The initial stages of nerve outgrowth carried out by growth cones occur in three fundamental cyclic steps. Each of these steps appears to require myosin II activity to variable degrees. The steps include the following: (a) exploration, involving extensions and retractions that are driven and controlled by the interaction of actin retrograde flow and polymerization; (b) adhesion of new extensions to the substrate, which has been shown to be mediated by complex interactions between extracellular matrix proteins, cell adhesion proteins, and the actin cytoskeleton; and (c) traction force generated during forward advance of the growth cone, resulting in the production of tension on the neurite. 相似文献

1. The slime produced by eight strains of Pseudomonas aeruginosa on a number of different media was demonstrated to be qualitatively the same. Small quantitative differences may be occasioned by differences in the extraction procedure, the growth medium or the strain of organism used. 2. The slime was shown to be predominantly polysaccharide with some nucleic acid material and a small amount of protein. 3. The hydrolysed polysaccharide fraction consists mainly of glucose with smaller amounts of mannose. This accounts for some 50-60% of the total slime. In addition, there is some 5% of hyaluronic acid. The nucleic acid material represents approx. 20% of the total weight, and is composed of both RNA and DNA. 4. Minor components are protein, rhamnose and glucosamine, the protein being less than 5% of the total. 5. Hyaluronic acid is produced in greater quantities from nutrient broth than from chemically defined media, and is more firmly attached to the cells than the other components. 相似文献

Boron-deficient soybean roots have a rosette appearance at the terminals caused by the death of the root primordium and the initiation of new root primordia. The root tips show some darkening, swelling, and then a collapse of the tissue. The petiole s of B-deficient plants are very brittle. Physiologically, B-deficiency in HA-soybeans does not appear to involve an interference in the transport of photosynthate (C14 distribution) or precursors required for citrate synthesis in the roots. More Ca 45 was found in the tops of B-sufficient than B-deficient plants, but the opposite trend existed in the root sap. 1. Lipoproteins or defatted proteins from the membrane of Halobacterium halobium were dissolved in 0.02m-phosphate buffer, pH7.4, with or without urea (8m). Physicochemical measurements were made on these solutions with and without addition of sodium chloride. 2. In the presence of 8m-urea the lipoproteins could be resolved into two sedimenting groups. The faster group had an average apparent molecular weight of 390 000; sedimentation coefficients of the slower group could not be determined. 3. Proteins that had been defatted by extraction with a chloroform-methanol mixture could also be resolved into two groups distinguished by their sedimentation velocities. Sedimentation coefficients of both groups were readily determined. The average apparent molecular weight of the faster group of defatted proteins in 8m-urea was 340 000 and that of the slower group was 97 000. It is concluded that attachment of lipid does not affect the state of aggregation of the proteins in 8m-urea. 4. Intrinsic viscosities of lipoproteins and defatted proteins were higher than is customary for globular proteins and suggest expanded highly-solvated molecules, consistent with the ;denaturing' action on the proteins of solutions of low ionic strength. 相似文献
