Calves derived from IVP embryos may suffer from the large offspring syndrome that has been related to effects of in vitro culture on the intrinsic quality of the embryo. Limited information is available on the role of the placenta in such cases. In this study, bovine pregnancy-associated glycoprotein (bPAG) was used as a marker to test whether placental function is influenced by the route of embryo production. Therefore, from day 7 until day 119 of ongoing gestations, resulting from transfer of MOET (n = 53), IVP-co-culture (n = 21) and IVP-SOF (n = 38) embryos, bPAG levels were compared in peripheral plasma of recipients. Plasma progesterone levels were compared as well. From day 25 of gestation onwards, bPAG could be detected in all recipients and the levels were significantly influenced by the day of gestation. Although IVP calves were significantly heavier than the in vivo produced calves, this difference was not reflected in the bPAG profiles of the embryo production groups. Yet, the mean bPAG level of the three last sampling moments (days 105-119) tended to be positively related to the birth weight of the calves, irrespective of the embryo production technique. Progesterone concentrations were not influenced by route of embryo production, but were significantly affected by parity of the recipient and day of gestation.  相似文献

Periviscerokinins (PVKs) are a distinct insect peptide family with unusual distribution in the central nervous system and neurohemal release sites. PVKs were first isolated from the abdominal perisympathetic organs of Periplaneta americana, but can be found in other insect species. Peptides with structural similarity to PVKs have been identified through searches of the Drosophila genome. The cardioacceleratory peptide CAP(2b) of the hawkmoth Manduca sexta shares close amino acid identity with the PVKs and may thus be included as a structural member of the PVK peptide family. In this review, we provide support for grouping CAP(2b) as a PVK family member based on published sequences, and new immunocytochemical findings and mass spectrometric data.  相似文献

In most bird species, pairs copulate many times before egg laying. The exact function of repeated inseminations (i.e. successful copulations) is unknown, but several suggestions have been made. We tested the hypothesis that repeated inseminations are required to ensure fertilization of eggs, by using an experimental method where free-ranging male collared flycatchers (Ficedula albicollis) were prevented from inseminating their mates. We show that egg fertility was lower when females had not copulated during the studied part of their fertile period. By counting sperm on the inner perivitelline layer of eggs, we estimated that a minimum of 86 sperm must reach the site of fertilization to ensure average fertility. Using the timing of inseminations and the numbers of sperm on successive eggs, we show that repeated copulations are necessary to achieve an average rate of fertilization of a single clutch. Our results thus provide evidence that repeated inseminations function to ensure fertilization success. We discuss possible constraints on sperm production and utilization that may have contributed to this pattern.  相似文献

The effect of some more or less harmful compounds like Cd, Pb, Ni, Ti salts and DCMU at low concentrations on the development of chloroplasts in maize and bean seedlings was investigated. Chlorophyll content, chlorophyll a/b ratio, photosynthetic activity (14CO2 fixation), chlorophyll-protein composition of thylakoid membranes, fluorescence spectra of chloroplasts, fluorescence induction parameters of leaves and electron microscopic structure of maize and bean chloroplasts as well as growth parameters were studied. Stimulation of chlorophyll synthesis and photosynthetic activity was observed at different intervals during all of the treatments, while chlorophyll a/b ratios and fluorescence properties of leaves or chloroplasts did not change considerably except in DCMU treated plants. Heavy metal treatments increased the amount of photosystem I and light-harvesting complex II, while decreased amount of photosystem I and higher amount of light-harvesting complex II was found in DCMU treated thylakoids. Electron microscopy showed only sligth differences in the morphology of chloroplast lamellar system (mostly in DCMU treated plants), while the status of the plasmalemma and tonoplast seemed to be altered as a result of certain metal treatments. Results showed the expression of a cytokinin-like effect on the development of chloroplasts. It is assumed, that these low-dose stressors generate non-specific alarm reactions in plants, which may involve changes of the hormonal balance.  相似文献

Ecological and comparative taxonomic investigations were carried out on 49 Aeromonas strains isolated from water samples of two moderately alkaline lakes of Hungary, Lake Balaton and Lake Fertó/Neusiedlersee together with 3 authentic strains of Aeromonas hydrophila. Five phena were created at greater than 92% similarity value using the UPGMA method with the Jaccard coefficient. Strains isolated from Lake Balaton were determined as A. hydrophila, while strains originated from Lake Fertó were identified as A. hydrophila and A. sobria. The Fertó isolates of A. hydrophila grew only at higher salt concentration (5% NaCl). This might be an adaptation to the higher salt contents in the water of Lake Fertó. However, no specific differences were detected in their behaviour against alkaline pH values. The wide range of their degradative enzymes indicate that aeromonads can play an important role in nutrient cycling.  相似文献

Functional morphology of Helix pomatia salivary gland cells was studied at light microscopic level by using different histochemical methods. Three cell types could be demonstrated in the salivary gland: mucocytes, granular and vacuolated cells. The distribution and the number of the different cell types were different in active and inactive snails. In active feeding animals, dilatated interlobular salivary ducts were observed, which were never present in inactive ones. In active animals an additional cell type, the cystic cell could also be observed. Periodic acid Schiff staining revealed both mucuos and serous elements in the salivary gland. Furthermore, hematoxyline-eosin staining indicated the occurrence of a cell layer with high mitotic activity in the acini. Applying immunohistochemical methods with monoclonal mouse anti-human Ki-67 clone, B56 and polyclonal rabbit anti-human Ki-67 antibodies, we also were able to demonstrate the occurrence of dividing cells in the salivary gland. Analysis of 1-2 microm semi-thin Araldite sections stained with toluidine-blue showed that the saliva can be released, in addition to possible exocytosis, by the lysis of cystic cells. Using an apoptosis kit, we could also establish that this process was due to rather an apoptotic than a necrotic mechanism. In the salivary gland of active snails, where an intensive salivation takes place, significantly more apoptotic cells occurred, if compared to that of inactive animals. It is suggested that programmed cell death may also be involved in the saliva release.  相似文献

Dogs' ability to recognise cues of human visual attention was studied in different experiments. Study 1 was designed to test the dogs' responsiveness to their owner's tape-recorded verbal commands (Down!) while the Instructor (who was the owner of the dog) was facing either the dog or a human partner or none of them, or was visually separated from the dog. Results show that dogs were more ready to follow the command if the Instructor attended them during instruction compared to situations when the Instructor faced the human partner or was out of sight of the dog. Importantly, however, dogs showed intermediate performance when the Instructor was orienting into 'empty space' during the re-played verbal commands. This suggests that dogs are able to differentiate the focus of human attention. In Study 2 the same dogs were offered the possibility to beg for food from two unfamiliar humans whose visual attention (i.e. facing the dog or turning away) was systematically varied. The dogs' preference for choosing the attentive person shows that dogs are capable of using visual cues of attention to evaluate the human actors' responsiveness to solicit food-sharing. The dogs' ability to understand the communicatory nature of the situations is discussed in terms of their social cognitive skills and unique evolutionary history.  相似文献

The effect of low concentrations of some stress-inducing compounds like Cd, Pb, Ni, and Ti salts and DCMU on the senescence of chloroplasts was investigated in detached primary leaves of bean. After the petioles of ageing leaves had developed roots, these low-dose stressors stimulated chlorophyll synthesis and photosynthetic activity, as compared to the control, thus causing rejuvenation in treated leaves. The amount of photosystem I (lowest in DCMU-treated leaves) and light-harvesting complex II increased, while that of photosystem II decreased or remained unchanged. Fluorescence induction parameters indicated unchanged electron transport (except for DCMU treatment). CO2 fixation and, in some cases, starch accumulation was stimulated. In parallel, the occurrence of large plastoglobuli seemed to decrease in plastids of heavy metal-treated leaves. A cytokinin bioassay of leaf extracts confirmed the cytokinin-mediated effect of low-dose stressors, as the slopes of Chl and cytokinin curves were similar during the rejuvenation process. It is assumed that these stressors generate non-specific alarm reactions, which involve changes in the hormonal balance by increasing the synthesis of cytokinins.  相似文献

The dramatic increase in the mortality of lip- and oral cancers in Hungary in the last decades points to the importance of primary and secondary prevention. Stomato-oncological screening examinations belong to the latter category, and might represent useful tools in the early diagnosis and treatment of oral carcinomas and precancerous lesions. The aim of the paper is to review the methods, results and effectivity of stomato-oncological screening examinations in Hungary. Between 1962 and 2000 nine screening examinations were performed: one on a population sample, one in an industrial setting, four connected to X-ray lung-screening examinations (one with the help of a mobile unit), one on voluntary persons, one on high risk people (homeless), one in general medical practice. Among these, in the last five years, in the course of the stomato-oncological examination of 17325 individuals, oral carcinoma has been found in 0.12%, and oral precanceroses in 2.63%. Although the general dentist is obliged by law to perform a stomato-oncological examination on the patients appearing in the practice, unfortunately, about 50-to-90% of the population does not visit a dentist regularly. The regular examination of these - high risk - groups by the help of the above methods, including the help of general medical practitioners is highly recommended.  相似文献
