Introduction
Synovial tissue macrophages play a key role in chronic inflammatory arthritis, but the contribution of different macrophage subsets in this process remains largely unknown. The main in vitro polarized macrophage subsets are classically (M1) and alternatively (M2) activated macrophages, the latter comprising interleukin (IL)-4 and IL-10 polarized cells. Here, we aimed to evaluate the polarization status of synovial macrophages in spondyloarthritis (SpA) and rheumatoid arthritis (RA).Methods
Expression of polarization markers on synovial macrophages, peripheral blood monocytes, and in vitro polarized monocyte-derived macrophages from SpA versus RA patients was assessed by immunohistochemistry and flow cytometry, respectively. The polarization status of the intimal lining layer and the synovial sublining macrophages was assessed by double immunofluorescence staining.Results
The expression of the IL-10 polarization marker cluster of differentiation 163 (CD163) was increased in SpA compared with RA intimal lining layer, but no differences were found in other M1 and M2 markers between the diseases. Furthermore, no significant phenotypic differences in monocytes and in vitro polarized monocyte-derived macrophages were seen between SpA, RA, and healthy controls, indicating that the differential CD163 expression does not reflect a preferential M2 polarization in SpA. More detailed analysis of intimal lining layer macrophages revealed a strong co-expression of the IL-10 polarization markers CD163 and cluster of differentiation 32 (CD32) but not any of the other markers in both SpA and RA. In contrast, synovial sublining macrophages had a more heterogeneous phenotype, with a majority of cells co-expressing M1 and M2 markers.Conclusions
The intimal lining layer but not synovial sublining macrophages display an IL-10 polarized-like phenotype, with increased CD163 expression in SpA versus RA synovitis. These differences in the distribution of the polarized macrophage subset may contribute to the outcome of chronic synovitis. 相似文献34.

125I-labeled derivatives of iodaryl-polyethylene oxide with mol. wt. equal to 3 X 10(3), containing and not containing dinitrophenyl (DNP) residues (haptens), were obtained. The capacity for anti-DNP response was shown to be unaffected by the presence of the iodaryl group. The pharmacokinetics of both derivatives of polyethylene oxide were studied in experiments on noninbred white rats receiving these derivatives intravenously in doses of 0.53-0.80 mg/kg. These experiments revealed the following differences in the pharmacokinetics of the derivatives under study: the presence of the DNP group led to an increase in the level of polymer fixation in the body of the animals, particularly in their blood, liver and spleen. Differences in the content of radioactive substances in other organs were not statistically significant. 相似文献

The hypothesis on the role of protonmotive force in the transport of DNA through the membrane of Bac. subtilis cell during initial stages of genetic transformation was tested. A genetic transformation of arsenate-treated cells was observed. Treatment of cells by the protonophorous uncoupler of oxidative phosphorylation-carbonylcyanide dichlorophenyl--hydrazone-led to the inhibition of initial stages of genetic transformation having no significant effect on the level of intracellular ATP concentration and on the viability of cells. The dissipation of protonmotive force by means of K+ and H+ fluxes catalyzed by valinomycin and nigericin also caused the inhibition of initial stages of genetic transformation. The inhibitory effect of cationic penetrant tetraphenyl phosphonium was observed, the effect being potentiated by low concentrations of anionic penetrant phenyldicarbaundecaborate. The value of the membrane potential in the energized valinomycin-treated cells calculated from the distribution of K+ was within the range of 70--100 mV (inside minus). These results support the conception that a protonmotive force drives DNA transport through the membrane of Bac. subtilis cells. 相似文献

Effects of different inhibitors of lipid peroxidation (LP), such as sulphur-containing oligoquinone hypoxen, natural flavonoid
dihydroquercetin (DHQ), and β-ionol, on Ca2+-induced calcium release from rat liver mitochondria (RLM) were investigated during oxidation of various substrates. The hypothesis
about interrelation between antioxidant properties and influence of selected substances on spontaneous calcium release from
mitochondria was verified. Degree of antioxidant activity of the selected substances was estimated by the inhibition of LP
induced by Fe2+/ATP complex in phospholipid emulsion or in rat liver mitochondria (RLM). According to the inhibition efficacy the investigated
substances were ordered as follows: β-ionol ≫ hypoxen > DHQ. 50% inhibition of oxygen consumption during LP of phospholipid
emulsion was reached in presence of 3.2 ± 0.6 μM of β-ionol, 15.0 ± 1.1 μM of hypoxen, or 19.8 ± 1.7 μM of DHQ. Among the
investigated antioxidants hypoxen only decreased spontaneous release of calcium from RLM after calcium accumulation by RLM.
The impact of the antioxidants onto calcium current depended on the oxidized substrate. Hypoxen effect was most expressed
during the oxidation of NAD-dependent substrate. The direct relationship between the antioxidant activity of the selected
antioxidants and their influence on calcium transport in RLM was not revealed. The results indicate that the choice of antiischemic
preparations should not only rely on their antioxidant activities. 相似文献

Properties and allelic ADH (alcohol dehydrogenase) control in Drosophila melanogaster were studied upon flies' cultivation under conditions of hypotherm of different intensity and duration. Lines homozygotic for F allele (vg) and S allele (cn) of the Adh gene as well as genetically enriched experimental cn' and vg' populations containing a small number of AdhF/AdhS heterozygotes at the initial stage were used. It was found out that physiological adaptation of the species to momentary influence of elevated temperature is accompanied by modification of physical properties of ADH-F according to ADH-S. Constant influence (during the life span of 25 generations) of elevated temperature on the population changes its genetic structure, due to selective advantages of the S allele of Adh under these conditions. 相似文献

It is established, that certain variants of replacement of chromosomes performed on wild (C-S, D) and mutant (cn, vg) lines as well as continuous backcrosses, leads to the change of heat resistance and activity of F- and S-allozymes of ADH in tissues of synthesized forms of drosophila. But nevertheless, the electrophoretic mobility of allozymes does not change. The activity, physico-chemical properties and multiple molecular forms of enzymes (alcohol dehydrogenase, superoxide dismutase, nonspecific alpha- and beta-esterases, hydroxide peptidohydrolase) were studied in ontogenesis of Drosophila inbred lines and their hybrids under conditions of high temperature (37-41 degrees C) and the presence in food of 10% ethanol. It was established that resistance of individuals to the effect of high temperature and alcohol, including manifestation of adaptive heterosis in hybrids not always depends on the level of the activity of enzymes analysed and is rather determined by allelic state of the appropriate structural genes. So, in conditions of the alcohol stress the individuals containing highly active F form of alcohol dehydrogenase have selective advantage and flie with hybrid F/S enzyme of higher activity and heat stability are more stable to the effect of high temperature. It is supposed that the complexes of adaptation genes (CGA) are formed in individuals of populations in response to the regular action of unfavourable environmental factors. These complexes condition optimal allelic control and most efficient regulation of enzyme activity in environment. Genotypic adaptation of individuals as well as occurrence of adaptive heterosis in hybrids seem likely to be connected with formation of CGA. 相似文献
