A novel killer protein (Pkkp) secreted by a Pichia kluyveri strain isolated from an Algerian soil was active against food and beverage spoilage yeasts of the genera Dekkera, Kluyveromyces, Pichia, Saccharomyces, Torulaspora, Wickerhamomyces and Zygosaccharomyces. After purification by gel filtration chromatography Pkkp revealed an apparent molecular mass of 54 kDa with SDS-PAGE. Minimum inhibitory concentrations (MICs) of purified Pkkp exhibited a high in vitro activity against Dekkera bruxellensis (MICs from 64,000- to 256,000-fold lower than that exhibited by potassium metabisulphite) and Saccharomyces cerevisiae (MICs from 32,000- to 64,000- fold lower than potassium sorbate). No in vitro synergistic interactions (calculated by FIC index ? Σ FIC) were observed when Pkkp was used in combination with potassium metabisulphite, potassium sorbate, or ethanol. Pkkp exhibited a dose–response effect against D. bruxellensis and S. cerevisiae in a low-alcoholic drink and fruit juice, respectively. The results of the present study suggest that Pkkp could be proposed as a novel food-grade compound useful for the control of food and beverage spoilage yeasts.  相似文献

The annual cycles of nutrients in the Etang de Berre were studied in 1985. From measurements twice a month at the input points, daily data for the freshwater flow and from daily measurements at the output point, an estimation is proposed for a nutrient budget. Seasonal variability is important, and a biological control of nutrient availability (locally remineralized products) results in a hydrologically controlled system (nutrients carried by continental waters) which produces the very high microalgal biomass observed in the lagoon. The seasonal halocline brings the euphotic layer very close to the remineralization site: nutrients are then constantly available to the phytoplankton.  相似文献

Cellobiose dehydrogenases (CDHs) are extracellular glycosylated haemoflavoenzymes produced by many different wood-degrading and phytopathogenic fungi. Putative cellobiose dehydrogenase genes are recurrently discovered by genome sequencing projects in various phylogenetically distinct fungi. The genomes from the basidiomycete Coprinopsis cinerea and the ascomycete Podospora anserina were screened for candidate cdh genes, and one and three putative gene models were evidenced, respectively. Two putative cdh genes were selected and successfully expressed for the first time in Aspergillus niger. CDH activity was measured for both constructions (CDHcc and CDHpa), and both recombinant CDHs were purified to homogeneity and subsequently characterised. Kinetic constants were determined for several carbohydrates including β-1,4-linked di- and oligosaccharides. Optimal temperature and pH were 60 °C and 5 for CDHcc and 65–70 °C and 6 for CDHpa. Both CDHs showed a broad range of pH stability between 4 and 8. The effect of both CDHs on saccharification of micronized wheat straw by an industrial Trichoderma reesei secretome was determined. The addition of each CDH systematically decreased the release of total reducing sugars, but to different extents and according to the CDH concentration. Analytical methods were carried out to quantify the release of glucose, xylose and gluconic acid. An increase of glucose and xylose was measured at a low CDHcc concentration. At moderated and high CDHcc and CDHpa concentrations, glucose was severely reduced with a concomitant increase of gluconic acid. In conclusion, these results give new insights into the physical and chemical parameters and diversity of basidiomycetous and ascomycetous CDHs. These findings also demonstrated that CDH drastically influenced the saccharification on a natural substrate, and thus, CDH origin, concentration and potential enzymatic partners should be carefully considered in future artificial secretomes for biofuel applications.  相似文献

Cellular senescence, a stable proliferation arrest, is induced in response to various stresses. Oncogenic stress-induced senescence (OIS) results in blocked proliferation and constitutes a fail-safe program counteracting tumorigenesis. The events that enable a tumor in a benign senescent state to escape from OIS and become malignant are largely unknown. We show that lysyl oxidase activity contributes to the decision to maintain senescence. Indeed, in human epithelial cell the constitutive expression of the LOX or LOXL2 protein favored OIS escape, whereas inhibition of lysyl oxidase activity was found to stabilize OIS. The relevance of these in vitro observations is supported by in vivo findings: in a transgenic mouse model of aggressive pancreatic ductal adenocarcinoma (PDAC), increasing lysyl oxidase activity accelerates senescence escape, whereas inhibition of lysyl oxidase activity was found to stabilize senescence, delay tumorigenesis, and increase survival. Mechanistically, we show that lysyl oxidase activity favors the escape of senescence by regulating the focal-adhesion kinase. Altogether, our results demonstrate that lysyl oxidase activity participates in primary tumor growth by directly impacting the senescence stability.  相似文献

Disassembly of the nuclear lamina is essential in mitosis and apoptosis requiring multiple coordinated enzymatic activities in nucleus and cytoplasm. Activation and coordination of the different activities is poorly understood and moreover complicated as some factors translocate between cytoplasm and nucleus in preparatory phases. Here we used the ability of parvoviruses to induce nuclear membrane breakdown to understand the triggers of key mitotic enzymes. Nuclear envelope disintegration was shown upon infection, microinjection but also upon their application to permeabilized cells. The latter technique also showed that nuclear envelope disintegration was independent upon soluble cytoplasmic factors. Using time-lapse microscopy, we observed that nuclear disassembly exhibited mitosis-like kinetics and occurred suddenly, implying a catastrophic event irrespective of cell- or type of parvovirus used. Analyzing the order of the processes allowed us to propose a model starting with direct binding of parvoviruses to distinct proteins of the nuclear pore causing structural rearrangement of the parvoviruses. The resulting exposure of domains comprising amphipathic helices was required for nuclear envelope disintegration, which comprised disruption of inner and outer nuclear membrane as shown by electron microscopy. Consistent with Ca++ efflux from the lumen between inner and outer nuclear membrane we found that Ca++ was essential for nuclear disassembly by activating PKC. PKC activation then triggered activation of cdk-2, which became further activated by caspase-3. Collectively our study shows a unique interaction of a virus with the nuclear envelope, provides evidence that a nuclear pool of executing enzymes is sufficient for nuclear disassembly in quiescent cells, and demonstrates that nuclear disassembly can be uncoupled from initial phases of mitosis.  相似文献

Over the last decade, bamboo has emerged as an interesting plant for the treatment of various polluted waters using plant-based wastewater treatment systems. In these systems, nitrogen and phosphorous concentrations in wastewater can exceed plant requirements and potentially limit plant growth. The effects of two nutrient rates on the growth of seven bamboo species were assessed in a one-year experiment: Dendrocalamus strictus, Thyrsostachys siamensis, Bambusa tuldoides, Gigantochloa wrayi, Bambusa oldhamii, Bambusa multiplex and Bambusa vulgaris. Nutrient rates were applied with a 20:20:20 NPK fertilizer as 2.6 and 13.2 t.ha.yr?1NPK to three-year-old bamboo planted in 70 L containers. Morphological characters, photosynthetic responses, and NPK content in bamboo tissues were investigated. Under high-nutrient supply rate, the main trend observed was an increase of culm production but the culms’ diameters were reduced. For the seven species, the aboveground biomass yield tended to increase with high-nutrient rate. Increasing in nutrient rates also improved the photosynthetic activity which is consistent with the increase of nitrogen and phosphorus contents measured in plant tissues. All the bamboo species tested appears suitable for wastewater treatment purposes, but the species Bambusa oldhamii and Gigantochloa wrayi showed the higher biomass yield and nutrient removal.  相似文献

In January 1993, a 24 hour survey based on an hourly sampling was conducted in a station (1 m depth) located in the Ebrié lagoon, Côte d'lvoire, in order to determine short-term changes in seston quality and quantity and relate them to physical conditions. Water samples were collected just below the surface and at 0.9m depth to assess the seston distribution in the water column. Located in an estuarine environment, the study site was submitted to periodic effect of tides with low currents. Under certain conditions, sediment resuspension by wind-induced waves was observed in the shallow parts of this ecosystem. From the analysis of hydrodynamic processes (tide, wind and sedimentation rates), four basic situations were distinguished in the course of the study: a period with no wind and high sedimentation rates, a period characterized by wind-induced resuspension, followed by a period of sedimentation and finallly, a period without wind during the second flood tide. The particle size spectra were similar throughout the study, the distributions being unimodal (mode of 4 μm diameter). The finer particles (<10 μm) were resuspended over the whole water column at the beginning of the resuspension period, whereas the larger particles were resuspended if the wind speed exceeded 3 m s?1. Phytoplankton species were found to provide information on the physical factors controlling the pelagic system: small cells, such as those of cyanobacteria (Synechococcus sp.), were always present over the whole tidal cycles, whereas large motile cells were predominant in the water column during the resuspension period. The seston characteristics of this part of Ebrié lagoon are mainly controlled by tidal currents, but wind-induced resuspension can modify the structure of the pelagic food web.  相似文献

A study of the comparative distribution of different types of bacterial populations in the sea area where the city of Marseille rejects its waste water shows that, when in contact with the marine environment, a great part of the allochthonic bacterial population undergoes physiological stress expressed by an abrupt reduction of the effectives and by weak values of the energetic charge. The presence of coliforms and streptococci in waste water lens, at important distances from discharging conduits, shows the possibility of a large diffusion for a fraction of the bacterial populations that withstand the initial stress. From mathematical analyses, an inverse correlation is revealed between detergents and the bacterial population as a whole, and between phenols and various allochthonic bacterial populations. The above results were evidenced through a sampling strategy based upon a 24-h-cycle study and a search for mathematical correlations between several biological, chemical, and physical parameters.  相似文献
