Introduction
Atherosclerotic diseases are the leading cause of death worldwide. Biomarkers of atherosclerosis are required to monitor and prevent disease progression. While mass spectrometry is a promising technique to search for such biomarkers, its clinical application is hampered by the laborious processes for sample preparation and analysis.Methods
We developed a rapid method to detect plasma metabolites by probe electrospray ionization mass spectrometry (PESI-MS), which employs an ambient ionization technique enabling atmospheric pressure rapid mass spectrometry. To create an automatic diagnosis system of atherosclerotic disorders, we applied machine learning techniques to the obtained spectra.Results
Using our system, we successfully discriminated between rabbits with and without dyslipidemia. The causes of dyslipidemia (genetic lipoprotein receptor deficiency or dietary cholesterol overload) were also distinguishable by this method. Furthermore, after induction of atherosclerosis in rabbits with a cholesterol-rich diet, we were able to detect dynamic changes in plasma metabolites. The major metabolites detected by PESI-MS included cholesterol sulfate and a phospholipid (PE18:0/20:4), which are promising new biomarkers of atherosclerosis.Conclusion
We developed a remarkably fast and easy method to detect potential new biomarkers of atherosclerosis in plasma using PESI-MS.84.

We compared forest light environments between a primary lowland tropical rainforest and a rainforest selectively logged 50 years ago in the Pasoh Forest Reserve, Peninsular Malaysia using two different approaches to assess forest light environments, hemispherical canopy photographs and continuous measurements of forest photosynthetic photon flux density (PPFD) and showed clear evidence of the long-term impact of selective logging on forest light environments. The selectively logged forest canopy consisted of shorter and smaller crowns with less variations of height and crown area than the primary forest. From the canopy structural characteristics of the selectively logged forest, we predicted that the selectively logged forest has brighter and more homogeneous forest light than the primary forest. Both hemispherical canopy photographs and measurements of PPFD showed that the selectively logged forest had more open canopies and longer sunfleck time than the primary forest. A significantly smaller variance of canopy openness and a shorter autocorrelation range in the selectively logged forest than in the primary forest were found, indicating that the selectively logged forest had a less heterogeneous light environment spatially than the primary forest. Therefore our predictions were confirmed. The levels of expression of interferon-stimulated genes (ISGs) in liver are associated with response to treatment with pegylated interferon (PEG-IFN) plus ribavirin (RBV). However, associations between the responses of ISGs to IFN-based therapy and treatment efficacy or interleukin-28B (IL28B) genotype have not yet been determined. Therefore, we investigated the early responses of ISGs and interferon-lambdas (IFN-λs) in peripheral blood mononuclear cells (PBMCs) during PEG-IFN/RBV plus NS3/4 protease inhibitor (PI) therapy. We prospectively enrolled 50 chronic hepatitis C patients with HCV genotype 1, and collected PBMCs at baseline, 8 and 24 h after the initial administration of PEG-IFN/RBV/PI. Levels of mRNAs for selected ISGs and IFN-λs were evaluated by real-time PCR. All 31 patients with a favorable IL28B genotype and 13 of 19 with an unfavorable genotype achieved sustained virological responses (SVR). Levels of mRNA for A20, SOCS1, and SOCS3, known to suppress antiviral activity by interfering with the IFN signaling pathway, as well as IRF1 were significantly higher at 8 h in patients with an unfavorable IL28B genotype than in those with a favorable one (P = 0.007, 0.026, 0.0004, 0.0006, respectively), especially in the non-SVR group. Particularly, the fold-change of IRF1 at 8 h relative to baseline was significantly higher in non-SVR than in SVR cases with an unfavorable IL28B genotype (P = 0.035). In conclusion, levels of several mRNAs of genes suppressing antiviral activity in PBMCs during PEG-IFN/RBV/PI differed according to IL28B genotypes, paralleling treatment efficacy. 相似文献

countries to control aphids in greenhouses. As developmental arrest in diapause lowers the effectiveness of natural enemies,
we studied the effect of photoperiod and temperature on the incidence of diapause in a Japanese strain of A. aphidimyza by examining diapause induction under different day-length conditions in the laboratory. The critical day length for diapause
induction was determined to be 12.7 h at 20°C. Diapause incidence was completely prevented at 30°C even though the photoperiod
used was 11L13D. We also examined diapause induction under changing temperature conditions while maintaining the critical
day length (12.7L11.3D). Diapause incidence was 100% in both field and greenhouse conditions under alternating temperatures
of 20/16 or 25/16°C while the critical day length of 12.7 h was maintained. The Japanese strain of A. aphidimyza was sensitive to diapause entry from the first to the third noncocooned instar larval stages. Its eggs do not seem to be
sensitive to diapause induction. Our results suggest that constant short-day conditions for at least four days are needed

Stripe rust is a devastating fungal disease of wheat caused by Puccinia striiformis f. sp tritici (Pst). The WHEAT KINASE START1 (WKS1) resistance gene has an unusual combination of serine/threonine kinase and START lipid binding domains and confers partial resistance to Pst. Here, we show that wheat (Triticum aestivum) plants transformed with the complete WKS1 (variant WKS1.1) are resistant to Pst, whereas those transformed with an alternative splice variant with a truncated START domain (WKS1.2) are susceptible. WKS1.1 and WKS1.2 preferentially bind to the same lipids (phosphatidic acid and phosphatidylinositol phosphates) but differ in their protein-protein interactions. WKS1.1 is targeted to the chloroplast where it phosphorylates the thylakoid-associated ascorbate peroxidase (tAPX) and reduces its ability to detoxify peroxides. Increased expression of WKS1.1 in transgenic wheat accelerates leaf senescence in the absence of Pst. Based on these results, we propose that the phosphorylation of tAPX by WKS1.1 reduces the ability of the cells to detoxify reactive oxygen species and contributes to cell death. This response takes several days longer than typical hypersensitive cell death responses, thus allowing the limited pathogen growth and restricted sporulation that is characteristic of the WKS1 partial resistance response to Pst. 相似文献
