Discomycetes of the family Hyaloscyphaceae were collected from across Japan and identified. In this report, non-glassy-haired members of the tribe Hyaloscypheae, subfamily Hyaloscyphoideae are described and illustrated. The five species involved include one new species (Phialina lacrimiformis) and four species less well known or newly reported in Japan (Dematioscypha dematiicola, Hamatocanthoscypha laricionis var.Iaricionis, Micropodia chrysostigma, M. grisella). cultural studies were carried out, and teleomorph-anamorph relationships are discussed forD. dematiicola, H. laricionis val.laricionis and their anamorphs. Moellerodiscus advenulus, a sclerotiniaceous fungus new to Japan, was collected and its ascospore morphology and cultural characteristics were studied. Ascospores possess previously unreported characters, such as gelatinous polar appendages and a sheath, and become brown and one-septate after discharge, prior to germination. The stromata produced in culture have a thin black rind typical ofLambertella. The taxonomic position ofM. advenulus is discussed, and a new combination,L. advenula, is proposed.  相似文献

As we previously reported, 1alpha,25-dihydroxyvitamin D3 (1,25(OH)2D3) dose-dependently inhibited not only proliferation of undifferentiated murine erythroleukemia (MEL) cells but also activin A-induced erythroid differentiation of MEL cells. However, the effect of 1,25(OH)2D3 on MEL cell proliferation was significantly greater by one order of magnitude than that on differentiation (IC(50): 9.2 vs 0.8 nM, respectively). The response of activin A-treated mature MEL cells to 1,25(OH)2D3 in the induction of 1,25(OH)2D3-24-hydroxylase (24-OHase) activity, a rapid effect of 1,25(OH)2D3, was enhanced to the same degree as in untreated immature cells, suggesting that differences in capacity of cells to inactivate 1,25(OH)2D3 did not contribute to augmentation of 1,25(OH)2D3 effect in activin A-treated mature cells. Furthermore, neither the number nor the affinity of vitamin D receptors (VDR) differed significantly between activin A-treated cells and untreated immature cells. The intracellular cAMP level, which affects 1,25(OH)2D3-mediated induction of 24-OHase activity, was significantly less in activin A-treated mature cells than in immature MEL cells. The addition of dibutyryl cAMP (dbc AMP) to activin A-treated MEL cells dose-dependently attenuated 1,25(OH)2D3-mediated induction of 24-OHase activity, finally to a level comparable to that of the untreated cells at the final concentration of 100 nM dbcAMP, while dbcAMP itself by 100 nM did not affect MEL cell differentiation by 24 h. In summary, we have shown for the first time that 1,25(OH)2D3 exerted its effect on leukemia cells at physiological concentration and that the magnitude of this effect depended on the changes in intracellular cAMP level through stages of differentiation, suggesting that the cAMP-protein kinase A system may be useful as a target for clinical application of vitamin D analogs by improving the sensitivity of leukemic cells to 1,25(OH)2D3.  相似文献

1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) is known to be involved in regulating the proliferation of parathyroid cells and PTH synthesis through reactions involving its nuclear receptor. We evaluated the effects of 1,25-(OH)2D3 and its hexafluorinated analog, 26,26,26,27,27,27-hexafluoro-1,25-dihydroxyvitamin D3 (26,27-F6-1,25-(OH)2D3), on parathyroid cells. The 1,25-(OH)2D3 and 26,27-F6-1,25-(OH)2D3 each inhibited [3H]thymidine incorporation and ornithine decarboxylase (ODC) activity, which is important in cell proliferation, in primary cultured bovine parathyroid cells. The inhibitory effect of 26,27-F6-1,25-(OH)2D3 on PTH secretion from parathyroid cells was significantly more potent than that of 1,25-(OH)2D 3 between 10−11 M and 10−8 M. Study of 26,27-F6-1,25-(OH)2D3 metabolism in parathyroid cells in vitro elucidated its slower degradation than that of 1,25-(OH)2D3. After 48 h of incubation with [1β-3H]26,27-F6-1,25-(OH)2D3, two HPLC peaks, one for [1β-3H]26,27-F6-1,25-(OH)2D3, and a second larger peak for [1β-3H]26,27-F6-1,23(S),25-(OH)3D3, were detected. No metabolites were detected after the same period of incubation with 1,25-(OH)2[26,27-3H]D3. We observed that 26,27-F6-1,23(S),25-(OH)3D3 was as potent as 1,25-(OH)2D3 in inhibiting the proliferation of parathyroid cells.Data suggest that the greater biological activity of 26,27-F6-1,25-(OH)2D3 is explained by its slower metabolisms and by the retention of the biological potency of 26,27-F6-1,25-(OH)2D3 even after 23(S)-hydroxylation.  相似文献

The mechanism by which taurine (2‐aminoethanesulfonic acid) protects hepatocytes injury induced by carbon tetrachloride (CCl4) is not fully understood. In a previous study, we reported that cellular polyamines play an important role in this mechanism. The relationship between cellular glutathione (GSH), protein‐SH levels, and lactate dehydrogenase (LDH), with respect to the effect of polyamine on the cytoprotective ability of taurine in CCl4‐induced toxicity in isolated rat hepatocytes, was examined. CCl4 induced a LDH release and decreased cellular thiols and polyamine levels. Treating with taurine reversed these depletions. The effect of CCl4 was also reversed by the addition of exogenous polyamines. Pretreating with α‐difluoromethylornithine, an irreversible inhibitor of ornithine decarboxylase, which is a key enzyme in polyamine biosynthesis and therefore used to deplete cellular polyamine, prevented the protective effect of taurine. Adding diethyl maleate, a cellular glutathione‐depleting agent, reduced the effect of exogenous polyamines. The role of polyamine in the cytoprotective effect of taurine in CCl4‐induced toxicity may therefore be by preventing, among others, GSH and protein‐SH depletions. © 1998 John Wiley & Sons, Inc. High solar radiation has been recognised as a contributing factor to exertional heat-related illness in individuals exercising outdoors in the heat. Although solar radiation intensity has been known to have similar time-of-day variation as body temperature, the relationship between fluctuations in solar radiation associated with diurnal change in the angle of sunlight and thermoregulatory responses in individuals exercising outdoors in a hot environment remains largely unknown. The present study therefore investigated the time-of-day effects of variations in solar radiation associated with changing solar elevation angle on thermoregulatory responses during moderate-intensity outdoor exercise in the heat of summer. Eight healthy, high school baseball players, heat-acclimatised male volunteers completed a 3-h outdoor baseball trainings under the clear sky in the heat. The trainings were commenced at 0900 h in AM trial and at 1600 h in PM trial each on a separate day. Solar radiation and solar elevation angle during exercise continued to increase in AM (672–1107 W/m2 and 44–69°) and decrease in PM (717–0 W/m2 and 34–0°) and were higher on AM than on PM (both < 0.001). Although ambient temperature (AM 32–36°C, PM 36–30°C) and wet-bulb globe temperature (AM 31–33°C, PM 34–27°C) also continued to increase in AM and decrease in PM, there were no differences between trials in these (both > 0.05). Tympanic temperature measured by an infrared tympanic thermometer and mean skin temperature were higher in AM than PM at 120 and 180 min (< 0.05). Skin temperature was higher in AM than PM at the upper arm and thigh at 120 min (< 0.05) and at the calf at 120 and 180 min (both < 0.05). Body heat gain from the sun was greater during exercise in AM than PM (< 0.0001), at 0–60 min in PM than AM (< 0.0001) and at 120–180 min in AM than PM (< 0.0001). Dry heat loss during exercise was greater at 0–60 min (< 0.0001), and lower at 60–120 min (< 0.05) and 120–180 min (< 0.0001) in AM than PM. Evaporative heat loss during exercise was greater in PM than AM at 120–180 min (< 0.0001). Total (dry + evaporation) heat loss at the skin was greater during exercise in PM than AM (< 0.0001), at 0–60 min in AM than PM (< 0.0001) and at 60–120 and 120–180 min in PM than AM (< 0.05 and 0.0001). Heart rate at 120–150 min was also higher in AM than PM (< 0.05). Neither perceived thermal sensation nor rating of perceived exertion was different between trials (both > 0.05). The current study demonstrates a greater thermoregulatory strain in the morning than in the afternoon resulting from a higher body temperature and heart rate in relation to an increase in environmental heat stress with rising solar radiation and solar elevation angle during moderate-intensity outdoor exercise in the heat. This response is associated with a lesser net heat loss at the skin and a greater body heat gain from the sun in the morning compared with the afternoon.  相似文献

Reproduction of tropical species beyond their geographic range associated with ocean warming is regarded as the key indicator of a range shift. However, the lack of historical breeding records poses challenges for detecting distinct range shifts of tropical fishes. To obtain baseline data of the current status of the occurrence and breeding activity of tropical pomacentrid and apogonid fishes in ocean warming hotspots of temperate reefs (Kochi and Wakayama, 33°N) of Japan, we conducted a two-year underwater visual survey and synthesized those data with recently published information. By combining data from the present as well as past studies, the results confirmed the occurrence of 52 pomacentrid and 34 apogonid species, whereas breeding activity was confirmed for 19 and 16 species, respectively. Species richness and abundance of recruitment periphery and breeding active species were high at the warmer site adjacent to the Kuroshio Current. Most observed species were found beyond their known geographic range. Some species showing active breeding were widespread tropical fishes (e.g., Amphiprion clarkii, Pomacentrus coelestis and Apogon notatus) and probably have established breeding populations irrespective of recent global warming. The winter sea water temperature around the study sites will continue to rise, increasing by >2 °C by the end of the century; therefore, our results are highly relevant and represent the first step to elucidate the potential range extension of tropical fishes into temperate reefs with climate change.  相似文献

The lactogenic hormone prolactin (PRL) regulates milk protein gene expression in mammary glands. To maintain homeostatic balance in the body, milk fat globule epidermal growth factor 8 (MFG-E8) is vital for phagocytic clearance of apoptotic cells. We investigated the effects of PRL on MFG-E8 expression in macrophages by evaluating its promoter function. Macrophages were stimulated with PRL, and the expression of MFG-E8 was determined using real-time PCR and Western blotting. The role of MFG-E8 on phagocytosis of apoptotic cells in PRL-treated macrophages was assessed using microscopy, while the response of PRL to MFG-E8 expression was evaluated using luciferase assay. Following treatment with PRL, significant up-regulations of the PRL receptor and MFG-E8 were observed in macrophages, though PRL-treated macrophages more efficiently engulfed apoptotic cells. The results of MFG-E8 promoter analysis showed considerable up-regulation of promoter activity in macrophages following PRL treatment and results from mutation analysis of the MFG-E8 promoter suggested that the C/EBPβ binding site was responsible for PRL-induced activation of the MFG-E8 promoter. C/EBPβ activity was found to be up-regulated in PRL-treated cells as revealed by an electrophoretic mobility shift assay (EMSA). In conclusion, PRL is a potent inducer of MFG-E8 expression in macrophages, while its effect is mediated by the presence of a responsive element in the MFG-E8 promoter.  相似文献
