Effect of stimulus strength and adaptation on the thermotactic response of Dictyostelium discoideum pseudoplasmodia

The thermotactic responses of Dictyostelium discoideum strain HL50 and mutants derived from this strain have been characterized by curves of stimulus-strength vs response. With gradient midpoint temperatures of 16 and 24 °C, these curves are typical of those of a single response, i.e., the strength of the response increases with increasing stimulus strength until at some strength the response saturates. However, with a gradient midpoint temperature close to the transition from negative to positive thermotaxis, the sign of the thermotactic response depends on gradient strength. These observations support the hypothesis that the transduction pathways for positive and negative thermotaxis act concurrently and contain separable elements. An investigation of the adaptation of thermotaxis indicated that the stimulus-strength-dependence and midpoint-temperature-dependence of both thermosensory responses was altered by shifting the growth and development temperature.     

Nodulation studies on legumes exotic to Australia: Hedysarum coronarium

Abstract Symbiotic experiments in glasshouse, controlled environment cabinet, and field were conducted with four lines of sulla ( Hedysarum coronarium ) and 15 strains of Rhizobium spp. This plant is highly Rhizobium -specific and appropriate strains are most unlikely to occur naturally in Australia. Under several sets of experimental conditions, H. coronarium nodulated abundantly and effectively with homologous rhizobia introduced from Spain and Italy. The optimum temperature for nitrogen fixation was relatively low (approx. 21°C) but significant interactions between line of host, strain of rhizobia, and growth temperature were frequent. The rhizobia were persistent in soil.     

Effects of estradiol-17β in the male xenopus laevis: Isolation and translation of cytoplasmic messenger rna populations

Summary Total Xenopus liver cytoplasmic RNA isolated following long-term estrogen administration (14 days) was fractioned using Sepharose 4B chromatography. One of the Sepharose 4B peaks was shown to contain RNA with a molecular weight reported for vitellogenin mRNA (34S). The presence of estrogeninduced vitellogenin mRNA in the peak 5 RNA was determined by translation of the RNA in the oocyte and analysis of the oocyte translational products by immunoprecipitation with anti-vitellogenin.Sepharose 4B peaks 2 and 3 were also observed to contain estrogen induced mRNA populations sedimenting between 9-18S. These findings suggest that Sepharose 4B chromatography might prove useful in separating different mRNA populations following estrogen-induced gene activation.     

Separation of double-label liquid scintillation samples by half-life

A method is presented for separation of double- or triple-labeled liquid scintillation samples based on the partial decay of one of the nuclides. Validity of the procedure has been demonstrated with the following combinations of nuclides commonly encountered in laboratory situations; ¹³¹I-¹⁴C, ³²P-¹⁴C, and ²⁰³Hg-¹⁴C, Separation of all nuclide pairs agrees favorably with predieted values except when the activity ratios exceed 10³. The determinations are independent of quenching and nuclide spectra and greatly augment the number of β-β and β-γ combinations resolved by liquid scintillation spectrometry. The utility of this method for separation of triple-labeled samples is discussed.     

The flight behaviour of individual passerine nocturnal migrants: A tracking radar study

Nocturnal passerine migrants were tracked with a small automatic tracking radar during spring migration in eastern New York. Climbing, descending and markedly non-linear tracks were selected for analysis because they may reveal relationships not evident in normal straight and level tracks. Climbing individuals ascended at 1 to 2 vertical metres per second by heading into the wind and increasing their ascent angles while air speed tended to remain constant. Within individual tracks, birds flew slower when flying downwind than when flying into the wind and changes in air speed were performed over periods of a few seconds. A small amount of data suggested that this behaviour did not occur under overcast skies. Both the direction and speed of the wind force were important in predicting air speed. Multiple regression analysis indicated that faster flying birds were more likely to fly in winds of high speed and at large angles into the wind.     

Light-harvesting pigment-protein complex deficiency in Hosta (Liliaceae)

A yellow-leaved plastome mutant of Hosta (Hosta sieboldii Ingram complex, Liliaceae) known as Wogan Gold lacks normal granal stacks, but has numerous stroma lamellae extending throughout the chloroplast. The chlorophyll a/b ratio is 0.76 in the mutant and 2.9 in wild type. The mutant contains a qualitatively normal pattern of other photosynthetic co-pigments. SDS-polyacrylamide gel electrophoresis showed a deficiency in the photosystem (PS) II light-harvesting complex. Since PS II is localized mainly in the granal region, the absence of the light-harvesting complex may explain the loss of granal stacking in this mutant.Abbreviation PS photosystem     

Rat cell surface antigens: Nonidentity of the H-4 and Ag-F traits

Animals partially congenic to Lewis have been bred that differ at the locus, linked to C., which controls the Ag-F antigenic trait. Skin grafts from these animals, placed on Lewis recipients, were not rejected. This result demonstrates that Ag-F does not mediate skin incompatibility and, thus, is distinct from H-4, which was defined based on a demonstrable incompatibility and which is also linked to C.
