dTDP-6-deoxy-L-lyxo-4-hexulose reductase (RmlD) catalyzes the final step in the conversion of dTDP-D-glucose to dTDP-L-rhamnose in an NAD(P)H- and Mg2+-dependent reaction. L-rhamnose biosynthesis is an antibacterial target. The structure of RmlD from Salmonella enterica serovar Typhimurium has been determined, and complexes with NADH, NADPH, and dTDP-L-rhamnose are reported. RmlD differs from other short chain dehydrogenases in that it has a novel dimer interface that contains Mg2+. Enzyme catalysis involves hydride transfer from the nicotinamide ring of the cofactor to the C4'-carbonyl group of the substrate. The substrate is activated through protonation by a conserved tyrosine. NAD(P)H is bound in a solvent-exposed cleft, allowing facile replacement. We suggest a novel role for the conserved serine/threonine residue of the catalytic triad of SDR enzymes.  相似文献

Peri-partum posture and behaviour of gilts from lines selected for different components of efficient lean growth were studied to determine if behavioural changes may have been associated with the observed responses in reproductive performance. The proportions of time that gilts expressed defined posture and behaviour traits and the locations of their piglets were determined from video recordings of observations made at 5min intervals in the period extending from 2h pre-farrowing to 2h post-farrowing. The 137 gilts studied were from four pairs of Large White lines which had been divergently (high and low) selected for either daily food intake (DFI), lean food conversion efficiency (LFC), lean growth rate on ad libitum feeding (LGA) or lean growth rate on a restricted feeding scale (LGS).Almost all the significant (P<05) changes occurred in the LGS pair of lines. In the pre-farrowing period, relative to the low LGS gilts, high LGS gilts spent a higher proportion of their time lying on their sides (0.92 versus 0.69), and less time in the upright postures of standing, sitting or lying on their bellies (0.08 versus 0.33) and engaging in nesting behaviour (0.02 versus 0.10). During farrowing, high LGS gilts again lay on their sides more often than low LGS gilts (0.96 versus 0.80) and were upright less often (0.04 versus 0.20). High LGS gilts changed posture less often than low LGS gilts (0.05 versus 0.31) but were more often alert (0.79 versus 0.61). During farrowing, high LGS piglets were seen less often at their mother's head, back and vulva or at the creep than low LGS piglets (0.06 versus 0.15). Post-farrowing, there were no significant differences between the lines, almost all gilts lying on their sides with their piglets at the udder. Divergent selection for components of efficient lean growth rate on ad libitum feeding was not associated with consistent responses in gilt posture and behaviour or in piglet location. Selection for high lean growth on restricted feeding had effects on gilt posture and behaviour which may have been beneficial to her welfare and that of her piglets.  相似文献

Experiments were conducted with soybean (Glycine max [L.] Merr. cv `Ransom') plants to determine if diurnal rhythms in net carbon dioxide exchange rate (CER), stomatal resistance, and sucrose-phosphate synthase (SPS) activity persisted in constant environmental conditions (constant light, LL; constant dark DD) and to assess the importance of these rhythms to the production of nonstructural carbohydrates (starch, sucrose, and hexose). Rhythms in CER, stomatal resistance, and SPS activity were observed in constant environmental conditions but the rhythms differed in period length, amplitude, and phase. The results indicated that these photosynthetic parameters are not controlled in a coordinated manner. The activity of UDPG pyrophosphorylase, another enzyme involved in sucrose formation, did not fluctuate rhythmically in constant conditions but increased with time in plants in LL. In LL, the rhythm in CER was correlated positively with fluctuations in total chlorophyll (r = 0.810) and chlorophyll a (r = 0.791) concentrations which suggested that changes in pigment concentration were associated with, but not necessarily the underlying mechanism of, the rhythm in photosynthetic rate. Assimilate export rate, net starch accumulation rate, and leaf sucrose concentration also fluctuated in constant light. No single photosynthetic parameter was closely correlated with fluctuations in assimilate export during LL; thus, assimilate export may have been controlled by interactions among the endogenous rhythms in CER, SPS activity, or other metabolic factors which were not measured in the present study.  相似文献

This study aims to evaluate the significance of the changes of erythrocyte reduced glutathione (GSH) in the course of diabetes mellitus including the pre-diabetes stage and cardiovascular disease co-morbidity. A total of 222 participants (female:male, 107:115) were selected and their erythrocyte GSH levels were measured. The participants were divided into four groups: (i) control; (ii) those with blood glucose level > or =5.6 mmol/l but < 6.9 mmol/l as pre-diabetes mellitus with no other pathology; (iii) diabetes without co-morbidity; and (iv) those with diabetes mellitus and cardiovascular disease. Statistical analysis was by ANOVA followed by a Fisher's LSD post hoc test. We observed that GSH concentration was significantly different between groups (P < 0.04). The Fisher's post hoc test indicated significant differences in erythrocyte GSH levels between the pre-diabetes mellitus and diabetes mellitus groups compared to control (P < 0.005 and P < 0.05, respectively). A statistically significant change (P < 0.001) involving an initial fall followed by a rise in erythrocyte GSH levels was observed when diabetes mellitus and diabetes mellitus+cardiovascular disease groups were combined and assessed with respect to period of diabetes. We conclude that oxidative stress is already present in the pre-diabetes stage as determined by the fall in GSH, representing the initial phase of oxidative stress in diabetes mellitus progression. This finding provides evidence that antioxidant markers such as GSH could be a useful tool for pre-diabetes mellitus screening.  相似文献
