The present investigation shows that autoreactive effector cells that transfer experimental allergic encephalomyelitis (EAE) can be activated from spleens and lymph nodes of Lewis rats given a single injection of 25 micrograms myelin basic protein (BP) in incomplete Freund's adjuvant (IFA), despite the fact that the cell donors do not develop EAE. Rather, these donor rats are unresponsive to EAE when given an encephalitogenic emulsion of BP in complete Freund's adjuvant (CFA). Lymphoid cells from rats given a single injection of BP-IFA were almost as effective as cells from BP-CFA-treated rats with respect to transferring EAE after in vitro activation with BP or concanavalin A (Con A). Irrespective of whether donors received BP in IFA or CFA, BP-cultured spleen and lymph node cells (SpC and LNC, respectively) transferred EAE, whereas Con A-cultured SpC but not LNC exhibited effector cell activity. Con A-cultured LNC were able to transfer EAE if the cultures were reconstituted with irradiated adherent phagocytic cells (which could be obtained from normal Lewis rat spleens) or with conditioned medium from these adherent SpC. These findings indicate that accessory cells are required for in vitro induction of this T cell-mediated autoimmune response.  相似文献

Damselfish of the genus Stegastes inhabit territories and cultivate algal gardens on branching corals of the genus Acropora, aggressively protecting their territories from other fish and preventing predation upon corals within the territory. This behaviour has important ecological impacts and could also be useful in reducing predation on outplanted corals during reef restoration efforts. However, the degree of protection from predators may depend on the ability of Stegastes spp. to recolonise outplanted or newly established coral colonies. Protection of bleaching-resilient massive corals within territories may be of particular importance due to the role of these corals in maintaining coral cover following bleaching events. This study examined whether the presence of Stegastes spp. reduces predation on the massive bleaching-resilient coral Porites lutea in the Mauritian lagoon, and whether Stegastes spp. readily colonise outplanted branching coral fragments and provide adjacent massive corals with indirect protection from predation. Predation levels on wild-occurring and outplanted P. lutea within and outside Stegastes spp. territories were measured. In addition, Acropora muricata branches were outplanted adjacent to wild P. lutea colonies outside Stegastes spp. territories, and recolonisation of these outplants by Stegastes spp. and the impacts of recolonisation on predation were monitored. Both wild and outplanted P. lutea colonies within Stegastes spp. territories sustained less predation damage compared to colonies outside territories. Stegastes spp. recolonized outplanted A. muricata colonies within six months of outplanting, and in doing so returned predation protection to adjacent P. lutea colonies. The ability of Stegastes spp. to colonise outplanted corals and provide indirect protection to adjacent massive bleaching-resilient corals may inform coral outplanting efforts in systems where Stegastes spp. are common. Encouraging Stegastes spp. recolonisation may help to reduce predation damage to corals within territories and potentially improve the success of rehabilitation efforts.

The susceptibility of a strain of Legionella pneumophila to disinfection by an organic halamine, free chlorine, and a mixture of the organic halamine and free chlorine was assessed. The organic halamine was found to have superior stability in solution and to exhibit adequate disinfectant potential over a period of 1 month of repeated reinoculations of fresh bacteria. The combined halamine exhibited great potential for use in maintaining closed-cycle cooling water systems free of L. pneumophila.  相似文献

We have obtained cDNA clones coding for the A, B1, and B2 chains of laminin by screening a cDNA library prepared from mouse EHS tumor poly(A)RNA in the lambda gt11 expression vector with polyclonal antibody against denatured laminin. These cDNA clones were used in combination with a cDNA clone coding for the alpha 1 type IV collagen chain to study the regulation of genes for these basement membrane proteins in retinoic acid-induced differentiating mouse F9 teratocarcinoma cells and in various adult murine tissues. The levels of mRNA for the laminin A, B1, and B2 chains and for the alpha 1 type IV collagen chain were increased simultaneously and reached a maximum at almost the same time during the differentiation of F9 cells, suggesting coordinate expression in these cells. The tissue levels of mRNA encoding for the basement membrane components, however, varied considerably. The highest level of the B1 chain mRNA was observed in kidney, whereas, the levels of mRNA for A and B2 chains were highest in heart. Almost the same levels of expression of the alpha 1(IV) collagen mRNA were found in kidney, lung, and heart. The results indicate that the expression of genes for the basement membrane proteins is not coordinately regulated in these tissues. It is thus possible that different subunit structures of the laminin molecule may exist in tissues.  相似文献

Neuromusculoskeletal models are a powerful tool to investigate the internal biomechanics of an individual. However, commonly used neuromusculoskeletal models are generated via linear scaling of generic templates derived from elderly adult anatomies and poorly represent a child, let alone children with a neuromuscular disorder whose musculoskeletal structures and muscle activation patterns are profoundly altered. Model personalization can capture abnormalities and appropriately describe the underlying (altered) biomechanics of an individual. In this work, we explored the effect of six different levels of neuromusculoskeletal model personalization on estimates of muscle forces and knee joint contact forces to tease out the importance of model personalization for normal and abnormal musculoskeletal structures and muscle activation patterns. For six children, with and without cerebral palsy, generic scaled models were developed and progressively personalized by (1) tuning and calibrating musculotendon units’ parameters, (2) implementing an electromyogram-assisted approach to synthesize muscle activations, and (3) replacing generic anatomies with image-based bony geometries, and physiologically and physically plausible muscle kinematics. Biomechanical simulations of gait were performed in the OpenSim and CEINMS software on ten overground walking trials per participant. A mixed-ANOVA test, with Bonferroni corrections, was conducted to compare all models’ estimates. The model with the highest level of personalization produced the most physiologically plausible estimates. Model personalization is crucial to produce physiologically plausible estimates of internal biomechanical quantities. In particular, personalization of musculoskeletal anatomy and muscle activation patterns had the largest effect overall. Increased research efforts are needed to ease the creation of personalized neuromusculoskeletal models.

The genes for the alpha-1 and alpha-2 chains of type IV collagen (COL4A1 and COL4A2) map to the same chromosomal band (13q34) and have a high degree of nucleotide homology. We have used pulsed field gel electrophoresis and cloned COL4A1 and COL4A2 DNA fragments as molecular probes to construct a 1200-kb macrorestriction map which encompasses both genes. The two genes are located within a 340-kb region with the 3' end of COL4A2 and the 5' region of COL4A1 separated by at least 100 kb but not more than 160 kb. These genes, therefore, are two members of a gene cluster on chromosome 13q34.  相似文献

This study investigated U.S. ethnic majority and minority youth's perceptions of fair and unfair treatment by authorities and evaluations of three situations of interracial exclusion (N = 685). Findings revealed that older adolescents who reported fair treatment by authorities were more likely to perceive wrongfulness in interracial exclusion scenarios than were their peers who reported unfair treatment. Additionally, in explaining the wrongfulness of interracial exclusion, older adolescents who had not experienced unfair treatment by authorities were more likely to mention racial prejudice than were their counterparts who had experienced unfair treatment. Results are discussed with reference to developmental theory on authority relations and the positive benefits of authority-adolescent relationships that reflect just and fair treatment.  相似文献

Children's evaluations of two parental roles, working outside the home, and staying home to take care of children, were assessed in second (M = 7.13 years, SD = .39) and fifth grade (M = 10.42, SD = .57) students (N = 121). Children viewed it as acceptable for both mothers and fathers to work full-time, and used personal choice and social conventional reasons as justifications. In contrast, children found it less acceptable for fathers to stay at home than for mothers to stay at home, and they used gender stereotypes about domestic roles as justifications. With age, children were more flexible in their reasoning and used fewer stereotypes in their evaluations; children from traditional family structures used more stereotypic expectations than did children from non-traditional ones. Overall, children's interpretations of competence in a caretaker role was highly contingent on the gender of the parent.  相似文献
