Synthetic iron oxides (goethite, -FeO·OH; hematite, Fe2O3; and ferrihydrite, Fe(OH)3) were used as model compounds to simulate the mineralogy of surface films on carbon steel. Dissolution of these oxides exposed to pure cultures of the metal-reducing bacterium, Shewanella putrefaciens, was followed by direct atomic absorption spectroscopy measurement of ferrous iron coupled with microscopic analyses using confocal laser scanning and environmental scanning electron microscopies. During an 8-day exposure the organism colonized mineral surfaces and reduced solid ferric oxides to soluble ferrous ions. Elemental composition, as monitored by energy dispersive x-ray spectroscopy, indicated mineral replacement reactions with both ferrihydrite and goethite as iron reduction occurred. When carbon steel electrodes were exposed to S. putrefaciens, microbiologically influenced corrosion was demonstrated electrochemically and microscopically.  相似文献

 The role of disturbed photoperiod on the developing surface layers of ripening Scots pine (Pinus sylvestris L.) seeds was studied from August 14 to October 30. The embryo sizes of both the light-treated and the control seeds were examined by the X-ray method prior to germination tests. The anatomical details of resin-embedded seeds were examined by fluorescence and light microscopy. The timing of the ripening process of the surface structures was described and documented. The greatest anatomical changes in the ripening seeds occurred in the sarcotesta and in the nucellar layers. Maturation of the surface structures was essentially slower than could have been interpreted by the size of the embryo and responded clearly to the disturbance of photoperiod. Accumulation of phenolic substances and degeneration of cells, particularly at the chalazal region, advanced faster in the light-treated than in the control seeds up to mid-September. The ripening effect of the altered photoperiod diminished, however, after mid-September. This was also confirmed by the brown colour of the seed coat getting darker only in the control seeds at the end of the test period. Consequently, fully ripe structures were first found about a fortnight earlier in the control than in the light-treated seeds. The coincidental advancement of the anatomical potential examined by the X-ray method supported the role of the surface structures on the anatomical maturity of pine seeds and the timing of cone collecting. Mitochondrial β-oxidation of long-chain fatty acids (LCFA) is essential for mammalian life. Because portions of this metabolic pathway are composed of enzymes that are coordinately regulated and share structural and functional similarities, we evaluated five of these enzyme genes for possible chromosomal linkages. Regulation of LCFA catabolism influences cell signal pathways and apoptosis, as well as energy production from LCFA. Partial cDNA fragments of the mouse mitochondrial proteins carnitine acetyltransferase (Crat), very-long-chain acyl coenzyme A dehydrogenase (Acadvl), the liver and muscle isoforms of carnitine acyltransferase I (Cpt1a and Cpt1b respectively), and a genomic PCR product of mitochondrial protein carnitine acyltransferase II (Cpt2) were used in a previously established mapping panel to determine their chromosomal locations. No pseudogenes were detected for any of the genes in Mus musculus, and all of the genes mapped to different chromosome locations, including the tissue-specific isoforms of carnitine palmitoyltransferase. Crat mapped to Chromosome (Chr) 2, at a position approximately 18 cM from the centromere and 2 cM proximal to the gene Ass1. Acadvl mapped to the middle of Chr 11, 8.3 cM distal to Il4 and 2.8 cM proximal to Mpmv2. Cpt1a mapped to the centromeric region of Chr 19, 8.7 cM proximal to Pomc-ps1. Cpt1b mapped to Chr 15, 4.9 distal to Gpt1 and 3.5 cM proximal to Wnt1. Cpt2 mapped to Chr 4 near the locus Pmv19. Received: 29 January 1998 / Accepted: 25 March 1998  相似文献

The Escherichia coli ATP-binding cassette (ABC) proteins   总被引:8,自引:1,他引:7  
The recent completion of the Escherichia coli genome sequence ( Blattner et al ., 1997 ) has permitted an analysis of the complement of genomically encoded ATP-binding cassette (ABC) proteins. A total of 79 ABC proteins makes this the largest paralogous family of proteins in E . coli . These 79 proteins include 97 ABC domains (as some proteins include more than one ABC domain) and are components of 69 independent functional systems (as many systems involve more than one ABC domain). The ABC domains are often, but not exclusively, the energy-generating domains of multicomponent membrane-bound transporters. Thus, 57 of the 69 systems are ABC transporters, of which 44 are periplasmic-binding protein-dependent uptake systems and 13 are presumed exporters. The genes encoding these ABC transporters occupy almost 5% of the genome. Of the 12 systems that are not obviously transport related, the function of only one, the excision repair protein UvrA, is known. A phylogenetic analysis suggests that the majority of ABC proteins can be assigned to 10 subfamilies. Together with statistical and, importantly, biological evidence, this analysis provides insight into the evolution and function of the ABC proteins.  相似文献

The structure of tubulin has recently been determined by electron crystallography, paving the way for a clearer understandin of the unique properties of tubulin that allow its varied functions within the cell. We investigated patch assessment by northern bobwhites (Collinusvirginianus) in an experimental arena where the distributionof resources in patches, preharvest information about thesepatches, and spacing of patches varied. We found that preharvestinformation about patch quality and a bimodal distribution ofpatch rewards allowed birds to selectively exploit patches highin resources. In contrast, uniform distribution of patch qualitiesand lack of preharvest information caused birds to forage nonselectivelyamong patches. Birds distinguished among patches of differentquality when these patches were spaced 13 m apart, but failedto react to patch quality differences when patches were 0 or3 m apart We also found a strong effect of the level of patchdepletion on foraging decisions: as resources in die arena becamescarce, birds increasingly foraged selectively in die most profitablepatches. Foraging decisions of bobwhites are biased by die waythey experience and memorize a spatially and temporally variableenvironment. The relative cost of this cognitive bias (i.e.,lost opportunity) is nonlinearty related to die mean resourcedensity in die environment and to die difference between thismean density and die resource density in die exploited patch.Cognitive bias should be considered when evaluating patch assessmentcapabilities of foragers in complex environments.  相似文献

Data from epidemiological studies might be seen as superior to data from animal bioassays for risk assessment purposes. Because humans are the population of interest, use of epidemiological data avoids interspecies extrapolation. However, one must not assume that an epidemiological study is necessarily valid at face value. We describe issues of validity that arise in the conduct and interpretation of epidemiological research and that affect the utility of epidemiological data in risk assessment. These issues include choice of study design, size and representativeness of the study sample, measurement of exposures and outcomes, control of confounding and specification of statistical model for analysis of data, all of which affect the accuracy and validity of study results.  相似文献

A new warming technique has been developed in a field experimental study of the potential effects of climatic change on N leaching from hill land plant/soil systems. Thermocouple compensating cable has been utilized to provide a small cross-section, flexible, low voltage heating cable, mounted on a framework of stainless steel mesh, to provide uniform heating at the vegetation/soil interface of zero-tension lysimeters and surrounding turf. We describe a specially designed heat controller capable of maintaining a temperature differential of 3 °C above ambient at a soil depth of 0.8 cm. The equipment raises temperatures down the soil profile and within the grass sward, whilst tracking normal diurnal temperature variation. Results presented here illustrate the efficacy of the warming technique, together with the consequences for the release of nitrate from lysimeters. The responses of soil solution concentrations of nitrate varied markedly between soil types, but showed a significant decrease in the brown earth during the first 5 months of additional heating. This suggests that increased nutrient release is masked by plant uptake in this soil, but the responses in the other two soils were less marked.  相似文献

GS4071 is a potent inhibitor of influenza neuraminidase. A precolumn fluorescence derivatization HPLC method is described for the analysis of GS4071 in rat plasma. Plasma samples were subjected to solid-phase extraction on C18 extraction columns. After extraction, GS4071 was derivatized with naphthalenedialdehyde in the presence of potassium cyanide to produce highly fluorescent cyano[f]benzoisoindole derivatives. Derivatized samples were stable for >24 h at 4°C. The samples were analyzed by an isocratic HPLC method using fluorescence detection at 420 nm excitation and 470 nm emission wavelength. The method was validated and applied to the analysis of plasma samples from pre-clinical pharmacokinetic studies in rats. The limit of detection for GS4071 was 20 ng/ml. For five replicate samples at 50, 400, and 1000 ng/ml, the within-day precision values were 16.9, 9.4 and 4.5%, respectively, and the between-day precision values were 16.9, 7.9, and 2.1%, respectively. The method was linear from 25 to 1600 ng/ml and the total recovery was >68% over this concentration range.  相似文献

One subclass of B12-requiring enzymes is now known to bind their B12 coenzymes “base-off,” with a histidine residue from the protein supplying an imidazole ligand to the cobalt center. Recent results from Sirovatka and Finke (J.M. Sirovatka and R.G. Finke, J.Am. Chem. Soc. 119, (1997) 3057) show that imidazole has an extraordinary trans effect on the mode of carbon–cobalt bond cleavage in coenzyme B12 analogs, compared to pyridine or the natural 5,6-dimethylbenzimidazole ligand, and it was suggested that a differential steric effect could, in part, account for the uniqueness of the imidazole ligand. Such a differential steric effect for imidazole and pyridine is now demonstrated by studies of the thermodynamics of ligation of these ligands to the α and β diastereomers of two alkylcobinamides (RCbi+s, derivatives of cobalamins which lack the normal axial nucleotide) based on the known differences in steric crowding of the α (“lower”) and β (“upper”) axial ligand positions of cobalt corrinoids. Imidazole binds more tightly than pyridine to both diastereomers of NCCH2Cbi+ and CF3Cbi+, in all cases due to a more favorable entropy change, which is the result of lowered steric interference with corrin side chain thermal motions.  相似文献
