The fermentation process of l-lactic acid is well known. Little importance was attached to d-lactic acid, but in the past 10 years, d-lactic acid gained significantly in importance. d-Lactic acid is an interesting precursor for manufacturing heat-resistant polylactic acid (PLA) bioplastics which can be widely used, for example as packaging material, coatings, for textiles or in the automotive industry.This review provides a comprehensive overview of the most recent developments, including a spectrum of studied microorganisms and their capabilities for the production of d-lactic acid. Additionally, the technological achievements in biotechnological d-lactic acid production including fermentation techniques like fed batch, simultaneous saccharification, and fermentation and continuous techniques are presented. Attention is also turned to suitable alternative substrates and their applicability in fermentation processes. Furthermore, advantages and disadvantages of product recovery and purification are discussed. Economic aspects of PLA are pointed out, and the present industrial producers of lactic acid are briefly introduced.  相似文献

Territorial passerines presumably benefit from their ability to use auditory cues to judge the distance to singing conspecifics, by increasing the efficiency of their territorial defence. Here, we report data on the approach of male territorial chaffinches, Fringilla coelebs, to a loudspeaker broadcasting conspecific song simulating a rival at various distances by different amounts of song degradation. Songs were degraded digitally in a computer-simulated forest emulating distances of 0, 20, 40, 80 and 120 m. The approach distance of chaffinches towards the loudspeaker increased with increasing amounts of degradation indicating a perceptual representation of differences in distance of a sound source. We discuss the interindividual variation of male responses with respect to constraints resulting from random variation of ranging cues provided by the environmental song degradation, the perception accuracy and the decision rules. Microbial community structure was linked to degradation potential in benzene-, toluene- or xylene- (BTX) degrading, iron-reducing enrichments derived from an iron-reducing aquifer polluted with landfill leachate. Enrichments were characterized using 16S rRNA gene-based analysis, targeting of the benzylsuccinate synthase-encoding bssA gene and phospholipid fatty acid (PLFA) profiling in combination with tracking of labelled substrate. 16S rRNA gene analysis indicated the dominance of Geobacteraceae, and one phylotype in particular, in all enrichments inoculated with polluted aquifer material. Upon cultivation, progressively higher degradation rates with a concomitant decrease in species richness occurred in all primary incubations and successive enrichments. Yet, the same Geobacteraceae phylotype remained common and dominant, indicating its involvement in BTX degradation. However, the bssA gene sequences in BTX degrading enrichments differed considerably from those of Geobacter isolates, suggesting that the first steps of toluene, but also benzene and xylene oxidation, are carried out by another member of the enrichments. Therefore, BTX would be synthrophically degraded by a bacterial consortium in which Geobacteraceae utilized intermediate metabolites. PLFA analysis in combination with (13)C-toluene indicated that the enriched Geobacteraceae were assimilating carbon originally present in toluene. Combined with previous studies, this research suggests that Geobacteraceae play a key role in the natural attenuation of each BTX compound in situ.  相似文献

In order to enhance understanding of the interrelationships among community members and between them and their environment, the concept of regulation analysis is extended from biochemistry into microbial ecology. Ecological regulation analysis quantifies how biogeochemical fluxes are regulated by the microorganisms performing the process; the degree to which changes in fluxes are due to changes in population size and to changes in activity cell(-1) (cellular activity). Regulation analysis requires data on biogeochemical fluxes and the numbers of cells through which these fluxes run. Its application to five biogeochemical processes (aerobic methane oxidation, aerobic nitrite oxidation, methanogenesis, sulfate reduction and reductive dehalogenation) revealed that in general, but not always, flux was primarily regulated by cellular activity, i.e. by changes in the size and properties of the enzyme pool and in the concentrations of substrates and metabolites. Thus, it is often not sufficient to count the numbers of cells performing a particular step in a biogeochemical process in order to estimate its flux. Ecological regulation analysis can be extended to address which aspects of cellular activity require quantification in order to describe biogeochemical fluxes better. Its application is discussed in the context of the complexity of microbial communities (e.g. functional redundancy) and their functioning.  相似文献

Over large areas of Europe, coniferous monocultures are being transformed into mixed forests by the re-introduction of broadleaf tree species belonging to the potential natural vegetation. One important species of interest in this changing forest policy is European beech (Fagus sylvatica). However, at present, this forest management directive has ignored potential adverse effects of global climate change on wide-spread re-introduction of beech to these areas. Average global surface temperatures have risen by approx. 0.8°C in the period between 1861 and 2005 and are expected to continue to increase until the end of this century by 1.5–5.8°C above the 1990 value. To estimate the climate change in the southern part of central Europe in future, we reviewed calculations from regional climate models. Temperature increase for the southern part of central Europe is projected to be up to 2°C within the next 40 years. In contrast, the annual precipitation will most likely remain constant over the same time period, but will experience significant changes in seasonal patterns. Rising intensities of individual precipitation events may result in increasing number and intensities of flooding events and reduced precipitation during the growing season in a higher frequency of summer droughts. Growth and competitive ability of European beech will not, necessarily, respond to increasing CO2 concentrations but may be strongly impacted by intensive drought that occurs during the growing season. Seedlings as well as adult trees may suffer from xylem embolism, restricted nutrient uptake capacity and reduced growth under limited water availability. However, it remains uncertain to what extent other environmental factors (e.g. soil properties, competitive interactions) may modify the drought response of beech, thus either enhancing susceptibility or increasing drought tolerance and resilience potential. Water-logged soils, predicted during the spring for several regions due to higher than average precipitation, could negatively impact nutrient uptake and growth of beech. Whereas other dominant species as, e.g. oak are well adapted to that environmental stress, beech is known to be sensitive to water-logging and flooding. Thus, the competitive capacity of beech might—depending on the other environmental conditions—be reduced under the expected future climate conditions. Silvicultural practices must be aware today of the potential risks which a changing climate may impose on sustainable forest development.  相似文献

A novel selective enrichment method is described for phototrophic green sulfur bacteria even in the presence of purple sulfur and purple nonsulfur bacteria using sulfanilate, which was discovered during efforts to selectively isolate sulfanilate-metabolizing anoxygenic phototrophic bacteria from marine habitats. Samples for these experiments were obtained from beaches, saltpans, subsurface mangrove soils, fish and prawn aquaculture ponds and backwaters of the East and West coasts of India. Photoorganoheterotrophic and photolithoautotrophic enrichments in the absence of sulfanilate predominantly yielded purple bacterial enrichments. In contrast, photolithoautotrophic enrichments in the presence of sulfanilate yielded green-colored enrichments from the same samples. Whole cell absorption spectra of the enrichment cultures revealed the presence of bacteriochlorophyll c and thus green phototrophic bacteria. Microscopic observation demonstrated the presence of sulfur globules outside the bacterial cells and the presence of non-motile cells, some of which had prosthecae. 16S rDNA sequences obtained from green sulfur bacterial strains isolated from enrichment cultures confirmed the presence of representatives of the green sulfur bacterial genera Prosthecochloris and Chlorobaculum. The selective pressure of sulfanilate exerted through inhibition of phototrophic purple sulfur bacteria was demonstrated by inhibition studies using the purple sulfur bacteria Marichromatium indicum JA100 and Marichromatium sp. JA120 (JCM 13533) and the green sulfur bacterium Prosthecochloris sp. JAGS6 (JCM 13299).  相似文献

The aim of this investigation was to evaluate the changes in PAF concentrations in the plasma, PBMC and BMMC of patients with acute lymphoblastic leukemia (ALL) and acute myeloblastic leukemia (AML). The plasma was from 23 healthy volunteers (HV) and 44 patients with AL (16 ALL, 28 AML). The PBMC were from 15 HV and 55 patients with AL (18 ALL, 37 AML), and the BMMC from 40 patients with AL (11 ALL, 29 AML). Methanol-chloroform phospholipid extraction from 60 × 106 cells (PBMC or BMMC) was performed according to a modified version of Folch’s method. 31P MRS data was obtained on an AMX 300 Bruker spectrometer (7.05 T). The PAF concentration in the plasma of the patients with ALL or AML was lower than that for the healthy volunteers. The PAF concentration in the plasma of the patients with ALL did not differ significantly from that of the patients with AML. In the case of both the PBMC and BMMC, the PAF concentration was significantly diminished in patients with ALL relative to the concentration for those with AML and for the healthy volunteers. No differences were observed in the PAF concentrations for the AML patients and the healthy volunteers.  相似文献
