Mutation of the TP53 tumor suppressor gene is the most common genetic alteration in cancer, and almost 1000 alleles have been identified in human tumors. While virtually all TP53 mutations are thought to compromise wild type p53 activity, the prevalence and recurrence of missense TP53 alleles has motivated countless research studies aimed at understanding the function of the resulting mutant p53 protein. The data from these studies support three distinct, but perhaps not necessarily mutually exclusive, mechanisms for how different p53 mutants impact cancer: first, they lose the ability to execute wild type p53 functions to varying degrees; second, they act as a dominant negative (DN) inhibitor of wild type p53 tumor-suppressive programs; and third, they may gain oncogenic functions that go beyond mere p53 inactivation. Of these possibilities, the gain of function (GOF) hypothesis is the most controversial, in part due to the dizzying array of biological functions that have been attributed to different mutant p53 proteins. Herein we discuss the current state of understanding of TP53 allele variation in cancer and recent reports that both support and challenge the p53 GOF model. In these studies and others, researchers are turning to more systematic approaches to profile TP53 mutations, which may ultimately determine once and for all how different TP53 mutations act as cancer drivers and whether tumors harboring distinct mutations are phenotypically unique. Abstract A second ecological survey of the Waikanae estuary, in the North Island of New Zealand, was carried out 30 years after the first (1941–1943), and was of similar duration. The observer for the first survey participated in the second, and the same methods were used. Brief accounts are given of the prevailing climate and geology, and of physical changes at the estuary. The effect of periodic storms and of the various encroachments by man is recorded; the latter appear to be particularly important. The vegetation of the estuary is described. Records of beach patrols and of other ornithologists supplement the authors' observations on the bird life. The numbers of birds recorded in the two surveys are compared by analyses of variance. Changes in the flora deduced from aerial photographs spanning the two surveys and from notes and photographs are related to man-induced disturbance, natural physical changes, and natural succession. The total of 79 bird species recorded at the estuary and in its immediate vicinity compares favourably with the 60 species recorded during the first survey. The increase of nearly one-third is due to such factors as the appearance of new Australian bird species, the information provided by the beach patrols, and the extension of sand-flat habitat. In terms of numbers observed, 10 bird species have increased, 11 remain unchanged, and 8 have declined. The distribution of birds is considered in relation to nine areas of habitat: the beach and open sea; open dunes at the beach front; river and sand flats; unfixed and fixed dunes; saltmarsh; grassed fields; and lakelets. The most species (29) were recorded on sand flats and the least (7) on unfixed dunes. Periodic rechannelling of the river mouth has led to expansion of the sand flats as a habitat for waders and some other shore birds, but other man-induced developments, such as housing, are threatening the present ecological balance of the estuary. Use of the area for recreation and for wildlife study is increasing, and we consider that action by conservation authorities is urgently required. 相似文献

A novel regulatory metal binding domain is present in the C terminus of Arabidopsis Zn2+-ATPase HMA2

HMA2 is a Zn2+-ATPase from Arabidopsis thaliana. It contributes to the maintenance of metal homeostasis in cells by driving Zn2+ efflux. Distinct from P1B-type ATPases, plant Zn2+-ATPases have long C-terminal sequences rich in Cys and His. Removal of the 244 amino acid C terminus of HMA2 leads to a 43% reduction in enzyme turnover without significant effect on the Zn2+ K(1/2) for enzyme activation. Characterization of the isolated HMA2 C terminus showed that this fragment binds three Zn2+ with high affinity (Kd = 16 +/- 3 nM). Circular dichroism spectral analysis indicated the presence of 8% alpha-helix, 45% beta-sheet, and 48% random coil in the C-terminal peptide with noticeable structural changes upon metal binding (8% alpha-helix, 39% beta-sheet, and 52% random coil). Zn K-edge XAS of Zn-C-MBD in the presence of one equivalent of Zn2+ shows that the average zinc complex formed is composed of three His and one Cys residues. Upon the addition of two extra Zn2+ ions per C-MBD, these appear coordinated primarily by His residues thus, suggesting that the three Zn2+ binding domains might not be identical. Modification of His residues with diethyl pyrocarbonate completely inhibited Zn2+ binding to the C terminus, pointing out the importance of His residues in Zn2+ coordination. In contrast, alkylation of Cys with iodoacetic acid did not prevent Zn2+ binding to the HMA2 C terminus. Zn K-edge XAS of the Cys-alkylated protein was consistent with (N/O)4 coordination of the zinc site, with three of those ligands fitting for His residues. In summary, plant Zn2+-ATPases contain novel metal binding domains in their cytoplasmic C terminus. Structurally distinct from the well characterized N-terminal metal binding domains present in most P1B-type ATPases, they also appear to regulate enzyme turnover rate. 相似文献

The results of elective truncal vagotomy and drainage in 547 duodenal ulcer patients are reported. Altogether, 204 patients were randomly allocated to pyloroplasty and 200 to gastrojejunostomy. In 101 patients gastrojejunostomy was electively chosen and in 42 patients the duodenum was opened to confirm the diagnosis. Operative mortality was 0·5%, the incidence of proved recurrent ulceration 3·3%, severe dumping 2%, and severe diarrhoea 1·1%.
