a solution of ammonium phosphate. It was concluded that the permeability of the inner mitochondrial membrane for the substrates decreases in hibernating animals, remaining unaffected for phosphate. Calcium-induced activation of membrane phospholipase A2 facilitates the transport of oxidative substrates into the mitochondria of hibernating ground squirrels, significant increase in the mitochondrial respiration being simultaneously observed. The data obtained suggest that inhibition of transport of oxidative substrates is one of the main factors which account for a low respiration rate in the mitochondria of hibernating animals. 相似文献

A highly specific proteolytic enzyme cleaving at the carboxyl group of valine has been isolated from Candida tropicalis. Its specificity has been determined by digesting beta-lactoglobulin and a number of synthetic peptides. The enzyme a glycoprotein, has a molecular mass of 40 +/- 7 kDa on the basis of sodium dodecyl sulphate polyacrylamide gel electrophoresis. The effect of excessive Cd on the growth and metal uptake by leafy vegetables Brassica chinensis L. (cv. Wuyueman) and Brassica pekinensis (Lour.) Rupr. (cv. Qingyan 87-114) were studied in hydroponic solution culture. The Cd concentration higher than 10 μM significantly
decreased the root elongation and leaf chlorophyll contents of both plant species. The shoots of B. pekinensis had significantly higher concentrations of total and water-soluble Cd than B. chinensis. The roots of both species accumulated more Cd than the shoots in all the Cd treatments. Most of the Cd in the roots was
found in the cell walls. The shoot/root ratio of Cd concentrations in B. pekinensis was always greater than that in B. chinensis. But, the concentration and proportion of Cd in the cell walls in B. chinensis were higher than that in B. pekinensis. Cadmium treatments also increased the concentrations of total non-protein thiols (NPT) in the shoots of the both species.

A method for polyethylene conjugation with recombinant asparaginase has been developed to improve therapeutically important properties of enzyme. Methoxy-p-nitrophenyl carbamate of polyethylene glycol with molecular weight 5000 was employed as the modification reagent. Optimization of the pegylation procedure resulted in high level of enzyme modification. Under 4.5 molar excess of the modification reagent more than 10 molecules of methoxy-polyethylene bound per one asparaginase molecular. The modified asparaginase retained 57% of initial activity. A simple and efficient pegylation procedure described in this work can be used for production of asparaginase with improved therapeutic properties. 相似文献

In situ sampling of rhizosphere solution chemistry is an important step in improving our understanding of soil solution nutrient
dynamics. Improved understanding will enhance our ability to model nutrient dynamics and on a broader scale, to develop effective
buffers to minimize nutrient movement to surface waters. However, only limited attention has been focused on the spatial heterogeneity
and temporal dynamics of rhizosphere solution, and still less is known about how rhizosphere solution chemistry varies among
plant species. Nutrients in rhizosphere soil solution and changes in root morphology of juvenile corn (Zea mays L. cv. Stine 2250), cottonwood (Populus deltoids L.), and switchgrass (Panicum virgatum L.) were monitored using mini-rhizotron technology. Plants were grown for 10 days in a fine-silty, mixed, superactive, mesic
Cumulic Hapludoll (Kennebec series). Micro-samples (100–200 μL) of rhizosphere and bulk soil solution were collected at 24-h
intervals at a tension of −100 kPa and analyzed for P, K, Ca, and Mg concentration using Capillary Electrophoresis techniques.
Plants were harvested at the end of the 10-day period, and tissue digests analyzed for nutrient content by Inductively Coupled
Plasma Spectroscopy. Corn plants produced roots that were 1.3 times longer than those of cottonwood, and 11.7 times longer
than those of switchgrass. Similar trends were observed in number of root tips and root surface area. At the end of 10 days,
rhizosphere solution P and K concentrations in the immediate vicinity of the roots (<1 mm) decreased by approximating 24 and
8% for corn, and 15 and 5% for cottonwood. A rhizosphere effect was not found for switchgrass. After correction for initial
plant nutrient content, corn shoot P, K, and Mg were respectively 385, 132, and 163% higher than cottonwood and 66, 37, and
10% higher than switchgrass. Cottonwood shoot Ca concentration, however, was 68 to 133% higher than that of corn or switchgrass.
There was no difference in root P concentration among the three species. Nutrient accumulation efficiency (μg nutrient mm−1 root length) of cottonwood was 26 to 242% higher for P, 25 to 325% higher for Ca, and 41 to 253% higher for Mg than those
of corn and switchgrass. However, K accumulation efficiency of corn was four to five times higher than that of the cottonwood
and switchgrass. Nutrient utilization efficiency (mg of dry weight produced per mg nutrient uptake) of P, K, and Mg was higher
in cottonwood than in corn and switchgrass. These differences are element-specific and depend on root production and morphology
as well as plant nutrient status. From a practical perspective, the results of this study indicate that potentially significant
differences in rhizosphere solution chemistry can develop quickly. Results also indicate that cottonwood would be an effective
species to slow the loss of nutrients in buffer settings.
