Plasma membranes from fusing embryonic muscle cells were assayed for phospholipase A activity to determine if this enzyme plays a role in cell fusion. The membranes were assayed under a variety of conditions with phosphatidylcholine as the substrate and no phospholipase A activity was found. The plasma membranes did contain a phosphatidic acid phosphatase which was optimally active in the presence of Triton X-100 and glycerol. The enzyme activity was constant from pH 5.2 to 7.0, and did not require divalent cations. Over 97% of the phosphatidic acid phosphatase activity was in the particulate fraction. The subcellular distribution of the phosphatidic acid phosphatase was the same as the distibutions of the plasma membrane markers, )-ATPase and the acetylcholine receptor, which indicates that this phosphatase is located exclusively in the plasma membranes. The effect of predation by fish and wading shore birds on the population dynamics of a nereid polychaete (Ceratonereis pseudoerythraeensis Hutchings & Turvey) was studied. The species occurs in high densities on the site studied probably owing to high nutrient input from a nearby sewage treatment farm. Predators were excluded from experimental areas with wire mesh cages. The effects of cage structures on predator behaviour, algal deposition and sediment depth and composition were studied, and the possibility that C. pseudoerythraeensis might migrate in or out of caged areas was experimentally tested. These effects appeared to be of minimal importance. Analyses of size classes of C. pseudoerythraeensis showed that the density of adult worms in control areas decreased relative to caged areas. This was attributed to predation, mainly by flounder and curlew sandpipers. Recruitment of juvenile C. pseudoerythraeensis was greater in control than caged treatments. The hypothesis that increased adult density decreases juvenile recruitment was confirmed by manipulating the densities of adults. The adult-recruit interaction effect apparently counter-balanced losses due to predators, so that if only the total densities of the polychaetes had been studied no effect of predation would have been apparent. 相似文献
1. Predation‐exclusion experiments have highlighted that top‐down control is pervasive in terrestrial communities, but most of these experiments are simplistic in that they only excluded a single group of predators and the effect of removal was evaluated on a few species from the community. The main goal of our study was to experimentally establish the relative effects of ants and birds on the same arthropod assemblage of canopy trees. 2. We conducted 1‐year long manipulative experiments in an organic citrus grove intended to quantify the independent effects of bird and ant predators on the abundance of arthropods. Birds were excluded with plastic nets whereas ants were excluded with sticky barriers on the trunks. The sticky barrier also excluded other ground dwelling insects, like the European earwig Forficula auricularia L. 3. Both the exclusion of ants and birds affected the arthropod community of the citrus canopies, but the exclusion of ants was far more important than the exclusion of birds. Indeed, almost all groups of arthropods had higher abundance in ant‐excluded than in control trees, whereas only dermapterans were more abundant in bird‐excluded than in control trees. A more detailed analysis conducted on spiders also showed that the effect of ant exclusion was limited to a few families rather than being widespread over the entire diverse spectrum of spiders. 4. Our results suggest that the relative importance of vertebrate and invertebrate predators in regulating arthropod populations largely depends on the nature of the predator–prey system. 相似文献
Background and AimsGlobal plant trait datasets commonly identify trait relationships that are interpreted to reflect fundamental trade-offs associated with plant strategies, but often these trait relationships are not identified when evaluating them at smaller taxonomic and spatial scales. In this study we evaluate trait relationships measured on individual plants for five widespread Protea species in South Africa to determine whether broad-scale patterns of structural trait (e.g. leaf area) and physiological trait (e.g. photosynthetic rates) relationships can be detected within natural populations, and if these traits are themselves related to plant fitness.MethodsWe evaluated the variance structure (i.e. the proportional intraspecific trait variation relative to among-species variation) for nine structural traits and six physiological traits measured in wild populations. We used a multivariate path model to evaluate the relationships between structural traits and physiological traits, and the relationship between these traits and plant size and reproductive effort.Key ResultsWhile intraspecific trait variation is relatively low for structural traits, it accounts for between 50 and 100 % of the variation in physiological traits. Furthermore, we identified few trait associations between any one structural trait and physiological trait, but multivariate regressions revealed clear associations between combinations of structural traits and physiological performance (R2 = 0.37–0.64), and almost all traits had detectable associations with plant fitness.ConclusionsIntraspecific variation in structural traits leads to predictable differences in individual-level physiological performance in a multivariate framework, even though the relationship of any particular structural trait to physiological performance may be weak or undetectable. Furthermore, intraspecific variation in both structural and physiological traits leads to differences in plant size and fitness. These results demonstrate the importance of considering measurements of multivariate phenotypes on individual plants when evaluating trait relationships and how trait variation influences predictions of ecological and evolutionary outcomes. 相似文献
Increasing rates of Anthropocene biodiversity extinctions suggest a possible sixth mass extinction event. Conservation planners are seeking effective ways to protect species, hotspots of biodiversity, and dynamic ecosystems to reduce and eventually eliminate the degradation and loss of diversity at the scale of genes, species, and ecosystems. While well-established, adequately enforced protected areas (PAs) increase the likelihood of preserving species and habitats, traditional placement methods are frequently inadequate in protecting biodiversity most at risk. Consequently, the Key Biodiversity Area (KBA) Partnership developed a set of science-based criteria and thresholds that iteratively identify sites where biodiversity is most in need of protection. KBA methodology has been rarely applied in the marine realm, where data are often extremely limited. We tested the feasibility of KBA population metrics in the Greater Caribbean marine region using occurrence and population data and threat statuses for 1669 marine vertebrates. These data identified areas where site-specific conservation measures can effectively protect biodiversity. Using KBA criteria pertaining to threatened and irreplaceable biodiversity, we identified 90 geographically unique potential KBAs, 34 outside and 56 within existing PAs. These provide starting points for local conservation managers to verify that KBA thresholds are met and to delineate site boundaries. Significant data gaps, such as population sizes, life history characteristics, and extent of habitats, prevent the full application of the KBA criteria to data-poor marine species. Increasing the rate and scope of marine sampling programs and digital availability of occurrence datasets will improve identification and delineation of KBAs in the marine environment.
Sequence similarity was used to predict the position of expressed sequence tags (ESTs) in the genome of the turkey (Meleagris gallopavo). Turkey EST sequences were compared with the draft assembly of the chicken whole-genome sequence and the chicken EST database by BLASTN. Among the 877 ESTs examined, 788 had significant matches in the chicken genome sequence. Position of orthologous sequences in the chicken genome and the predicted position of the EST loci in the turkey genome are presented. Genetic assignments suggest a high level of accuracy for the COMPASS predictions. 相似文献
N-Acylethanolamines (NAEs) are fatty-acid derivatives with potent biological activities in a wide range of eukaryotic organisms. Polyunsaturated NAEs are among the most abundant NAE types in seeds of Arabidopsis thaliana, and they can be metabolized by either fatty acid amide hydrolase (FAAH) or by lipoxygenase (LOX) to low levels during seedling establishment. Here, we identify and quantify endogenous oxylipin metabolites of N-linolenoylethanolamine (NAE 18:3) in Arabidopsis seedlings and show that their levels were higher in faah knockout seedlings. Quantification of oxylipin metabolites in lox mutants demonstrated altered partitioning of NAE 18:3 into 9- or 13-LOX pathways, and this was especially exaggerated when exogenous NAE was added to seedlings. When maintained at micromolar concentrations, NAE 18:3 specifically induced cotyledon bleaching of light-grown seedlings within a restricted stage of development. Comprehensive oxylipin profiling together with genetic and pharmacological interference with LOX activity suggested that both 9-hydroxy and 13-hydroxy linolenoylethanolamides, but not corresponding free fatty-acid metabolites, contributed to the reversible disruption of thylakoid membranes in chloroplasts of seedling cotyledons. We suggest that NAE oxylipins of linolenic acid represent a newly identified, endogenous set of bioactive compounds that may act in opposition to progression of normal seedling development and must be depleted for successful establishment. 相似文献
