Using intra-abdominal miniature data loggers, we measured core body temperature in female springbok (Antidorcas marsupialis) of three colour morphs (black, normal and white), free-living in the Karoo, South Africa, for one year. During winter, white springbok displayed lower daily minimum body temperatures (37.4 ± 0.5 °C), than both black (38.1 ± 0.3 °C) and normal (38.0 ± 0.6 °C) springbok. During spring, black springbok displayed higher daily maximum body temperatures (40.7 ± 0.1 °C) than both white (40.2 ± 0.2 °C) and normal (40.2 ± 0.2 °C) springbok. These high maximum body temperatures were associated with larger daily amplitudes of nychthemeral rhythm of body temperature (2.0 ± 0.2 °C), than that of white (1.6 ± 0.1 °C) and normal (1.7 ± 0.2 °C) springbok. Biophysical properties of sample springbok pelts were consistent with these patterns, as the black springbok pelt showed lower reflectance in the visible spectral range, and higher heat load from simulated solar radiation, than did the pelts of the other two springbok. Black springbok had lower diurnal activity in winter, consistent with them having to forage less because their metabolic cost of homeothermy was lower, but were disadvantaged in hot periods. White springbok, by contrast, were more protected from solar heat load, but potentially less able to meet the energy cost of homeothermy in winter. Thus energy considerations may underlie the rarity of the springbok colour morphs.  相似文献

A microsatellite‐enriched library of Job's tears (Coix lacryma‐jobi L. var. Ma‐yuen Stapf) was constructed using a modified biotin–streptavidin capture method. After screening, 17 polymorphic microsatellites were used for diversity analysis among 30 Job's tears accessions. The number of alleles ranged from one to five alleles per locus with an average of 2.8 alleles. Expected heterozygosity (HE) and polymorphism information content (PIC) ranged from 0 to 0.676 and from 0 to 0.666, respectively. The newly developed microsatellite markers are expected to be very valuable tools for evaluation of genetic diversity among large germplasm collection of Job's tears present in our Korean Gene Bank.  相似文献

In experiments involving different germ-cell stages, we had previously found meiotic prophase of the male mouse to be vulnerable to the induction of several types of genetic damage by the topoisomerase-II inhibitor etoposide. The present study of etoposide effects involved two end points of meiotic events known to occur in primary spermatocytes--chromosomal crossing-over and segregation. By following assortment of 13 microsatellite markers in two chromosomes (Ch 7 and Ch 15) it was shown that etoposide significantly affected crossing-over, but did not do so in a uniform fashion. Treatment generally changed the pattern for each chromosome, leading to local decreases in recombination, a distal shift in locations of crossing-over, and an overall decrease in double crossovers; at least some of these results might be interpreted as evidence for increased interference. Two methods were used to explore etoposide effects on chromosome segregation: a genetic experiment capable of detecting sex-chromosome nondisjunction in living progeny; and the use of FISH (fluorescence in situ hybridization) technology to score numbers of Chromosomes X, Y, and 8 in spermatozoa. Taken together these two approaches indicated that etoposide exposure of pachytene spermatocytes induces malsegregation, and that the findings of the genetic experiment probably yielded a marked underestimate of nondisjunction. As indicated by certain segregants, at least part of the etoposide effect could be due to disrupted pairing of achiasmatic homologs, followed by precocious sister-centromere separation. It has been shown for several organisms that absent or reduced levels of recombination, as well as suboptimally positioned recombination events, may be associated with abnormal segregation. Etoposide is the only chemical tested to date for which living progeny indicates an effect on both male meiotic crossing-over and chromosome segregation. Whether, however, etoposide-induced changes in recombination patterns are direct causes of the observed malsegregation requires additional investigation.  相似文献

Recent findings have suggested that there has been a change in the structural nature of forests in the Eastern Cape, South Africa, which explains decreases in forest fauna. One of these (rare) forest species is the tree hyrax Dendrohyrax arboreus, a nocturnal, arboreal folivore, which makes use of cavity‐bearing trees as dens. However, a conflict exists between the use of forest resources and in protecting habitats for the tree hyrax. In order to design appropriate management strategies, conservation authorities and forest managers require information regarding the specific habitat requirements of fauna in their forest, particularly those that are already threatened by resource use. The aims of this study were thus: (i) to characterise the den trees of D. arboreus, (ii) to determine whether D. arboreus selects for certain den tree characteristics (i.e. relative to the abundance of that characteristic); and (iii) to develop a habitat assessment model for D. arboreus. The tree hydrax was found to select for den trees with particular characteristics: seven tree species were selected as den trees, which were usually the tallest trees in the canopy (4–8 m). Den trees were usually only partly decayed, with multiple cavity entrances and trunk angles of between 45° and 68°. Cavity entrance and orientation did not appear to play a role in den tree selection by D. arboreus. A function which discriminated between den trees and non‐den trees was calculated, and can be used to determine the suitability of a tree as a den for a tree hyrax (and thus to prevent it from being removed during commercial logging operations), or to assess the suitability of an area for habitation by tree hyraxes. This model can therefore help to alleviate the conflict between forest conservation managers and resource users in the Eastern Cape.  相似文献

The ability of Lupinus albus L. to adapt to a heterogeneous soil profile containing acid subsoil below limed topsoil of the same type, and to utilize nutrients by significantly altering its root system structure, was investigated using specially constructed soil profile tubes. Plants grown in homogeneous acid profiles had the fastest growth while those grown in homogeneous limed-soil profiles showed the slowest growth and exhibited some chlorosis after 19 days. Limed topsoil combined with an acid subsoil profile initially retarded plant growth similar to that in a homogeneous limed soil. However, after 68 days significantly greater growth had occurred in the limed/acid soil treatment relative to the homogeneous limed soil, indicating plants had benefited from the acid subsoil stratum. Plants in the homogeneous limed soil profile had lower concentrations of P, Fe and Mn in shoots compared with those in heterogeneous soils. In contrast, the concentration of Ca increased by 74%, due mainly to an increase in the water-soluble Ca fraction. When grown in a heterogeneous limed/acid soil profile, concentrations of P, Ca, K, Mg, Fe, Mn and Zn in shoots were comparable to those grown in a soil with a homogeneous acid profile. Although total root production was lower in the homogeneous limed-soil profile compared to the acid-soil containing profiles, cluster root mass was maintained at a level comparable with that in acid soil. The roots in heterogeneous soil profiles exhibited extensive plasticity, demonstrating a root-type specific, morphological response to the soil conditions. Within the acid subsoil of a heterogeneous profile, there was a large increase in cluster root mass compared with non-cluster roots. The proliferation of cluster roots in acid soil below limed topsoil may enhance the plant's ability to exploit this soil and facilitate the cultivation of L. albus on limed soil. The inhibitory action of tetracycline on the development of embryonic mouse incisors cultured in vitro was examined. Explants exposed to tetracycline were severely inhibited in development. In contrast, tooth germs cultured in the presence of both tetracycline and iron escaped inhibition and attained a stage of development which compared favorably with the controls.  相似文献

Understanding and monitoring the population status of endangered species is vital for developing appropriate management interventions. We used noninvasive genetic analyses to obtain ecological and genetic data on the last remaining Far Eastern leopard population in the world. During seven winters from 2000–2001 to 2007–2008, we collected feces, hair, and saliva from most of the leopard habitat. Of the 239 leopard samples collected during the study period, 155 were successfully genotyped at 13 microsatellite loci and 37 individuals (18 males and 19 females) were identified. Population size estimates based on the Capwire model were 28 (95 % CI 19–38) in 2002–03 and 26 (95 % CI 13–33) in 2007–2008. The leopard population had a low level of genetic diversity (expected and observed heterozygosity = 0.43; average number of alleles per locus = 2.62), and effective population size was estimated to be low (N e = 7–16) by two genetic-based methods. We observed little improvement in the genetic diversity during the study period and did find an indication of allele loss compared with individuals from the mid-1990s, suggesting that the remaining population will continue to suffer loss of genetic diversity. Given the small population size and the low genetic diversity, with little expectation of replenishment of the genetic variation by natural immigration, successful expansion of available habitat and development of a second population based on captive individuals may be crucial for persistence of this leopard subspecies in the wild.  相似文献
