A new kinetic model is presented for analysis of experimental data of oxidation process catalyzed by milk xanthine oxidase. Hyperosmotic stress initiates adaptive responses, including phosphorylation of myosin light chain (MLC) and concomitant activation of Na+-K+-Cl- cotransporter (NKCC). Because the small GTPase Rho is a key regulator of MLC phosphorylation, we investigated 1) whether Rho is activated by hyperosmotic stress, and if so, what the triggering factors are, and 2) whether the Rho/Rho kinase (ROK) pathway is involved in MLC phosphorylation and NKCC activation. Rho activity was measured in tubular epithelial cells by affinity pulldown assay. Hyperosmolarity induced rapid (<1 min) and sustained (>20 min) Rho activation that was proportional to the osmotic concentration and reversed within minutes upon restoration of isotonicity. Both decreased cell volume at constant ionic strength and elevated total ionic strength at constant cell volume were capable of activating Rho. Changes in [Na+] and [K+] at normal total salinity failed to activate Rho, and Cl- depletion did not affect the hyperosmotic response. Thus alterations in cellular volume and ionic strength but not individual ion concentrations seem to be the critical triggering factors. Hyperosmolarity induced mono- and diphosphorylation of MLC, which was abrogated by the Rho-family blocker Clostridium toxin B. ROK inhibitor Y-27632 suppressed MLC phosphorylation under isotonic conditions and prevented its rise over isotonic levels in hypertonically stimulated cells. ML-7 had a smaller inhibitory effect. In contrast, it abolished the hypertonic activation of NKCC, whereas Y-27632 failed to inhibit this response. Thus hyperosmolarity activates Rho, and Rho/ROK pathway contributes to basal and hyperosmotic MLC phosphorylation. However, the hypertonic activation of NKCC is ROK independent, implying that the ROK-dependent component of MLC phosphorylation can be uncoupled from NKCC activation. 相似文献

The ability of Phaseolus vulgaris, Mentha aquatica, and Pteris cretica to release arsenic (As) species from contaminated soil was tested in rhizobox experiments in three soils differing in their
physicochemical parameters and total and mobile As concentration. Relatively low uptake of arsenic by P. vulgaris and M. aquatica resulted in very low and ambiguous changes in rhizosphere soil compared to bulk soil. However, there were observed differences
in the distribution of the mobile As portion in soil to individual As species as affected by plant species and/or plantation
conditions of these plants. Higher percentage of mobile arsenite in mint rhizosphere seems to be related to more reducing
conditions during cultivation of these wetland plants. P. cretica planted in the soils containing between 36 and 1436 mg As kg−1 was able to accumulate between 80 and 500 mg As kg−1 in aboveground biomass. The extractable concentrations of As compounds in rhizosphere soil of P. cretica showed a clear depletion of arsenate (representing more than 90% of extractable arsenic) with the distance from plant roots.
However, the As uptake mechanisms, as well as As transformation within hyperaccumulating fern plants, differ substantially
from those in higher plants. Therefore the finding of suitable higher plant tolerant to the As soil contamination with good
ability to accumulate As in aboveground biomass remains for the further research. 相似文献

Ten minutes of exposure to increasing concentrations of sucrose caused a proportional decrease in the volume of sheep late morulae, their relative volume changed as a linear function of the reciprocal of the osmolality of the medium. Day 6 sheep and Day 7 cattle embryos responded to the addition of permeating cryoprotectants by an initial shrinkage which was followed by gradual reexpansion. After 1.25 min exposure the relative volumes of sheep and cattle embryos respectively were 20 and 25% smaller in glycerol than in ethylene glycol. The volumes of cattle and sheep embryos remained smaller in glycerol than in ethylene glycol up to the final observation at 30 min. The osmotic response of sheep late morulae to 2.0 M propylene glycol was intermediate between their response to 2.0 M glycerol and to 2.0 M ethylene glycol. These results indicate that Day 6 sheep and Day 7 cattle embryos are more permeable to ethylene glycol than to glycerol. 相似文献

Primary African green monkey kidney, continuous African green monkey kidney cell line BS-C-1, and buffalo green monkey kidney cultures were infected with a uniform inoculum of hepatitis A virus (HAV). Although both the cell line BS-C-1 and primary African green monkey kidney cultures produced useful amounts of virus, HAV was detected earlier and in greater quantities in primary African green monkey kidney cultures. A persistently infected primary African green monkey kidney culture was developed. The influence of incubation time (4 to 40 days) and concentration (2 to 15%) of fetal calf serum in the maintenance medium on production of HAV by this culture was examined. An incubation period of 24 to 28 days was found to be optimal; reducing this period led to decreased yields of HAV. No significant difference in the amount of HAV produced was observed with differing concentrations of fetal calf serum. Three different methods of extraction and the effect of multiple extractions on the recovery of HAV from cell lysates were examined. Sonication was a critical factor. Two extractions yielded more than 90% recoverable virus. Yields in excess of 10(11) physical particles of HAV per 850-cm2 roller bottle were routine. The total yield could be increased by concentrating the HAV present in spent maintenance medium by using bentonite or organic flocculation. 相似文献

Genetic parentage studies of socially monogamous birds reveal a widespread prevalence of extra-pair paternity. Variation in
extra-pair paternity among individuals may depend on how different individuals benefit from extra-pair fertilisations and
on the opportunity to pursue extra-pair copulations. A long-term study of sand martins (Riparia riparia) in Hungary allowed us to examine patterns of extra-pair fertilisations in a large colony of over 3,000 breeding pairs with
many known age individuals. We used multi-locus DNA fingerprinting to determine whether extra-pair fertilisations occur when
females are paired to (1) presumably low quality mates, or (2) genetically similar or dissimilar mates, and whether extra-pair
fertilisations result in offspring of higher quality. Extra-paternal young were found in 38% of 47 broods and comprised 19%
of 190 offspring. Males that lost paternity did not differ significantly from others in age or body condition. Social mates
of broods containing extra-pair offspring did not differ in genetic similarity from pairs without extra-pair offspring. Furthermore,
there was no significant difference in body condition between extra-pair young and their maternal half-siblings. We were unable
to assign paternity and therefore cannot exclude the possibility that extra-pair males differed from the within-pair males
they cuckolded, in age, body condition or genetic similarity with the female. We found a positive relationship between paternity
losses and breeding density, suggesting that low breeding density may constrain opportunities for seeking extra-pair copulations. 相似文献

Sperm mobility is known to be an important determinant of a male's sperm competitive ability. Although more debated, sperm length and its relation to sperm swimming ability has also been proposed to determine a male's fertilisation potential. Furthermore, both mobility and length may covary with a male's phenotype, either positively (the phenotype‐linked fertility hypothesis) or negatively if, for instance, low‐quality males have less access to females but invest more in sperm production. Using dummy females, we collected sperm samples from wild sand martins Riparia raparia males. We investigated the relationship between sperm length and sperm swimming speed as measured by sperm straight line velocity (VSL), and determined whether sperm traits are correlated with male body size and condition. We found that total sperm length is repeatable within‐ejaculate and shows substantial inter‐male variation. Sperm length was associated with sperm velocity: males with short sperm have sperm that swim initially faster but die sooner, whereas males with longer sperm have sperm that swim more slowly but for a longer time. Smaller males produced sperm with higher overall velocity. This correlation between male size and sperm behaviour may reflect alternative fertilisation strategies where small males having less mating opportunities invest more in sperm competitive ability. The existence of such alternative strategies would participate in maintaining variation in sperm length and velocity in this species. 相似文献

Vernalization, the requirement of a period of low temperature to induce transition from the vegetative to reproductive state,
is an evolutionarily and economically important trait in the Triticeae. The genetic basis of vernalization in cultivated barley (Hordeum vulgare subsp. vulgare) can be defined using the two-locus VRN-H1/VRN-H2 model. We analyzed the allelic characteristics of HvBM5A, the candidate gene for VRN-H1, from ten cultivated barley accessions and one wild progenitor accession (subsp. spontaneum), representing the three barley growth habits – winter, facultative, and spring. We present multiple lines of evidence, including
sequence, linkage map location, and expression, that support HvBM5A being VRN-H1. While the predicted polypeptides from different growth habits are identical, spring accessions contain a deletion in the
first intron of HvBM5A that may be important for regulation. While spring HvBM5A alleles are typified by the intron-localized deletion, in some cases, the promoter may also determine the allele type. The
presence/absence of the tightly linked ZCCT-H gene family members on chromosome 4H perfectly correlates with growth habit and we conclude that one of the three ZCCT-H genes is VRN-H2. The VRN-H2 locus is present in winter genotypes and deleted from the facultative and spring genotypes analyzed in this study, suggesting
the facultative growth habit (cold tolerant, vernalization unresponsive) is a result of deletion of the VRN-H2 locus and presence of a winter HvBM5A allele. All reported barley vernalization QTLs can be explained by the two-locus VRN-H1/VRN-H2 model based on the presence/absence of VRN-H2 and a winter vs. spring HvBM5A allele.

Winterhardiness has three primary components: photoperiod (day length) sensitivity, vernalization response, and low temperature
tolerance. Photoperiod and vernalization regulate the vegetative to reproductive phase transition, and photoperiod regulates
expression of key vernalization genes. Using two barley mapping populations, we mapped six individual photoperiod response
QTL and determined their positional relationship to the phytochrome and cryptochrome photoreceptor gene families and the vernalization
regulatory genes HvBM5A, ZCCT-H, and HvVRT-2. Of the six photoreceptors mapped in the current study (HvPhyA and HvPhyB to 4HS, HvPhyC to 5HL, HvCry1a and HvCry2 to 6HS, and HvCry1b to 2HL), only HvPhyC coincided with a photoperiod response QTL. We recently mapped the candidate genes for the 5HL VRN-H1 (HvBM5A) and 4HL VRN-H2 (ZCCT-H) loci, and in this study, we mapped HvVRT-2, the barley TaVRT-2 ortholog (a wheat flowering repressor regulated by vernalization and photoperiod) to 7HS. Each of these three vernalization
genes is located in chromosome regions determining small photoperiod response QTL effects. HvBM5A and HvPhyC are closely linked on 5HL and therefore are currently both positional candidates for the same photoperiod effect. The coincidence
of photoperiod-responsive vernalization genes with photoperiod QTL suggests vernalization genes should also be considered
