The aim of the present study was to test the assumption that asymmetric trunk loading requires a higher total muscle force and consequently entails a higher compression forces on the spine as compared to symmetric loading. When the trunk musculature is modelled in sufficient detail, optimisation shows that there is no mechanical necessity for an increase in total muscle force (or compression force) with task asymmetry. A physiologically based optimisation does also not predict an increase in total muscle force or spinal loading with asymmetry. EMG data on 14 trunk muscles collected in eight subjects showed antagonistic coactivity to be present in both conditions. However, estimates of total muscle force based on the EMG were lower when producing an asymmetric moment. In conclusion, producing an asymmetric moment appears to cause slightly lower forces on the lumbosacral joint as compared to a symmetric moment. Only lateral shear forces increase with asymmetry but these remain well below failure levels.  相似文献

A more fundamental understanding about the act of manual lifting can be provided by the assessment of the total production of power and the power generated in joints. The present study is concerned with the validity of the estimations of these parameters. Four subjects lifted an 18.8 kg load while they were filmed and ground reaction forces were measured. The total generated power was calculated in three ways: (1) by summation of joint powers, (2) on the basis of the rate of change of the summed energy contents of human body segments, and (3) on the basis of the rate of change of the body energy estimated from ground reaction forces. The results were compared. Furthermore, at a segmental level the power supplied to or absorbed from a segment was compared to the rate of change of its energy content. The resulting instantaneous power curves from the three different methods showed a high level of agreement, which supports their validity. However, some minor discrepancies were observed. The major cause of the observed difference between the rate of change of the summed segmental energy contents and the summed joint powers was found at a segmental level. It was observed that segmental link lengths (i.e. distances between proximal and distal markers) changed during movement, which yielded discrepancies between the power flow to or from a segment and the rate of change of its energy content.  相似文献

In this study we investigated polyamine metabolism during inhibition of two polyamine-catabolizing enzymes. This was performed by treating rats with aminoguanidine [an inhibitor of Cu-dependent amine oxidase (CuAO)], NN'-bis(buta-2,3-dienyl)butane-1,4-diamine [MDL 72527, an inhibitor of FAD-dependent polyamine oxidase (PAO)], tetrachloromethane (hepatotoxic agent) and combinations of these compounds. Emphasis was laid on the origin and possible clinical usefulness of two polyamine metabolites: acetylisoputreanine-gamma-lactam and N1N12-diacetylspermine. Acetylisoputreanine-gamma-lactam is a normal constituent of human and rat urine. Treatment of rats with aminoguanidine led to undetectable urinary levels of acetylisoputreanine-gamma-lactam, whereas MDL 72527 treatment resulted in a 12-fold increase. Under normal conditions this compound represents a minor CuAO catabolite of N1-acetylspermidine, but may become of more importance under CuAO-induced conditions. N1N12-diacetylspermine was undetectable in urine samples from non-pregnant adults and rats, but became detectable after treating rats with MDL 72527. Additional tetrachloromethane poisoning resulted in a 35-fold increase of N1N12-diacetylspermine in urine and its appearance in liver. Hence urinary excretion of N1N12-diacetylspermine during PAO inhibition may serve as a sensitive marker for cell death. This was confirmed by myeloid-leukaemia-bearing rats treated with MDL 72527, which also excreted N1N12-diacetylspermine in urine in relatively high amounts from at least day 14 until spontaneous death.  相似文献

Pseudomonas sp. GJ1 is able to grow with 2-chloroethanol as the sole carbon and energy source, but not with 2-bromoethanol, which is toxic at low concentrations (1 mM). A muatnt that could grow on 2-bromoethanol with a growth rate of 0.034 h–1 at concentrations up to 5 mM was isolated and designated strain GJ1M9. Measurement of enzyme activities showed that mutant and wild-type strains contained a PMS-linked alcohol dehydrogenase that was active with halogenated alcohols and that was threefold overexpressed in the mutant when grown on 2-chloroethanol, but only slightly overproduced when grown on 2-bromoethanol. Both strains also contained an NAD-dependent alcohol dehydrogenase that had no activity with halogenated alcohols. Haloacetate dehalogenase levels were similar in the wild-type and the mutant. Activities of NAD-dependent aldehyde dehydrogenase were only slightly higher in extracts of the mutant grown with 2-bromoethanol than in those of the wild-type grown with 2-chloroethanol. SDS-PAGE, however, showed that this enzyme amounted to more than 50% of the total cellular protein in extracts of the mutant from 2-bromoethanol-grown cells, which was fourfold higher than in extracts of the wild-type strain grown on 2-chloroethanol. The enzyme was purified and shown to be a tetrameric protein consisting of subunits of 55 kDa. The enzyme had low Km values for acetaldehyde and other non-halogenated aldehydes (0.8–4 μM), but much higher Km values for chloroacetaldehyde (1.7 mM) and bromoacetaldehyde (10.5 mM), while Vmax values were similar for halogenated and non-halogenated aldehydes. Cultures that were pregrown on 2-chloroethanol rapidly lost aldehyde dehydrogenase activity after addition of 2-bromoethanol and chloroamphenicol, which indicates that bromoacetaldehyde inactivates the enzyme. To achieve growth with 2-bromoethanol, the high expression of the enzyme thus appears to be necessary in order to compensate for the high Km for bromoacetaldehyde and for inactivation of the enzyme by bromoacetaldehyde. Zusammenfassung Hyphomicrobium vulgare ist aus den Tropfen langsam tropfender, wenig gebrauchter Wasserhähne zu isolieren auf Natriumformiat-Agar mit Nitrat als N-Quelle; gegebenenfalls kann es in einer Nährlösung für Nitritbildner angereichert werden.Der Organismus kann organische Verunreinigungen der Luft als Kohlenstoffquelle ausnutzen. Er wächst gut mit Formiaten und Acetaten, während Zucker und Asparagin nicht verwertet werden, aber die Verwertung der übrigen C-Quellen auch nicht hemmen. Hyphomicrobium besitzt eine polare Geißel. Am entgegengesetzten Ende sprossen Fäden, die nur in Dunkelfeldbeleuchtung sichtbar sind, und an denen wieder Zellen entstehen, die sich loslösen. Teilung der Zellen fehlt; dagegen findet auf trockenerem Substrat anscheinend eine Art Sprossung statt. Eine systematische Einreihung des Organismus ist zur Zeit nicht möglich.  相似文献
