Cerebral malaria is the most severe complication of Plasmodium falciparum infection, and a leading cause of death in children under the age of five in malaria-endemic areas. We report high therapeutic efficacy of a novel formulation of liposome-encapsulated water-soluble glucocorticoid prodrugs, and in particular β-methasone hemisuccinate (BMS), for treatment of experimental cerebral malaria (ECM), using the murine P. berghei ANKA model. BMS is a novel derivative of the potent steroid β-methasone, and was specially synthesized to enable remote loading into nano-sterically stabilized liposomes (nSSL), to form nSSL-BMS. The novel nano-drug, composed of nSSL remote loaded with BMS, dramatically improves drug efficacy and abolishes the high toxicity seen upon administration of free BMS. nSSL-BMS reduces ECM rates in a dose-dependent manner and creates a survival time-window, enabling administration of an antiplasmodial drug, such as artemisone. Administration of artemisone after treatment with the nSSL-BMS results in complete cure. Treatment with BMS leads to lower levels of cerebral inflammation, demonstrated by changes in cytokines, chemokines, and cell markers, as well as diminished hemorrhage and edema, correlating with reduced clinical score. Administration of the liposomal formulation results in accumulation of BMS in the brains of sick mice but not of healthy mice. This steroidal nano-drug effectively eliminates the adverse effects of the cerebral syndrome even when the treatment is started at late stages of disease, in which disruption of the blood-brain barrier has occurred and mice show clear signs of neurological impairment. Overall, sequential treatment with nSSL-BMS and artemisone may be an efficacious and well-tolerated therapy for prevention of CM, elimination of parasites, and prevention of long-term cognitive damage.  相似文献

A collection of paromomycin-based di-alkylated cationic amphiphiles differing in the lengths of their aliphatic chain residues were designed, synthesized, and evaluated against 14 Gram positive pathogens that are known to cause skin infections. Paromomycin derivatives that were di-alkylated with C7 and C8 linear aliphatic chains had improved antimicrobial activities relative to the parent aminoglycoside as well as to the clinically used membrane-targeting antibiotic gramicidin D; several novel derivatives were at least 16-fold more potent than the parent aminoglycoside paromomycin. Comparison between a di-alkylated and a mono-alkylated paromomycin indicated that the di-alkylation strategy leads to both an improvement in antimicrobial activity and to a dramatic reduction in undesired red blood cell hemolysis caused by many aminoglycoside-based cationic amphiphiles. Scanning electron microscopy provided evidence for cell surface damage by the reported di-alkylated paromomycins.  相似文献

目的研究生长休止蛋白7(Gas7)在成年大鼠肾脏、心脏和肝脏的表达。方法成年SD大鼠16只,分别采用逆转录聚合酶链反应(RT-PCR)方法和免疫组织化学方法检测Gas7基因mRNA和蛋白在成年SD大鼠肾脏、心脏和肝脏的表达,并进行图像分析和统计学处理。结果RT—PCR结果显示,Gas7mRNA在肾脏高表达,在心脏的表达弱于肾脏(P〈0.05),而在肝脏的表达最弱,基本检测不到。免疫组化结果显示,在肾脏中,Gas7免疫阳性产物在近髓肾单位的近曲小管呈强阳性反应,在集合管表达较弱,在肾小球和其余肾小管未见表达;在心脏中,Gas7免疫阳性产物均匀分布于心肌细胞,呈中等强度反应,弱于肾脏(P〈O.05);在肝脏中,Gas7蛋白未见明显表达,与其mRNA在肝脏的表达相似。结论Gas7在大鼠肾脏、心脏和肝脏表达的不同,尤其在肾脏组织分布的差异性,提示Gas7在成年大鼠肾脏和心脏结构以及功能的维持中可能起着重要作用。  相似文献

目的观察不同时辰电针“足三里”和“三阴交”穴对氯胺酮成瘾大鼠内侧前额叶皮质(mPFC)酪氨酸羟化酶(TH)表达的影响,探讨电针治疗氯胺酮滥用成瘾的作用机制。方法将56只SD大鼠随机分为正常对照组、生理盐水对照组、模型组、电针治疗组,电针治疗组再分为子时(23:00)、卯时(05:00)、午时(11:00)、酉时(17:00)4个电针组,每组8只。每天1次经腹腔注射氯胺酮复制氯胺酮成瘾模型,不同时辰电针组在给药7d后分别选取一侧“足三里”和“三阴交”穴给予低频(2Hz)电针治疗,每次30min,连续治疗7d。采用免疫组织化学染色方法检测mPFC内TH的表达。结果与正常对照组和生理盐水对照组相比较,模型组mPFC内TH免疫反应阳性神经元的数量明显增多(P〈0.01),细胞平均灰度值降低(P〈0.01),与模型组相比较,午时、酉时电针组TH免疫反应阳性神经元的数量明显减少(P〈0.01),细胞平均灰度值升高(P〈0.01);子时、卯时电针组则无明显变化(P〉0.05)。结论氯胺酮成瘾使mPFC内TH的表达明显增加;午时、酉时电针“足三里”和“三阴交”穴可明显下调mPFC内TH的表达,改善氯胺酮成瘾症状。  相似文献

The mutualistic symbiosis between forest trees and ectomycorrhizal fungi (EMF) is among the most ubiquitous and successful interactions in terrestrial ecosystems. Specific species of EMF are known to colonize specific tree species, benefitting from their carbon source, and in turn, improving their access to soil water and nutrients. EMF also form extensive mycelial networks that can link multiple root‐tips of different trees. Yet the number of tree species connected by such mycelial networks, and the traffic of material across them, are just now under study. Recently we reported substantial belowground carbon transfer between Picea, Pinus, Larix and Fagus trees in a mature forest. Here, we analyze the EMF community of these same individual trees and identify the most likely taxa responsible for the observed carbon transfer. Among the nearly 1,200 EMF root‐tips examined, 50%–70% belong to operational taxonomic units (OTUs) that were associated with three or four tree host species, and 90% of all OTUs were associated with at least two tree species. Sporocarp 13C signals indicated that carbon originating from labelled Picea trees was transferred among trees through EMF networks. Interestingly, phylogenetically more closely related tree species exhibited more similar EMF communities and exchanged more carbon. Our results show that belowground carbon transfer is well orchestrated by the evolution of EMFs and tree symbiosis.  相似文献

Effective translation of the viral genome during the infection cycle most likely enhances its fitness. In this study, we reveal two different strategies employed by cyanophages, viruses infecting cyanobacteria, to enhance their translation efficiency. Cyanophages of the T7-like Podoviridae family adjust their GC content and codon usage to those of their hosts. In contrast, cyanophages of the T4-like Myoviridae family maintain genomes with low GC content, thus sometimes differing from that of their hosts. By introducing their own specific set of tRNAs, they appear to modulate the tRNA pools of hosts with tRNAs that fit the viral low GC preferred codons. We assessed the possible effects of those viral tRNAs on cyanophages and cyanobacterial genomes using the tRNA adaptation index, which measures the extent to which a given pool of tRNAs translates efficiently particular genes. We found a strong selective pressure to gain and maintain tRNAs that will boost translation of myoviral genes when infecting a high GC host, contrasted by a negligible effect on the host genes. Thus, myoviral tRNAs may represent an adaptive strategy to enhance fitness when infecting high GC hosts, thereby potentially broadening the spectrum of hosts while alleviating the need to adjust global parameters such as GC content for each specific host.  相似文献

Precursor proteins that are imported from the cytosol into the matrix of mitochondria carry positively charged amphipathic presequences and cross the inner membrane with the help of vital components of the TIM23 complex. It is currently unclear which subunits of the TIM23 complex recognize and directly bind to presequences. Here we analyzed the binding of presequence peptides to purified components of the TIM23 complex. The interaction of three different presequences with purified soluble domains of yeast Tim50 (Tim50IMS), Tim23 (Tim23IMS), and full-length Tim44 was examined. Using chemical cross-linking and surface plasmon resonance we demonstrate, for the first time, the ability of purified Tim50IMS and Tim44 to interact directly with the yeast Hsp60 presequence. We also analyzed their interaction with presequences derived from precursors of yeast mitochondrial 70-kDa heat shock protein (mHsp70) and of bovine cytochrome P450SCC. Moreover, we characterized the nature of the interactions and determined their KDs. On the basis of our results, we suggest a mechanism of translocation where stronger interactions of the presequences on the trans side of the channel support the import of precursor proteins through TIM23 into the matrix.  相似文献

Carbonyl sulfide (COS) and C(18)OO exchange by leaves provide potentially powerful tracers of biosphere-atmosphere CO(2) exchange, and both are assumed to depend on carbonic anhydrase (CA) activity and conductance along the diffusive pathway in leaves. We investigated these links using C(3) and C(4) plants, hypothesizing that the rates of COS and C(18)OO exchange by leaves respond in parallel to environmental and biological drivers. Using CA-deficient antisense lines of C(4) and C(3) plants, COS uptake was essentially eliminated and discrimination against C(18)OO exchange ((18)Δ) greatly reduced, demonstrating CA's key role in both processes. (18)Δ showed a positive linear correlation with leaf relative uptake (LRU; ratio of COS to CO(2) assimilation rates, A(s)/A(c), normalized to their respective ambient concentrations), which reflected the effects of stomatal conductance on both COS and C(18)OO exchange. Unexpectedly, a decoupling between A(s) and (18)Δ was observed in comparing C(4) and C(3) plants, with a large decrease in (18)Δ but no parallel reduction in A(s) in the former. This could be explained by C(4) plants having higher COS concentrations at the CA site (maintaining high A(s) with reduced CA) and a high phosphoenolpyruvate carboxylase/CA activity ratio (reducing (18)O exchange efficiency between CO(2) and water, but not A(s)). Similar A(s) but higher A(c) in C(4) versus C(3) plants resulted in lower LRU values in the former (1.16 ± 0.20 and 1.82 ± 0.18 for C(4) and C(3), respectively). LRU was, however, relatively constant in both plant types across a wide range of conditions, except low light (<191 μmol photon m(-2) s(-1)).  相似文献

In the foraging game between gerbils and their predators, gerbils manage risk of predation using the tools of time allocation (where, when and for how long to forage) and vigilance. The optimal level of a forager's vigilance should be affected by its encounter rate with predators and the effectiveness of its vigilance in reducing mortality risk. The physical structure of the environment can alter the effectiveness of its vigilance and therefore alter its foraging behaviour. We tested this for gerbils at risk of predation from barn owls or foxes in a large vivarium. In particular, we reduced the effectiveness of vigilance by placing obstructions around feeding trays that blocked sight lines along either the vertical (vigilance directed against owls) or horizontal axis (vigilance directed against foxes), thereby changing the physical structure of the environment. In addition, we manipulated the presence of foxes and owls. In general, gerbils harvested fewer seeds, allocated less time to foraging in dangerous patches, and used more vigilance while foraging where and when risks were higher (i.e. in the presence of predators and in bright moonlight). Vertical and horizontal sightline treatments interacted synergistically to further raise perceived risk. These results imply that blocking sight lines reduces the effectiveness of vigilance, causing gerbils to use it less. Moreover, in the presence of a predator, the gerbils’ response to the blocked sightlines was more severe – harvesting less food and spending less time and vigilance – in the patches with the increased risk. This was especially so in the presence of the predator that was expected to most benefit from blocking that particular type of sight line: cover that blocked vertical sight lines was riskiest in the presence of owls, and cover that blocked horizontal sight lines was riskiest in the presence of foxes. These results strongly indicate the importance of sightlines and landscape features such as bushes in the risk management and forging decisions of gerbils, demonstrating that bush cover provides mixed blessing to gerbils by providing cover, but making vigilance ineffective.  相似文献
