Developing cardiac myocytes divide a limited number of times before they stop and terminally differentiate, but the mechanism that stops their division is unknown. To help study the stopping mechanism, we defined conditions under which embryonic rat cardiac myocytes cultured in serum-free medium proliferate and exit the cell cycle on a schedule that closely resembles that seen in vivo. The culture medium contains FGF-1 and FGF-2, which stimulate cell proliferation, and thyroid hormone, which seems to be necessary for stable cell-cycle exit. Time-lapse video recording shows that the cells within a clone tend to divide a similar number of times before they stop, whereas cells in different clones divide a variable number of times before they stop. Cells cultured at 33 degrees C divide more slowly but stop dividing at around the same time as cells cultured at 37 degrees C, having undergone fewer divisions. Together, these findings suggest that an intrinsic timer helps control when cardiac myocytes withdraw from the cell cycle and that the timer does not operate by simply counting cell divisions. We provide evidence that the cyclin-dependent kinase inhibitors p18 and p27 may be part of the timer and that thyroid hormone may help developing cardiac myocytes stably withdraw from the cell cycle. 相似文献

Xanthan with various pyruvic acid and acetate contents has been prepared from a single commercial polysaccharide sample using optimised chemical conditions (acid and alkali hydrolysis, respectively) for removal of acetal and acyl groups. The only significant change found on analysis of the modified xanthans was loss of pyruvic acid and/or acetate; no low moleculur weight carbohydrate-containing material was released. Contrary to some previous reports, evidence is presented to show that the pyruvic acid acetal and o-acetyl contents of xanthan do not affect solution viscosity. The viscosities of native, pyruvate-free and pyruvate/acetate-free xanthan solutions (0·3% w/v) were similar at shear rates 8·8–88·3 s?1 in both distilled water and 1% KCl. Over the concentration range 0·2-1·5%, the viscosities of native and pyruvate-free xanthan at 10 s?1 were similar. The viscosity increase on addition of 1% KCl to salt-free xanthan solutions was independent of pyruvic acid acetal substitution. Our results suggest that xanthan samples with various pyruvic acid acetal and o-acetal contents, prepared under different fermentation conditions of Xanthomonas campestri should not normally be used for assessing the contribution of these groups to solution viscosity. 相似文献

A new extracellular microbial polysaccharide, XM-6, has been isolated from cultures of an Enterobacter species and shows unusual gelation properties of potential technological significance. The polysaccharide contains d-glucose, l-fucose and d-glucuronate in the approximate molar ratio 3:1:1. No significant amounts of acetate or pyruvate were detected. d-Glucuronate and some d-glucose are destroyed on periodate oxidation, but l-fucose and some d-glucose may be recovered intact, indicating the presence of some 1,3 linkages in the primary structure. The major oligosaccharide isolated from autohydrolysates was an aldobiuronic acid containing equal amounts of d-glucose and l-fucose.Thermally-reversible gels are formed on addition of salt to solutions of the polysaccharide. A preliminary investigation of the mechanism of gelation by optical rotation, circular dichroism, high resolution n.m.r. and mechanical spectroscopy suggests interchain association through conformationally ordered ‘junction zones’, with specific incorporation of site-bound cations within the ordered structures. In the sol state the polysaccharide shows the shear-rate and temperature dependence of viscosity typical of a disordered (‘random coil’) polymer solution. Divalent cations are, in general, more effective than monovalent cations in promoting gelation of XM-6, while trivalent cations normally cause precipitation. Within Groups I and II, optimum gelation is achieved with Na+ and Ca2+ (ionic radius ? 0·1 nm), with larger and smaller ions becoming progressively less effective. Both gel strength and melting temperature increase with increasing salt concentration.XM-6 forms gels of reasonable strength at unusually low concentrations of the polysaccharide. For example, gels comparable to those required for normal industrial or food applications may be obtained using 0·3% w/v XM-6 and 1% w/v NaCl. Gel strength increases with increasing polymer concentration but there is no systematic variation in melting point. The sol-gel transition of XM-6 is unusually sharp and, by suitable adjustment of salt concentration, can be made to occur just below body temperature (e.g. 30–35°C), with obvious implications for biomedical or food applications. 相似文献

Background
Antiretroviral therapy (ART) has evolved rapidly since its beginnings. This analysis describes trends in first-line ART use in Asia and their impact on treatment outcomes.Methods
Patients in the TREAT Asia HIV Observational Database receiving first-line ART for ≥6 months were included. Predictors of treatment failure and treatment modification were assessed.Results
Data from 4662 eligible patients was analysed. Patients started ART in 2003–2006 (n = 1419), 2007–2010 (n = 2690) and 2011–2013 (n = 553). During the observation period, tenofovir, zidovudine and abacavir use largely replaced stavudine. Stavudine was prescribed to 5.8% of ART starters in 2012/13. Efavirenz use increased at the expense of nevirapine, although both continue to be used extensively (47.5% and 34.5% of patients in 2012/13, respectively). Protease inhibitor use dropped after 2004. The rate of treatment failure or modification declined over time (22.1 [95%CI 20.7–23.5] events per 100 patient/years in 2003–2006, 15.8 [14.9–16.8] in 2007–2010, and 11.6 [9.4–14.2] in 2011–2013). Adjustment for ART regimen had little impact on the temporal decline in treatment failure rates but substantially attenuated the temporal decline in rates of modification due to adverse event. In the final multivariate model, treatment modification due to adverse event was significantly predicted by earlier period of ART initiation (hazard ratio 0.52 [95%CI 0.33–0.81], p = 0.004 for 2011–2013 versus 2003–2006), older age (1.56 [1.19–2.04], p = 0.001 for ≥50 years versus <30years), female sex (1.29 [1.11–1.50], p = 0.001 versus male), positive hepatitis C status (1.33 [1.06–1.66], p = 0.013 versus negative), and ART regimen (11.36 [6.28–20.54], p<0.001 for stavudine-based regimens versus tenofovir-based).Conclusions
The observed trends in first-line ART use in Asia reflect changes in drug availability, global treatment recommendations and prescriber preferences over the past decade. Objectives
Restoration of Cytomegalovirus-specific-CD4 T cell (CMV-Sp-CD4) responses partly accounts for the reduction of CMV-disease with antiretroviral-therapy (ART), but CMV-Sp-CD4 may also drive immune activation and immunosenescence. This study characterized the dynamics of CMV-Sp-CD4 after ART initiation and explored associations with CD4 T cell recovery as well as frequency of naïve CD4 T cells at week 96.Methods
Fifty HIV-infected, ART-naïve Thai adults with CD4 T cell count ≤350cells/µL and starting ART were evaluated over 96 weeks (ClinicalTrials.gov identifier NCT01296373). CMV-Sp-CD4 was detected by co-expression of CD25/CD134 by flow cytometry after CMV-antigen stimulation.Results
All subjects were CMV sero-positive, 4 had quantifiable CMV-DNA (range 2.3-3.9 log10 copies/mL) at baseline but none had clinically apparent CMV-disease. Baseline CMV-Sp-CD4 response was positive in 40 subjects. Those with CD4 T cell count <100cells/µL were less likely to have positive baseline CMV-Sp-CD4 response (P=0.003). Positive baseline CMV-Sp-CD4 response was associated with reduced odds of quantifiable CMV-DNA (P=0.022). Mean CD4 T cell increase at week 96 was 213 cells/µL. This was associated positively with baseline HIV-VL (P=0.001) and negatively with age (P=0.003). The frequency of CMV-Sp-CD4 increased at week 4 (P=0.008), then declined. Those with lower baseline CMV-Sp-CD4 (P=0.009) or CDC category C (P<0.001) had greater increases in CMV-Sp-CD4 at week 4. At week 96, CD4 T cell count was positively (P<0.001) and the frequency of CMV-Sp-CD4 was negatively (P=0.001) associated with the percentage of naïve CD4 T cells.Conclusions
Increases in CMV-Sp-CD4 with ART occurred early and were greater in those with more advanced immunodeficiency. The frequency of CMV-Sp-CD4 was associated with reduced naïve CD4 T cells, a marker associated with immunosenescence. 相似文献66.

Isochrysis galbana, one of the most widely usedmarine microalgae in the rearing of finfish and shellfish larvae, is masscultured frequently in outdoor tanks. Under prolonged and repeated culture,severe contamination occurs. Organismal dispersal through mountain passes should be more constrained by temperature‐related differences between lowland and highland sites in montane environments. This may lead to higher rates of diversification through isolation of existing lineages toward the tropics. This mechanism, proposed by Janzen, could influence broad‐scale patterns of biodiversity across mountainous regions and more broadly across latitudinal gradients. We constructed two complementary analyses to test this hypothesis. First, we measured topographically‐derived thermal gradients using recently‐developed climatic data across the Americas, reviewing the main expectations from Janzen's climatic model. Then, we evaluated whether thermal barriers predict assemblage similarity for amphibians and mammals along elevational gradients across most of their latitudinal extent in the Americas. Thermal barriers between low and high elevation areas, initially proposed to be unique to tropical environments, are comparably strong in some temperate regions, particularly along the western slopes of North American dividing ranges. Biotic similarity for both mammals and amphibians decreases between sites that are separated by elevation‐related thermal barriers. That is, the stronger the thermal barrier separating pairs of sites across the latitudinal gradient, the lower the similarity of their species assemblages. Thermal barriers explain 10–35% of the variation in latitudinal gradients of biotic similarity, effects that were stronger in comparisons of sites at high elevations. Mammals' stronger dispersal capacities and homeothermy may explain weaker effects of thermal barriers on gradients of assemblage similarity than among amphibians. Understanding how temperature gradients have shaped gradients of montane biological diversity in the past will improve understanding of how changing environments may affect them in the future. 相似文献

Reading disabilities (RD) are the most common neurocognitive disorder, affecting 5% to 17% of children in North America. These children often have comorbid neurodevelopmental/psychiatric disorders, such as attention deficit/hyperactivity disorder (ADHD). The genetics of RD and their overlap with other disorders is incompletely understood. To contribute to this, we performed a genome‐wide association study (GWAS) for word reading. Then, using summary statistics from neurodevelopmental/psychiatric disorders, we computed polygenic risk scores (PRS) and used them to predict reading ability in our samples. This enabled us to test the shared aetiology between RD and other disorders. The GWAS consisted of 5.3 million single nucleotide polymorphisms (SNPs) and two samples; a family‐based sample recruited for reading difficulties in Toronto (n = 624) and a population‐based sample recruited in Philadelphia [Philadelphia Neurodevelopmental Cohort (PNC)] (n = 4430). The Toronto sample SNP‐based analysis identified suggestive SNPs (P ~ 5 × 10?7) in the ARHGAP23 gene, which is implicated in neuronal migration/axon pathfinding. The PNC gene‐based analysis identified significant associations (P < 2.72 × 10?6) for LINC00935 and CCNT1, located in the region of the KANSL2/CCNT1/LINC00935/SNORA2B/SNORA34/MIR4701/ADCY6 genes on chromosome 12q, with near significant SNP‐based analysis. PRS identified significant overlap between word reading and intelligence (R2 = 0.18, P = 7.25 × 10?181), word reading and educational attainment (R2 = 0.07, P = 4.91 × 10?48) and word reading and ADHD (R2 = 0.02, P = 8.70 × 10?6; threshold for significance = 7.14 × 10?3). Overlap was also found between RD and autism spectrum disorder (ASD) as top‐ranked genes were previously implicated in autism by rare and copy number variant analyses.
