We prepared antiserum that reacted with a major core polypeptide of approximately 62,000 daltons (62K polypeptide), designated 4b, and its 74K precursor, designated P4b. A cell-free translation product of vaccinia virus late mRNA that comigrated with P4b was specifically immunoprecipitated. The late mRNA encoding P4b hybridized to restriction fragments derived from the left end of the HindIII A fragment and to a lesser extent from the right side of the HindIII D fragment. A polypeptide that comigrated with P4a, the precursor of another major core polypeptide, was synthesized by mRNA that hybridized to DNA segments upstream of the P4b gene. Complete nucleotide sequence analysis of the P4b gene revealed an open reading frame, entirely within the HindIII A fragment, that was sufficient to encode a 644-amino-acid polypeptide of 73K. The 5' end of the P4b mRNA was located at or just above the translational initiation site.  相似文献

Summary Field observations of the adult European skipper, Thymelicus lineola (Ochs), feeding on concentrated nectars (40–65% sucrose) from a variety of flower species led us to question recent literature stating that butterflies feed primarily, and most effectively, on dilute nectars. Rate of sucrose solution intake, volume consumed and feeding duration were measured for males and females at 25 and 35°C under laboratory conditions. As sucrose concentration increased, the volume of solution ingested per meal first increased and then decreased gradually, while sucrose intake was highest at concentrations 40%. Females fed more than males at all concentrations >10% while temperature had no significant effect on meal size. Feeding duration increased with concentration, was shorter at 35 than at 25°C, and was longer for females than males.The rate of volume intake decreased as concentration incresed, but not nearly as rapidly as predicted by earlier models. Rates did not differ between the sexes but were faster at 35 than 25°C. This increase was contributed to equally by a reduction in viscosity and an increase in power output of the cibarial pump. The form of the relations was similar, with maximum rate of sucrose intake occurring at 40% sucrose.A new mathematical model was developed to describe the rate — concentration relation based on the Hagen-Poiseuille equation for laminar fluid flow through pipes. Our model differs from previous models principally in that the power output of the insect's cibarial pump remains relatively constant while the pressure drop created by the pump to induce suction is highly variable. This change results in a very different feeding rate — sucrose concentration function with the optimal rate of sucrose intake at a concentration of approximately 40%. The model indicates that the same relation should hold for a wide range of proboscis shape and size and type of suction pump, and should therefore be applicable to all other nectar feeders with sucking mouth parts. Independent verifications of the model were carried out by measuring the rate of uptake of sucrose solutions of the adult common armyworm, Pseudaletia unipuncta (Haw.), and of human subjects using a volumetric pipette, both of which gave an excellent fit.Nectar concentrations which correspond to optimal rates of sucrose intake should be highly preferred by insects with high feeding costs, those which are time-limited, or which are very vulnerable while feeding. High transport costs and severe water stress may shift preferences to higher and lower concentrations respectively.  相似文献

The effects of ethylene and auxin on the morphology and anatomy of root organ cultures of Pinus mugo Turra var. mugo were investigated to test the hypothesis that changes in root morphology associated with formation of ectomycorrhizae may be related to ethylene produced by ectomycorrhizal fungi or by host plant roots in response to fungus-produced auxin. Morphological changes characteristic of mycorrhizal infection include dichotomous branching of lateral roots, inhibition of root hair formation and enlargement of cortical cells. Lateral roots on non-mycorrhizal root organ cultures, grown in a defined medium, underwent dichtotomous branching while root hair formation was inhibited in response to the ethylene released by 50 and 100 μ M ethephon (2-chloroethylphosphonic acid), but no effect on cortical cell dimensions was observed. The auxin, naphthaleneacetic acid (1 and 10 μ M ) also stimulated dichotomous branching and inhibited root hair formation, but to a lesser extent and with a greater lag time than ethephon. Auxin-stimulated ethylene production by root organ cultures was demonstrated. This appeared to be responsible, at least in part, for the auxin-induced dichotomous branching since the ethylene action inhibitor, silver thiosulfate (0.1 m M ) inhibited the response to auxin by 35%.  相似文献

Nutrients, chlorophyll, phaeophytin and algal abundances were investigated in the surface microlayer and at subsurface depths in a small eutrophic bog pond. Nutrient levels were consistently higher in the microlayer while algal abundance was sometimes higher but sometimes lower in the microlayer than at near subsurface depths. Algal diversity values were strongly influenced by the depth of flagellate blooms, and in contrast to previous studies, diversity in the microlayer was higher than at near subsurface depths. These results are discussed in terms of weather parameters, affinity of algal species for the surface and differences between microlayer ecology in shallow and deep water systems.  相似文献

The effects of iontophoretically applied human pancreatic growth hormone-releasing factor (hpGRF), peptide histidine isoleucine (PHI-27), and somatostatin (SS) on the extracellular activity of single cells in the hypothalamus, thalamus, and cortex of the rat brain were studied in urethane-anesthetized, male rats. Neurons with membrane sensitivity to hpGRF, PHI-27, and SS were present in each brain region. Although neurons excited by these peptides were encountered in thalamus and hypothalamus, depression of neuronal firing was the predominant response observed. Overall, the neurons responding to hpGRF also possessed membrane sensitivity to PHI-27, whereas, the hpGRF sensitive neurons appeared to be more divided as to their ability to respond to SS. The results clearly demonstrate that hpGRF and PHI-27 are capable of affecting the membrane excitability of neurons in several brain regions. The distribution of neurons sensitive to hpGRF suggests that hypothalamic GRF, in addition to its well documented role in the regulation of pituitary growth hormone secretion, may subserve other physiological events in the rat central nervous system as a neurotransmitter and/or neuromodulator.  相似文献

Samples from sixty-two geolittoral sites along two rivers in northern Sweden were analysed in order to quantify possible differences in the vascular plant flora related to differences in the type of natural disturbance of the habitat (more disturbed: rapids (water flow) and lakes (wave action); less disturbed: slow-flowing stretches). We found that the mean species richness was lower along the slow-flowing stretches, than along the rapids and lakes, which were quite similar. When the geolittoral was subdivided into the three vertical belts, the floras of both the upper and middle belts exhibited this same pattern, whereas the species richness of the lower geolittoral belt was also low along the stretches of rapids. Along both rivers, the steepest gradient in mean species richness was recorded on the shores bordering rapids. The highest numbers of 'constant' species (frequency (F) > 60%); and of those which showed a statistically significant (probability (p) > 0.05) preference for a certain type of stretch in at least one of the three geolittoral belts, or in the entire geolittoral, were found along the rapids, in comparison to the flora of the slow-flowing stretches. The flora of the lake shores also comprised a large number of specific species compared to that along the slow-flowing stretches. The predominant species along the above-mentioned stretches reflect both phytogeographical and habitat differences.  相似文献

Probabilistic models of the cell cycle maintain that cell generation time is a random variable given by some distribution function, and that the probability of cell division per unit time is a function only of cell age (and not, for instance, of cell size). Given the probability density, f(t), for time spent in the random compartment of the cell cycle, we derive a recursion relation for n(x), the probability density for cell size at birth in a sample of cells in generation n. For the case of exponential growth of cells, the recursion relation has no steady-state solution. For the case of linear cell growth, we show that there exists a unique, globally asymptotically stable, steady-state birth size distribution, *(x).
