Summary Barley (Hordeum vulgare L.) has both NADH-specific and NAD(P)H-bispecific nitrate reductases. Genomic and cDNA clones of the NADH nitrate reductase have been sequenced. In this study, a genomic clone (pMJ4.1) of a second type of nitrate reductase was isolated from barley by homology to a partial-length NADH nitrate reductase cDNA and the sequence determined. The open reading frame encodes a polypeptide of 891 amino acids and its interrupted by two small introns. The deduced amino acid sequence has 70% identity to the barley NADH-specific nitrate reductase. The non-coding regions of the pMJ4.1 gene have low homology (ca. 40%) to the corresponding regions of the NADH nitrate reductase gene. Expression of the pMJ4.1 nitrate reductase gene is induced by nitrate in root tissues which corresponds to the induction of NAD(P)H nitrate reductase activity. The pMJ4.1 nitrate reductase gene is sufficiently different from all previously reported higher plant nitrate reductase genes to suggest that it encodes the barley NAD(P)H-bispecific nitrate reductase.Scientific Paper No. 9101-14. Summary The addition of sublethal concentrations of methanol to yeast cultures was found to give strong induction of alacZ gene under heat shock element (HSE) control. Methanol addition is therefore an easily-executed alternative to temperature upshift for the induction of HSE-directed gene expression, although in this study it gave slightly lower levels of induction compared to heat shock.  相似文献

Seasonal variations in numbers and biomass and breeding of 28 species ofScarabaeidae-Scarabaeinae andGeotrupidae were studied at 2 temperate sites and 2 mediterranean sites in southern France in 1978 and 1979, using cattle-dung baited pitfall traps. Maximum dung burial occurred when females were parous and ovipositiong (i.e. during spring at the mediterranean sites and summer at the temperate sites). Using biomass as a measure of dung burial the most important spring active beetles at the mediterranean sites were:Budas bison, B. bubalus, Scarabaeus laticollis, Onthophagus lemur andCopris hispanus, and at the temperate sites:C. lunaris, O. vacca, andO. ovatus. These results confirmed the previous choice ofC. hispanus, C. lunaris, B. bison andO. vacca for introduction into Australia. In addition it is suggested thatB. bubalus, S. laticollis andO. lemur be considered for introduction into mediterranean areas of south western Australia to complement those species already introduced for bush fly control.   相似文献

Summary A total of 345 haemolysates previously phenotyped by starch gel electrophoresis and known to contain the products of the PGM 1 3 , PGM 1 6 , and PGM 1 7 alleles have been analyzed by thin layer polyacrylamide gel isoelectric focussing in the pH range 5–7. Two common subtypes, 3+and 3-, of the PGM 1 3 allele have been found in a number of Pacific populations. A single form of the PGM 1 7 allele was observed in the Western Caroline Islands. In contrast, one of two Indian PGM17 variants focussed to a different position when compared with the form found at polymorphic frequency in the Western Caroline Islands. Only one type of the PGM 1 6 allele was detected during the present investigation.  相似文献

A double-blind controlled trial of azathioprine in a dose of 2-2.5 mg/kg body weight over six months was conducted among 44 patients with active chronic ulcerative colitis. Three patients treated with placebo did not complete the trial because their disease became so severe that colectomy was performed. Among patients who completed the trial the mean dose of prednisolone necessary to control the disease decreased in those treated with azathioprine and those treated with placebo; the reduction was greater among those who took azathioprine (p less than 0.001). Activity of the disease apparently improved in both treatment groups but a significant (p less than 0.001) trend was observed only in those patients treated with azathioprine. No serious side effects from azathioprine occurred during the trial but seven of 24 patients had to stop the drug because of nausea. Azathioprine may have a role in the treatment of a few patients wih troublesome chronic colitis for whom conventional drug treatment is ineffectual, or for whom continuous systemic corticosteroid treatment is needed to control symptoms, and for whom surgical treatment is inappropriate.  相似文献

The supraoptic-hypophyseal tract is a primary system for the synthesis and release of vasopressin. Angiotensin II (AII) has been shown to release vasopressin when injected into the cerebral ventricles (IVT). However, intravenous (IV) AII injections have not produced consistent results. The present studies were conducted to examine the effects of AII delivered by either route on the unit activity of supraoptic nucleus (SON) magnocellular neurons. Rats were prepared with intracranial cannulas to insure delivery of drugs to the left lateral ventricle and with polyethylene catheters in the left jugular vein, femoral vein, and femoral artery for systemic injections and arterial pressure recordings. A ventral approach permitted recording from the SON without violating the ventricular-SON partition. Magnocellular neurons were electrophysiologically identified. In the majority of identified cells, IVT AII increased activity. In others pressor doses of AII IV inhibited firing while blood pressure was elevated. After sino-aortic denervation, AII IV excited SON neurons. Based on latency, and the fact that lesioning the anteroventral third ventricle blocked the action of AII IVT, the results indicate that AII IVT acts on a periventricular site to influence SON magnocellular neurons. Furthermore, systemic AII may have two effects on SON neurons: a central excitatory action, and an inhibition due to a baroreceptor reflex.  相似文献

Using immunoelectron microscopy and isoform-specific antibodies against Na,K-ATPase to study changes in Na,K-ATPase in rat erythroblastic cells during maturation, we unexpectedly observed numerous antigenic sites against the 3-isoform in the cytoplasmic phase. There was an increase in the number of 3-isoforms after denucleation of the erythroblast. The increase was transient. As the reticulocyte matured into a red blood cell, the number of 3-isoforms was reduced drastically. This 3-isoform was distributed in a reticular pattern resembling the double layers of endoplasmic reticulum. Western blot analysis confirms the presence of the 3-isoform in these cells. X-ray microanalysis of the erythroid series of cells in the bone marrow shows that sodium concentration in the young reticulocyte is higher than that in the nucleated erythroblast. The reason for the transient increase in this pump p rotein is not clear. It is possible that the increase in sodium concentration in the reticulocyte plays a role in the increase in pump protein synthesis.  相似文献
