A colorimetric assay for the quantitation of microbial cytotoxicity has been developed using cells from a monocyte-like human cell line (U937), epithelial cells (Hela), and fibroblast-like cells (Vero) as targets. The fraction of surviving cells was determined by their content of the dye neutral red which is retained only by live cells and can be quantitated photometrically after controlled lysis. The neutral red retention assay was at least as sensitive as the 51Cr-release assay; it was considerably less laborious, faster, and avoided handling of radioactivity. Among the different Candida species tested, the highest cytotoxicity was associated with C. albicans and C. tropicalis; a lower degree of cytotoxicity was exhibited by C. glabrata, C. guilliermondii, C. krusei, C. parapsilosis, and C. pseudotropicalis. Among the strains of a given fungal species cytotoxicity varied by up to 40%. 相似文献

Vasopressin, angiotensin II and the catecholamines decreased ketogenesis from oleate but increased ketogenesis from butyrate in hepatocytes from fed rats. The hormones increase CO2 production from both oleate and butyrate. It is suggested that whereas the mitochondrial uptake of butyrate is linked to its rate of oxidation, that of oleate is independent of its intramitochondrial metabolism, and consequently the oxidation of oleate to CO2 occurs at the expense of ketogenesis. Effects of the hormones on ketogenesis from oleate or butyrate were not observed after pre-treatment of the hepatocytes with dibutyryl cyclic AMP for 1 hour. The insensitivity of ketogenesis to the hormones after this treatment (which mimics the effects of acute carbohydrate deprivation in vivo) questions the physiological significance of hormonal regulation of ketogenesis other than at the onset of starvation. 相似文献

Population genetics of the vitamin D binding protein (GC) subtypes in the Asian-Pacific area: Description of new alleles at the GC locus 总被引:2,自引:1,他引:1
Isoelectric focussing (IEF) in thin layer polyacrylamide gels pH range 4-6.5 has been used to analyse the GC phenotypes of 4233 individuals from 28 different population groups in the Asian, Pacific, and Australian area. Because this technique reveals subtypes of the common GC*1 allele, there is almost a two-fold increase in the mean heterozygosity at the GC locus using IEF compared with conventional electrophoresis. The highest frequency (above 50%) of the GC*1S allele was encountered in Indian populations, reflecting genetic affinities with Europeans. By comparison, east and south east Asians are unique offing maximum values of the GC*1F allele (50%). With the exception of a few Pacific populations which show similar frequencies to east Asians, all other groups in the Pacific area, including Australia, have values of GC*1F similar to GC*1S ranging from 27% to 40%. The GC*2 frequency in most populations varies from 20% to 30%. However, some Polynesian groups have values up to 40% and Australian Aborigines less than 10%. Among other alleles, GC*1A1 is found to be widely distributed among Australian Aborigines and Melanesians and occurs sporadically in Polynesians, Micronesians, and in the Lesser Sunda Islands. Four new alleles, GC*1C24, GC*1C35 Aborigine, GC*1A21, and GC*1A22 are described. The Ki for the interaction of 2-fluorourocanic acid with urocanase (from ) is 1000 times as great as Km for the natural substrate, urocanic acid, whereas enzymatic hydration of the fluoro analog occurs . 100 times more slowly. Inhibition is competive and is eventually overcome by utilization of the analog. Comparative subcellular fractionation studies on rat kidney and bovine choroid plexus using differential centrifugation and free flow electropheresis were undertaken because of the morphological and functional similarities of the epithelial cells of both tissues. The activities of three enzymes commonly used as markers for brush border membranes in kidney were measured in fractions of each tissue. γ-Glutamyl transpeptidase, alkaline phosphatase, and 5'-nucleotidase copurified in membrane fractions of renal cortex collected by differential centrifugation. Application of a similar fractionation procedure to choroid plexus gave relatively similar results, except for alkaline phosphatase, the yield of which was substantially reduced in a fraction enriched with two marker enzymes. Further fractionation of γ-glutamyl transpeptidase and alkaline phosphatase activities in these membrane fractions was achieved using free flow electropheresis. The two enzymes from kidney exhibited discrete peaks with a small separation, while the electropheretic pattern of γ-glutamyl transpeptidase from choroid plexus was biphasic. Photoreactions of cytochrome b-559 and cyclic electron flow in Photosystem II of intact chloroplasts
The high potential cytochrome b-559 of intact spinach chloroplasts was photooxidized by red light with a high quantum efficiency and by far-red light with a very low quantum efficiency, when electron flow from water to Photosystem II was inhibited by a carbonyl cyanide phenylhydrazone (FCCP or CCCP). Dithiothreitol, which reacts with FCCP or CCCP, reversed the photooxidation of cytochrome b-559 and restored the capability of the chloroplasts to photoreduce CO2 showing that the FCCP/CCCP effects were reversible. The quantum efficiency of cytochrome b-559 photooxidation by red or far-red light in the presence of FCCP was increased by 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone which blocks oxidation of reduced plastoquinone by Photosystem I. When the inhibition of water oxidation by FCCP or CCCP was decreased by increased light intensities, previously photooxidized cytochrome b-559 was reduced. Red light was much more effective in photoreducing oxidized high potential cytochrome b-559 than far-red light. The red/far-red antagonism in the redox state of cytochrome b-559 is a consequence of the different sensitivity of the cytochrome to red and far-red light and does not indicate that the cytochrome is in the main path of electrons from water to NADP. Rather, cytochrome b-559 acts as a carrier of electrons in a cyclic path around Photosystem II. The redox state of the cytochrome was shifted to the oxidized side when electron transport from water became rate-limiting, while oxidation of water and reduction of plastoquinone resulted in its shifting to the reduced side. 相似文献

Summary Detailed growth analyses of cultured skin fibroblasts from two patients with Huntington's Disease (HD) were compared with those from controls matched for age and sex. In contrast to control cells, HD fibroblasts plated more efficiently at the low seeding densities used. Subsequent exponential growth of HD cultures was more stable towards routine trypsinisation than that of controls. However, the most striking feature of HD cultures was their ability to grow to significantly higher cell saturation densities. Experiments with trypsinised and untrypsinised cultures imply an inherent alteration in the HD cell membrane. 相似文献
