The cytoplasmic step of posttranslational secretion in Escherichia coli is catalyzed by export-specific chaperone SecB and translocational ATPase SecA. In addition, the efficiency of secretion depends on the charge of the signal peptide (SP). Replacement of positively charged Lys(–20) with uncharged Ala or negatively charged Glu in the N-terminal region of SP of the alkaline phosphatase precursor (prePhoA) was shown to decrease the PhoA secretion in the periplasm. The effect on secretion increased in the absence of SecB and was especially high on SecA inactivation. A change in SP charge strengthened the SecA and SecB dependences of secretion. On evidence of immunoprecipitation, the charge of the N-terminal region of SP had no effect on prePhoA interaction with the cytoplasmic secretion factors, suggesting no direct binding between this region and SecA or SecB. Yet the charge of the N-terminal region proved to affect the functions of SP as an intramolecular chaperone and a factor of prePhoA targeting to the membrane in cooperation with SecA and SecB. 相似文献

Similarly to Helicobacter pylori but unlike Vibrio cholerae O1/O139, Campylobacter jejuni is non‐motile at 20°C but highly motile at ≥37°C. The bacterium C. jejuni has one of the highest swimming speeds reported (>100 μm/s), especially at 42°C. Straight and spiral bacterial shapes share the same motility. C. jejuni has a unique structure in the flagellate polar region, which is characterized by a cup‐like structure (beneath the inner membrane), a funnel shape (opening onto the polar surface) and less dense space (cytoplasm). Other Campylobacter species (coli, fetus, and lari) have similar motility and flagellate polar structures, albeit with slight differences. This is especially true for Campylobacter fetus, which has a flagellum only at one pole and a cup‐like structure composed of two membranes. 相似文献

Recoverin is an N-myristoylated 23 kDa calcium-binding protein from retina, which modulates the Ca2+-sensitive deactivation of rhodopsin via Ca2+-dependent inhibition of rhodopsin kinase. It was shown by intrinsic and bis-ANS probe fluorescence, circular dichroism, and differential scanning calorimetry that myristoylated recombinant recoverin interacts specifically with zinc ions. Similar to the calcium binding, the binding of zinc to Ca2+-loaded recoverin additionally increases its alpha-helical content, hydrophobic surface area, and environmental mobility/polarity of its tryptophan residues. In contrast to the calcium binding, the binding of zinc decreases thermal stability of the Ca2+-loaded protein. Zn2+-titration of recoverin, traced by bis-ANS fluorescence, reveals binding of a single Zn2+ ion per protein molecule. It was shown that the double-mutant E85Q/E121Q with inactivated Ca2+-binding EF-hands 2 and 3 (Alekseev, A. M.; Shulga-Morskoy, S. V.; Zinchenko, D. V.; Shulga-Morskaya, S. A.; Suchkov, D. V.; Vaganova, S. A.; Senin, I. I.; Zargarov, A. A.; Lipkin, V. M.; Akhtar, M.; Philippov, P. P. FEBS Lett. 1998, 440, 116-118), which can be considered as an analogue of the apo-protein, binds Zn2+ ion as well. Apparent zinc equilibrium binding constants evaluated from spectrofluorimetric Zn2+-titrations of the protein are 1.4 x 10(5) M(-1) (dissociation constant 7.1 microM) for Ca2+-loaded wild-type recoverin and 3.3 x 10(4) M(-1) (dissociation constant 30 microM) for the E85Q/E121Q mutant (analogue of apo-recoverin). Study of the binding of wild-type recoverin to ROS membranes showed a zinc-dependent increase of its affinity for the membranes, without regard to calcium content, suggesting further solvation of a protein myristoyl group upon Zn2+ binding. The distribution of parasites among individual hosts is characterised by high variability that is believed to be a result of variations in host traits. To find general patterns of host traits affecting parasite abundance, we studied flea infestation of nine rodent species from three different biomes (temperate zone of central Europe, desert of Middle East and tropics of East Africa). We tested for independent and interactive effects of host sex and body mass on the number of fleas harboured by an individual host while accounting for spatial clustering of host and parasite sampling and temporal variation. We found no consistent patterns of the effect of host sex and body mass on flea abundance either among species within a biome or among biomes. We found evidence for sex-biased flea infestation in just five host species (Apodemus agrarius, Myodes glareolus, Microtus arvalis, Gerbillus andersoni, Mastomys natalensis). In six rodent species, we found an effect of body mass on flea abundance (all species mentioned above and Meriones crassus). This effect was positive in five species and negative in one species (Microtus arvalis). In M. glareolus, G. andersoni, M. natalensis, and M. arvalis, the relationship between body mass and flea abundance was mediated by host sex. This was manifested in steeper change in flea abundance with increasing body mass in male than female individuals (M. glareolus, G. andersoni, M. natalensis), whereas the opposite pattern was found in M. arvalis. Our findings suggest that sex and body mass are common determinants of parasite infestation in mammalian hosts, but neither of them follows universal rules. This implies that the effect of host individual characteristics on mechanisms responsible for flea acquisition may be manifested differently in different host species. 相似文献

Abstract. 1. The fleas Xenopsylla conformis and Xenopsylla ramesis exploit the same rodent host, Meriones crassus , and replace each other between two different habitats situated at the opposite sides of a steep precipitation gradient. It was hypothesised that the reason for this paratopic distribution is competition between larvae of the two species for food resources.
2. This hypothesis was tested by studying the performance of larvae of the two species in terms of their developmental success in mixed-species and single-species treatments under different air temperatures, relative humidities, substrate textures, and food abundance.
3. The number of individuals of X. conformis that survived until emergence depended significantly on the presence of competing species, being, in general, lower in mixed-species compared with single-species treatments. The decrease in developmental success of X. conformis in mixed-species treatments was found mainly during food shortage. In contrast, presence of the competitor did not affect the number of X. ramesis that survived until emergence. No effect of the presence of the competitor on duration of development or sex ratio was found in either species.

Aim Spatial variation in the diversity of fleas parasitic on small mammals was examined to answer three questions. (1) Is the diversity of flea assemblages repeatable among populations of the same host species? (2) Does similarity in the composition of flea assemblages among populations of the same host species decay with geographical distance, with decreasing similarity in the composition of local host faunas, or with both? (3) Does the diversity of flea assemblages correlate with climatic variables? Location The study used previously published data on 69 species of small mammals and their fleas from 24 different regions of the Holarctic. Methods The diversity of flea assemblages was measured as both species richness and the average taxonomic distinctness of their component species. Similarity between flea assemblages was measured using both the Jaccard and Morisita–Horn indices, whereas similarity in the composition of host faunas between regions (host ‘faunal’ distance) was quantified using the Jaccard index. Where appropriate, a correction was made for the potentially confounding influence of phylogeny using the independent contrasts method. Results Flea species richness varied less within than among host species, and is thus a repeatable host species character; the same was not true of the taxonomic distinctness of flea assemblages. In almost all host species found in at least five regions, similarity in flea assemblages decreased with increases in either or both geographical and faunal distance. In most host species, the diversity of flea assemblages correlated with one or more climatic variable, in particular mean winter temperature. Main conclusions Spatial variation in flea diversity among populations of the same mammal species is constrained by the fact that it appears to be a species character, but is also driven by local climatic conditions. The results highlight how ecological processes interact with co‐evolutionary history to determine local parasite biodiversity. 相似文献

A negative interspecific correlation between the degree of habitat specialization and the size of a species' geographic range has been documented for several free living groups of organisms, providing support for the niche breadth hypothesis. In contrast, practically nothing is known about the geographic range sizes of parasitic organisms and their determinants. In the context of the niche breadth hypothesis, parasites represent ideal study systems, because of the well documented variation in host specificity among parasite species. Here, we investigated the relationship between host specificity (a measure of niche breadth) and geographic range size among flea species parasitic on small mammals, using data from seven distinct geographical regions. Two measures of host specificity were used: the number of host species used by a flea species, and a measure of the average taxonomic distance between the host species used by a flea; the latter index provides an evolutionary perspective on host specificity. After correcting for phylogenetic influences, and using either of our two measures of host specificity, the degree of host specificity of fleas was negatively correlated with the size of their geographic range in all seven regions studied here, with only one minor exception. Overall, these results provide strong support for the niche breadth hypothesis, although other explanations cannot be ruled out. 相似文献

We studied the co-occurrence of flea species in infracommunities of 16 rodents from four regions (South Africa, Tanzania,
central Europe and western Siberia) using null models, and predicted that flea co-occurrences will be expressed more strongly
in male than in female hosts. We examined patterns of co-occurrence (measured as the C score) in infracommunities of fleas that are parasitic on male and female hosts by comparing co-occurrence frequencies with
those expected by chance. When a significant degree of nonrandomness in flea co-occurrences was detected, it indicated aggregative
infracommunity structure. In Tanzanian rodents, no significant flea co-occurrences were detected in either male or female
hosts. In a South African rodent, significant flea co-occurrences were not detected in males, but were found in females in
some localities. In Palaearctic rodents, significant nonrandomness was detected either equally for males and females or more
frequently in males than in females. Meta-analyses demonstrated that the frequency of the detection of nonrandomness in flea
co-occurrences was significantly higher in male than in female hosts. The values of the standardized effect size (SES) for
the C score differed significantly among host species, but not between host genders. When the Palaearctic hosts were analyzed separately,
the effects of both host gender and species appeared to be significant, with the SES values for the C score in males being smaller than those in females. The strength of the gender difference in the manifestation of flea community
structure increased with increasing gender difference in flea species richness, and with decreasing gender difference in flea
prevalence for the Palaearctic hosts. We conclude that male hosts are the main drivers of flea infracommunity structure. However,
the manifestation of gender bias in flea community structure varies among host species, and is likely determined by the pattern
