The composition of intracellular granules from the metal-accumulating cells of the common garden snail (Helix aspersa). 下载免费PDF全文
Certain cells in the hepatopancreas of the common garden snail (Helix aspersa) contain intracellular granules that are sites of metal-ion accumulation. These granules have been extracted and investigated by u.v. and i.r. spectroscopy, atomic-absorption spectroscopy, X-ray microanalysis, thermogravimetric analysis, enzymic assay and microanalysis. The deposits contain about 18% (w/w) water, 5% (w/w) organic matter and 76% (w/w) inorganic material of which the main components are Ca2+, Mg2+ and P2O7(4)-. The possible origin of these granules is discussed, as is their role in detoxifying heavy-metal ions. 相似文献

Methanol and formate oxidation supported the assimilation of [14C]acetate by cell suspensions of Methylococcus capsulatus; oxidation of other primary alcohols, except ethanol, did not. The extent of [1-14C]acetate assimilation supported by methanol oxidation was decreased in the presence of primary alcohols, except ethanol. Potassium cyanide (0.33 mM) completely inhibited the oxidation of formate and its stimulation of [1-14C]acetate assimilation. Various concentrations of oxygen were used to determine the optimum culture medium PO2 for survival and proliferation of attached human and mouse fibroblasts grown from different inoculum sizes. When T-15 flasks were seeded with less than or equal to 2 X 10(4) cells (less than or equal to 1.3 X 10(3) cells/cm2), the highest plating efficiencies and cell yields were obtained with a culture medium PO2 of 40-60 mm Hg. At higher inoculum sizes (10(5) cells per T-15) used routinely for mass cultured, no difference in cell yield or glycolytic activity was observed between cultures gassed with atmospheric, i.e., 18% O2 (growth medium PO2 approximately equal to 125-135 mm Hg) and those gassed with 1% O2 (growth medium PO2 approximately euqal to 40-60 mm Hg). The enhanced clonal growth observed at the latter PO2 results from an increased proliferation rate rather than more efficient attachment and survival of inoculated cells. Glucose uptake and lactic acid accumulation were increased in sparse cultures sparged with 1% O2. Summary Mark-release-recapture experiments performed with natural populations of Drosophila at Mather, California show that flies tend to return to their area of original capture or an area ecologically similar to it. Such habitat choice explains the microgeographic genetic differentiation we observed in the population. This behavioral difference between the flies may have a genetic component or may be environmentally induced. Either way, the results help explain how high levels of genetic variation are maintained by natural selection in these species. 相似文献

Fertile and sterile frond segments of the lyginopterid seed fern Feraxotheca culcitaus are described. In both the fertile and sterile specimen three orders of frond axes are borne alternately and in the same plane. Antepenultimate and penultimate frond axes are characterized by a C-shaped vascular bundle and numerous canals containing a golden-colored material in the fossilized condition. Synangia with only four sporangia demonstrate radial symmetry; bilateral symmetry is present in larger synangia. Pinnules possess dichotomous venation, with a single vein entering each lobe of the deeply incised lamina. A single canal occurs below each vein, and vein ends have an amplified vascular bundle. Morphologically Feraxotheca compares most closely with the compression genus Crossotheca. The morphology of the vascular bundle in the largest pinna is similar to a Lower Carboniferous specimen of Rhodea. 相似文献

The milk-clotting enzyme pepsin was immobilized onto beads of alumina, titania, glass, stainless steel, iron oxide, and Teflon for treating skim milk in a fluidized-bed reactor. Two covalent attachment procedures using silanized supports and glutaraldehyde and two adsorption procedures were evaluated. The three best catalysts were titania and glass, using the covalent attachment procedure, and alumina, using the adsorption procedure at pH 1.2. The pepsin adsorbed on alumina catalyst has commercial potential compared to the previously used glass catalyst. Attempts to increase the stability of pepsin adsorbed on alumina by cross-linking with glutaraldehyde were unsuccessful owing to the low pH necessary for optimum pepsin adsorption; Desorption of pepsin from alumina during reactor operation was determined. Summary Our current knowledge of the control of ventilation in fish is incomplete at all levels. The respiratory rhythm originates in a medullary central pattern generator (CPG), which has yet to be clearly identified and characterized. Its activity is directly modulated by inputs from elsewhere in the CNS and from peripheral mechanoreceptors. The central location of respiratory motoneurones, innervating the various respiratory muscles, has been described in detail for some fish, particularly elasmobranchs. We are still unclear, however, about the link between the CPG and the sequential firing of the motoneurones, which result in coordinated contractions of the respiratory muscles, and about the mechanisms that result in recruitment of feeding muscles into forced ventilation. In teleosts, ventilation is matched to oxygen requirements by stimulation of gill chemoreceptors, which seem to respond to oxygen content or supply. There is little evidence of a role for these receptors in elasmobranchs.Chemoreceptor stimulation evokes a number of reflex changes in the respiratory and cardiovascular systems of fish that are rapid in onset and seem adaptive (e.g. increased ventilation and a bradycardia in response to hypoxia). Conditions that result in hypoxaemia and the consequent ventilatory changes also cause an elevation in circulating catecholamine levels. We have explored the possibility of a causal relationship between these levels and the ventilatory response. Strong evidence for this relationship arises from experiments on hypoxia and acid infusion, which trigger a ventilatory increase and a rise in circulating catecholamines. Both ventilatory responses are blocked by an injection of propranolol, indicating that adrenoreceptors are involved in the response.The ventilatory response to hypoxia, in teleosts at least, occurs very rapidly, perhaps before any marked increase in circulating catecholamines and almost certainly before any blood-borne catecholamines could reach the respiratory neurones. This argues for an immediate neuronal reflex based on chemoreceptors in the gill region responding to hypoxia. Clearly, circulating catecholamines also affect ventilation through some action in the medulla and could act in concert with a direct neuronal chemoreceptive drive during hypoxia. The studies on acid infusion during hyperoxia, where there is an acidosis but no increase in ventilation or blood catecholamines, would argue against any hydrogen ion receptor, either peripheral or central, being involved in the reflex ventilatory response to acidotic conditions in fish.The release of catecholamines into the circulation, therefore, seems to be an absolute requirement for the ventilatory response to acidosis in fish. Present evidence supports a role for -adrenergic receptors on respiratory neurones, stimulated by changes in the levels of circulating catecholamines, in the control of ventilatory responses to marked changes in oxygen availability in fish, such as those occurring in the post-exercise acidotic state. 相似文献

In contrast to the Gram-negative bacteria, Gram-positive bacteria such as Streptomyces lack a mucopolysaccharide cell wall which allows them to produce and secrete a variety of proteins directly into their environment. In an effort to understand and eventually exploit the synthesis and secretion of proteins by Streptomyces, we identified and characterized two naturally occurring abundantly produced proteins in culture supernatants of Streptomyces lividans and Streptomyces longisporus. We purified these 10-kDa proteins and obtained partial amino acid sequence information which was then used to design oligonucleotide probes in order to clone their genes. Analysis of the sequence data indicated that these proteins were related to each other and to several other previously characterized Streptomyces protein protease inhibitors. We demonstrate that both proteins are protein protease inhibitors with specificity for trypsin-like enzymes. The presumptive signal peptidase cleavage sites and subsequent aminopeptidase products of each protein are characterized. Finally, we show that the cloned genes contain all of the information necessary to direct synthesis and secretion of the proteins by Streptomyces spp. or Escherichia coli. 相似文献
