The Diego Ramirez Islands lie 60 nautical miles southwest of Cape Horn and are the breeding site for three species of burrowing
seabirds: blue petrels (Halobaena caerulea), common diving petrels (Pelecanoides urinatrix) and sooty shearwater (Puffinus griseus). Burrowing seabirds are highly vulnerable to predation by introduced vertebrate pests, and Diego Ramirez is an important
breeding site because it is one of a few remaining subantarctic island groups with no introduced predators. Diego Ramirez
is the only known breeding site for blue petrels in the southeast Pacific region, holding about 80% of the global population
of that species, and with a population ten times larger than any other population in the world. We estimated the population
size in 2002, using a novel application of the distance sampling technique to determine burrow density, and a burrow-scope
with excavations to determine occupying species. We found that density was correlated with slope angle and soil wetness. Burrow
densities in flatter terrain with drier soils were 2.03 burrows/m2 (95% confidence intervals: 1.82–2.27) and 1.11 burrows/m2 (0.84–1.48) in steeper terrain with wetter soils. The occupation rate of burrows were significantly different between habitat
types (t=2.74, d.f. 11, P<0.05); in flatter drier habitats the proportion of burrows that led to a nest was 0.85 (0.74–0.96), in steeper wetter habitats
this decreased to 0.64 (0.50–0.78). We used a digital elevation model to calculate true area rather than planar area for the
two habitat types on the main island of Bartolome, and charts to calculate planar area for the remainder of the archipelago.
There were 1.35 (1.15–1.54) million pairs of blue petrels and 99,000 (65,000–134,000) pairs of common diving petrels on the
archipelago. These are similar figures to those from the only previous estimate, made in 1980. We found breeding sooty shearwaters
for the first time, and estimated a population of several thousand pairs. We emphasise the facility of distance sampling as
an unbiased technique with practical advantages over commonly used area search methods for monitoring populations of burrowing
seabirds. These advantages include increased survey efficiency allowing a larger sample size for a given effort and a correspondingly
tighter estimation of density. 相似文献

As genome-scale metabolic reconstructions emerge, tools to manage their size and complexity will be increasingly important. Flux balance analysis (FBA) is a constraint-based approach widely used to study the metabolic capabilities of cellular or subcellular systems. FBA problems are highly underdetermined and many different phenotypes can satisfy any set of constraints through which the metabolic system is represented. Two of the main concerns in FBA are exploring the space of solutions for a given metabolic network and finding a specific phenotype which is representative for a given task such as maximal growth rate. Here, we introduce a recursive algorithm suitable for overcoming both of these concerns. The method proposed is able to find the alternate optimal patterns of active reactions of an FBA problem and identify the minimal subnetwork able to perform a specific task as optimally as the whole. Our method represents an alternative to and an extension of other approaches conceived for exploring the space of solutions of an FBA problem. It may also be particularly helpful in defining a scaffold of reactions upon which to build up a dynamic model, when the important pathways of the system have not yet been well-defined. 相似文献

A popular explanation for the small number of women at the top level of intellectually demanding activities from chess to science appeals to biological differences in the intellectual abilities of men and women. An alternative explanation is that the extreme values in a large sample are likely to be greater than those in a small one. Although the performance of the 100 best German male chess players is better than that of the 100 best German women, we show that 96 per cent of the observed difference would be expected given the much greater number of men who play chess. There is little left for biological or cultural explanations to account for. In science, where there are many more male than female participants, this statistical sampling explanation, rather than differences in intellectual ability, may also be the main reason why women are under-represented at the top end. 相似文献

Fibulin 5 is a 52-kDa calcium-binding epidermal growth factor (cbEGF)-rich extracellular matrix protein that is essential for the formation of elastic tissues. Missense mutations in fibulin 5 cause the elastin disorder cutis laxa and have been associated with age-related macular degeneration, a leading cause of blindness. We investigated the structure, hydrodynamics, and oligomerization of fibulin 5 using small angle x-ray scattering, EM, light scattering, circular dichroism, and sedimentation. Compact structures for the monomer were determined by small angle x-ray scattering and EM, and are supported by close agreement between the theoretical sedimentation of the structures and the experimental sedimentation of the monomer in solution. EM showed that monomers associate around a central cavity to form a dimer. Light scattering and equilibrium sedimentation demonstrated that the equilibrium between the monomer and the dimer is dependent upon NaCl and Ca2+ concentrations and that the dimer is dominant under physiological conditions. The dimerization of fragments containing just the cbEGF domains suggests that intermolecular interactions between cbEGFs cause dimerization of fibulin 5. It is possible that fibulin 5 functions as a dimer during elastinogenesis or that dimerization may provide a method for limiting interactions with binding partners such as tropoelastin.Fibulins are a family of seven extracellular matrix glycoproteins, some of which associate with elastic fibers and basement membranes (1, 2). They are involved in the assembly, organization, and stabilization of macromolecular complexes (3). Fibulins contain arrays of cbEGF2-like domains and a fibulin-type C-terminal (Fc) module (4). Fibulins 3–5 have a modified N-terminal cbEGF domain, followed by five cbEGF domains (4).Fibulin 5 (supplemental Fig. S1) is highly expressed in developing arteries with a low expression in adult vessels that is up-regulated following vascular injury and in atherosclerosis (5, 6). Expression has been detected in other elastin-rich tissues, including aorta, skin, uterus, lung, heart, ovary, and colon (5, 6). The extensibility of such tissues is provided by elastic fibers (7), and aging is associated with a loss of elasticity (8). Fibulin 5 is essential for elastinogenesis. The fibulin 5 knock-out mouse exhibits disorganized elastic fibers resulting in severe elastinopathies, with loose skin, vascular abnormalities, and emphysematous lungs. Similar changes are seen in an aged phenotype (9, 10). Mutations in fibulin 5 lead to the elastin disorder cutis laxa (11–13) and have been associated with age-related macular degeneration (14, 15).It has been shown that fibulin 5 binds elastic fibers (16) and interacts with tropoelastin (10), fibrillin 1 (17), lysyl oxidase-like protein 1 (18), -2, and -4 (19), latent transforming growth factor-β-binding protein 2 (19), emilin 1 (20), apolipoprotein (a) (21), and superoxide dismutase (22). Through an RGD motif fibulin 5 interacts with integrins (6, 9, 23).The assembly of elastic fibers is a complex hierarchical process. A model proposes that fibulin 5 associates with microfibrils via interactions with fibrillin 1; tropoelastin molecules bind fibulin 5 and coacervate, and lysyl oxidase-like protein 1 enzymes cross-link tropoelastin to form mature elastin (7, 16). Data that support this model indicate that fibulin 5 potentially increases the coacervation of tropoelastin, enhancing elastic fiber formation (24). However, other data suggest that fibulin 5 slows the maturation of elastin assemblies (25).Rotary-shadowing EM has suggested that fibulin 5 exists as a short rod with a globular domain at one end (26). We used size-exclusion column multiangle laser light scattering (SEC-MALLS), small angle x-ray scattering (SAXS), EM single particle analysis, analytical ultracentrifugation (AUC), CD, and isoelectric focusing to investigate the structures of fibulin 5 in monomeric and dimeric form, and the equilibrium between the two forms. 相似文献

A new homobimetallic complex [Cu2(tpp)2(dppm)Br2] (1) of copper(I) bromide with triphenylphosphine (tpp) and bis-diphenylphosphinomethane (dppm) has been synthesized and charaterized by m.p., elemental analysis, FT-IR, 1H NMR, mass spectrometry, thermal studies and single crystal X-ray analysis. The solid-state molecular structure of 1, belonging to the monoclinic crystal system with space group P21/n, describes it as a neutral dinuclear species in which two copper atoms are bridged together through two bromides and a dppm ligand and each copper atom possesses a distorted tetrahedral geometry. Complex 1 was studied as a single-source precursor for the fabrication of phase pure thin films of Cu3P by aerosol-assisted chemical vapour deposition. The films have been characterized by PXRD, SEM and ED-XRF analyses and found to exhibit the particles size range 200−400 nm with high purity and surface uniformity. 相似文献

Introduction – Commercially available herbal mixture FE, a proprietary natural health product manufactured by Flora Manufacturing and Distributing Ltd (Flora), is a unique North American traditional herbal product. FE is a chemically complex mixture of eight herbs and has not been subjected to phytochemical analysis. Objective – To develop analytical methods to undertake detailed phytochemical analyses of FE, and its eight contributing herbs, including burdock (Arctium lappa L.), sheep sorrel (Rumex acetosella L.), Turkish rhubarb (Rheum palmatum L.), slippery elm Muhl. (Ulmus rubra), watercress (Nasturtium officinale R. Br.), red clover (Trifolium pratense L.), blessed thistle (Cnicus benedictus L.) and kelp (Laminaria digitata Lmx.). Methodology – The identification was undertaken by a combination of reversed phase high performance liquid chromatography–diode array detection–atmospheric pressure chemical ionisation–mass selective detection (RP‐HPLC‐DAD‐APCI‐MSD) analysis and phenolics metabolomic library matching. Results – New separation methods facilitated the identification of 43 markers in the individual herbs which constitute FE. Sixteen markers could be identified in FE originating from four contributing herbs including four caffeoyl quinic acids, three dicaffeoyl quinic acids and two caffeic acid derivatives from A. lappa, luteolin‐7‐O‐glucoside, luteolin, five apigenin glycosides and apigenin from R. acetocella and N. officinale and sissostrin from T. pretense. A validated method for quantitative determination of three markers is reported with good intraday, interday and interoperator repeatability using a reliable alcohol based extraction technique. Conclusion – FE and its contributing herbs predominantly contain phenolics. This methodology can be applied to further develop full‐scale validation of this product.
