A late step in cytokinesis requires the central spindle, which forms during anaphase by the bundling of antiparallel nonkinetochore microtubules. Microtubule bundling and completion of cytokinesis require ZEN-4/CeMKLP-1, a kinesin-like protein, and CYK-4, which contains a RhoGAP domain. We show that CYK-4 and ZEN-4 exist in a complex in vivo that can be reconstituted in vitro. The N terminus of CYK-4 binds the central region of ZEN-4, including the neck linker. Genetic suppression data prove the functional significance of this interaction. An analogous complex, containing equimolar amounts of a CYK-4 ortholog and MKLP-1, was purified from mammalian cells. Biochemical studies indicate that this complex, named centralspindlin, is a heterotetramer. Centralspindlin, but not its individual components, strongly promotes microtubule bundling in vitro.  相似文献

Coaggregation assays were performed to investigate interactions between oral Bifidobacterium adolescentis and other oral bacterial species. Bifidobacterium adolescentis OLB6410 isolated from the saliva of healthy humans did not coaggregate with Actinomyces naeslundii JCM8350, Streptococcus mitis OLS3293, Streptococcus sanguinis JCM5708, Veillonella parvula ATCC17745 or Porphyromonas gingivalis OB7124, but it did coaggregate with Fusobacterium nucleatum JCM8532. Subsequent examination of biofilm formation on saliva-coated hydroxyapatite discs using FISH revealed that B. adolescentis OLB6410 could not directly adhere to the coated discs. It did, however, adhere to biofilms of A. naeslundii, V. parvula, and F. nucleatum, although it did not coaggregate with A. naeslundii nor with V. parvula. These results suggest that the adhesion of B. adolescentis to tooth surfaces is mediated by other oral bacteria. Heat- or proteinase K-treated F. nucleatum could not coaggregate with B. adolescentis. Similarly, the coaggregation and coadhesion of proteinase K-treated B. adolescentis were strongly inhibited. It is therefore probable that proteinaceous factors on the cellular surface of B. adolescentis and F. nucleatum are involved in their interaction. The data presented in this study add to our understanding of bifidobacterial colonization in the human oral cavity.  相似文献

We isolated oral bacteria that coexisted with Porphyromonas gingivalis in a hamster periodontitis model. As predominant bacteria in the periodontitis site, Collinsella-reltaed strains, Eubacterium-reltaed strains, Streptococcus suis-related strains, and Veillonella parvula-reltaed strains were detected. In addition, Actinomyces, Bacteroides, and P. gingivalis were also isolated predominantly. The results suggest that the bacterial composition of the periodontitis site in hamsters is complex, as in human periodontitis.  相似文献

Periodontitis is one of the most common oral diseases in humans. This caused by infection by the oral bacterium Porphyromonas gingivalis. Our strategy to prevent this infection is to establish a passive immunization system in which endogenous antibodies can be applied directly to neutralize virulent factors associated with this bacterium. We focused our attention on the P. gingivalis 35 kDa surface protein, or HBP35, since this protein is involved not only in the coaggregation with oral miroflora but also in hemin binding. In addition, nucleotide sequencing of the gene, hbp35, coding for this protein revealed the presence of a catalytic center for thioredoxin, and we further attempted to characterized the protein by amino acid substitution. A total of four Cys residues were substituted for Ser residues by combining the simple method for site-directed mutagenesis and the heterodimer system, an approach designed to construct chimeric plasmids readily. Native and mutagenized hbp35 were introduced into the Eschericha coli dsbA mutant strain, JCB 572, defective in both alkaline phosphatase and motile activities due to inefficient disulfide bond formation. Transformant harboring the native hbp35 could complement the dsbA mutation, suggesting a role of disulfide bond formation of this protein in P. gingivalis cells. Possible roles of the Cys residues in complementation are discussed.  相似文献

Abundance of drifting seaweeds in eastern East China Sea   总被引:3,自引:0,他引:3  
Drifting seaweeds play important ecological roles in offshore waters. Recently, large amounts of drifting seaweed rafts were found in the eastern East China Sea between the continental shelf and the oceanic front of the Kuroshio Current. However, so far there have been no quantitative reports about this particular area. Two research cruises were organized to survey abundance and standing crop of drifting seaweeds in eastern East China Sea in May 2002 and March 2004, using visual census and net sampling of drifting seaweeds. Visual census data were composed of drifting seaweed raft diameter, perpendicular distance from the transect (navigation course of the research vessel) to the raft, and positions. Using these data, we calculated the “effective stripe width” using the DISTANCE software. Drifting seaweed abundance (composed exclusively of Sargassum horneri) in waters located between the continental shelf peripheral area and the Kuroshio oceanic front was estimated to be higher than in any other area within eastern East China Sea in March and May. Abundance means in May 2002 and March 2004 were 6.14 and 29.05 rafts km−2, respectively, while standing crop reached 126.81 and 20.35 kg km−2 (wet weight). Mean diameter and drifting seaweed rafts in May 2002 were significantly greater than in March 2004, reflecting seasonal growth of Sargassum horneri.  相似文献

We report the multi-functionalized cubic silsesquioxane (POSS) as the ratiometric multimodal chemosensors for monitoring solvent polarity with fluorescence and (19)F NMR. The alteration of the dispersion state of the modified POSS by changing solvent polarity can be reflected into the orthogonal signal responses for the fluorescence and (19)F NMR. In addition, the ratiometric dual monitoring for the enzymatic reaction was performed using the POSS-based chemosensor.  相似文献

During our research using a high-throughput screening system for discovery of a new class of human liver glycogen phosphorylase a (hLGPa) inhibitors, a series of 3-(3,4-dichlorophenyl)acrylamide derivatives were synthesized, and their inhibitory activities toward hLGPa were evaluated. Among the derivatives, (2E,2'E)-N,N'-pentane-1,5-diylbis[3-(3,4-dichlorophenyl)acrylamide] (6c) inhibited hLGPa with an IC(50) value of 0.023 microM. An X-ray crystallographic study of the enzyme-6c complex showed that the inhibitor is bound at the dimer interface site, where the 3,4-dichlorophenyl moiety interacts hydrophobically with the enzyme.  相似文献

With the original aim of surveying the role of exopolysaccharide (EPS) in Lotus-Mesorhizobium symbiosis, we carried out Tn5 mutagenesis of Mesorhizobium loti and obtained 32 mutants with defects in EPS biosynthesis. One of the mutants, HIA22, formed pseudonodules and failed to fix nitrogen with Lotus japonicus. However, complementation analysis unexpectedly revealed that the potential gene with the locus tag, mll2073, interrupted by Tn5 was responsible for neither normal EPS synthesis nor symbiosis. Further analysis uncovered that HIA22 had a genome deletion of approximately 20 kbp, resulting in the loss of two separate genes responsible for EPS biosynthesis and symbiosis. One gene with the locus tag, mll5669, was needed to synthesize normal EPS that fluoresced on medium containing Calcofluor and encoded a homolog of O-antigen acetyl transferase in Salmonella typhimurium. A specific mutant of mll5669, EMB-B58, successfully fixed nitrogen when infected onto L. japonicus. Another gene, mlr5647, was needed to establish fully functional nodules and encoded ornithine carbamoyl transferase [ArgF (EC 2.1.3.3)], which participates in arginine biosynthesis. A specific mutant of mlr5647, EMB-Y2, showed arginine auxotrophy and formed infection threads, but the nodules formed by this strain had few infected cells filled with bacteroids. These mutant phenotypes were complemented by supplementation of arginine or citrulline to bacterial or plant medium. EMB-Y2 represented a novel class of rhizobial arginine auxotrophs with symbiotic deficiency, and its phenotypes indicated that sufficient supply of citrulline or its derivative is essential for successful infection or for a stage in the infection process in Lotus-Mesorhizobium symbiosis.  相似文献
