Xanthomonas maltophilia ATCC 17666 is an obligate aerobe that accumulates nitrite when grown on nitrate. Spectra of membranes from nitrate-grown cells exhibited b-type cytochrome peaks and A615-630 indicative of d-type cytochrome but no absorption peaks corresponding to c-type cytochromes. The nitrate reductase (NR) activity was located in the membrane fraction. Triton X-100-extracted reduced methyl viologen-NRs were purified on DE-52, hydroxylapatite, and Sephacryl S-300 columns to specific activities of 52 to 67 μmol of nitrite formed per min per mg of protein. The cytochrome-containing NRI separated on sodium dodecyl sulfate-polyacrylamide gel electrophoresis into a 135-kDa α-subunit, a 64-kDa β-subunit, and a 23-kDa γ-subunit with relative band intensities indicative of a 1:1:1 α/β/γ subunit ratio and a Mr of 222,000. The electronic spectrum of dithionite-reduced purified NR displayed peaks at 425, 528, and 558 nm, indicative of the presence of a cytochrome b, an interpretation consistent with the pyridine hemochrome spectrum formed. The cytochrome b of the NR was reduced under anaerobic conditions by menadiol and oxidized by nitrate with the production of nitrite. This NR contained 0.96 Mo, 12.5 nonheme iron, and 1 heme per 222 kDa: molybdopterin was detected with the Neurospora crassa nit-1 assay. A smaller reduced methyl viologen-NR (169 kDa), present in various concentrations in the Triton X-100 preparations, lacked a cytochrome spectrum and did not oxidize menadiol. The characteristics of the NRs and the absence of c-type cytochromes provide insights into why X. maltophilia accumulates nitrite.  相似文献

Samples of water, sediments and aquatic weeds were collected from 26 sites in the Nyando River catchment of the Lake Victoria basin in 2005–2006. The objective was to investigate levels of organochlorine pesticides that have either been banned or are restricted for use in Kenya. The pesticides investigated were lindane, aldrin, endosulfan, endrin, dieldrin, DDT, heptachlor and methoxychlor. These pesticides had previously found wide applications in public health and agriculture in Kenya for control of disease vectors and crop pests respectively. Results showed that mean concentrations were highest for methoxychlor (8.817 ± 0.020?µg l?1) in water, sediments (92.893 ± 3.039 µg kg?1), and weeds (39.641 ± 3.045?µg kg?1), the weeds also tended to accumulate aldrin (15.519 ± 3.756?µg kg?1). The results show that the pesticides are still in use and are detected in the catchment. Stringent management and public awareness measures are required to enforce the ban on the organochlorine pesticides in order to safeguard the environment and ecosystems of Lake Victoria.  相似文献

Adult Schistosoma monsoni grown in hamsters and transferred directly to the mesenteric vessels of C57BL10J mice previously immunized with hamster cells survive normally despite evidence of cytoxic and hemagglutinating antibodies directed against hamster cells. 1989~2009年在澳门山林和沿海滩涂湿地对鸟类进行了调查和记录,共记录鸟类297种,加上文献中记录的3种,整理出澳门鸟类名录共300种,隶属17目56科.其中非雀形目有30科160种,占澳门鸟类总种数的53.33%;雀形目有26科140种,占澳门鸟类种数46.67%.就数量来看,澳门鸟类中的优势种有9种,常见种44种,偶见种96种,稀有种则有151种.白腹海雕Haliaeetus leucogaster属于国家Ⅰ级重点保护野生鸟类.属于国家Ⅱ级重点保护的鸟类有33种.澳门鸟类属于<濒危野生动植物种国际贸易公约,2007>公约附录Ⅰ的有游隼Falco peregrinus peregrinus、小青脚鹬Tringa guttifer和小杓鹬Numenius minutus 3种,属于公约附录Ⅱ的有29种.勺嘴鹬Eurynorhynchus pygmeus被世界自然保护联盟(2009)列入极危(CR)鸟类,黑脸琵鹭Platalea minor和小青脚鹬被列入濒危(EN)鸟类.易危(VU)鸟类有4种,近危(NT)鸟类有5种.本文成果可为今后长期监测和保护澳门鸟类资源提供依据.  相似文献

Key aspects of lysosomal function are affected by the ionic content of the lysosomal lumen and, therefore, by the ion permeability in the lysosomal membrane. Such functions include regulation of lysosomal acidification, a critical process in delivery and activation of the lysosomal enzymes, release of metals from lysosomes into the cytoplasm and the Ca2+-dependent component of membrane fusion events in the endocytic pathway. While the basic mechanisms of lysosomal acidification have been largely defined, the lysosomal metal transport system is not well understood. TRPML1 is a lysosomal ion channel whose malfunction is implicated in the lysosomal storage disease Mucolipidosis Type IV. Recent evidence suggests that TRPML1 is involved in Fe2+, Ca2+ and Zn2+ transport across the lysosomal membrane, ascribing novel physiological roles to this ion channel, and perhaps to its relatives TRPML2 and TRPML3 and illuminating poorly understood aspects of lysosomal function. Further, alterations in metal transport by the TRPMLs due to mutations or environmental factors may contribute to their role in the disease phenotype and cell death.  相似文献

Sixty nine hammerhead sharks, Sphyrna lewini, were tagged at Malpelo Island (Colombia) with ultrasonic transmitters during March 2006, 2007 and 2008, as part of a study to understand their residency at the island and their horizontal and vertical movements. Five sharks visited Cocos Island, 627 km distant from Malpelo. One of the sharks that appeared at Cocos Island also visited the Galapagos Islands, 710 km from Cocos, a month later. There is connectivity of Sphyrna lewini between Malpelo, Cocos and the Galapagos Islands, but the frequency of movements between the islands appears to be relatively low (<7% of the tagged sharks). The most common depth at which the sharks swam coincided with the thermocline (rs = 0.72, p < 0.01). The depth of the thermocline varied depending on the time of the year. Nocturnal detections of the sharks were more frequent during the cold season than during the warm season (W = 60, p < 0.01). We also found that hammerheads spent significantly more time on the up-current side of the island (Kruskal-Wallis = 31.1008; p < 0.01). This study contributes to the knowledge of hammerhead sharks not only in Malpelo Island but also at a regional level in the Eastern Tropical Pacific.  相似文献

Sharks segregate by sex and size, but few studies have attempted to explain such behaviors. To address this, we examined aggregations and the foraging ecology of whale sharks in Bahía de La Paz (BLP) with aerial and ship surveys and direct observation. Zooplankton abundance and composition, and hydrographic conditions were analyzed in relation to whale shark occurrence to explore underlying factors causing segregations. We observed large aggregations of juveniles (<9 m total length, TL) inshore, comprised by 60 % male individuals, and small aggregations of adults (>9 m TL) offshore, composed of 84 % females. Juvenile sharks were associated to turbid shallow waters in BLP, where they performed stationary and dynamic suction feeding on dense copepod swarms. Adults occurred in oceanic waters and fed by ram-filtering on diffuse patches of euphausiids, with no association to oceanographic conditions. Such segregation may be advantageous to juvenile R. typus utilizing shallow coastal waters to find abundant preferred prey needed for their fast growth rates. Our studies suggest that the main driving forces of whale shark segregation by sex and size in BLP may be diet preference for juveniles and habitat preference for adult sharks.  相似文献

The trophic state of Lake Baringo and factors that could be limiting the development of algal biomass in it were investigated during one wet/dry hydrological cycle in 2014–2015. Water samples were analysed for dissolved inorganic nutrients, including , and , total phosphorus and Chlorophyll a. Light attenuation was estimated using Secchi depth. The trophic state was determined using Carlson trophic state indices (CTSI). Deviations in CTSI, nutrient ratios and ambient nutrient concentrations were used to identify factors limiting phytoplankton growth. The mean values measured for Secchi depth, nitrate, total phosphorus and Chlorophyll a showed significant seasonal variation (p < 0.05). Based on the Carlson trophic state index, the results show that Lake Baringo is eutrophic. However, the lake is also experiencing phosphorus limitation and poor light penetration, because of high turbidity, which is more pronounced during the wet season.  相似文献
