In the growing culm of C. alternifolius, surgical removal of parts indicated that the stimulus for the prolonged activity of the internodal intercalary meristem (IM) came from the matured leaves and upper internode and that buds were not involved in maintaining internodal growth. Decapitation of the culm resulted in cessation of internodal extension. Various growth regulators were applied to the decapitated internode, and both the total extension and growth rates were analyzed statistically. Gibberellin A3 (GA) and benzyladenine (BA) substituted for the excised parts in their effect on internodal extension. Indoleacetic acid (IAA) had little effect. (2-chloroethyl) trimethylammonium chloride (CCC) inhibited internodal growth, and its effects were reversed by GA. IAA was antagonistic to BA but not to GA. BA and GA were somewhat antagonistic. The quantitative effects of growth regulators on epidermal and ground parenchyma cell length and number of interstomatal cells were examined. Extension induced by GA was due to both cell division and cell elongation in the IM. Cells were longer, and fewer stomates differentiated than in the control. In internodes induced to extend by GA + BA cell division, cell length, and stomate differentiation were similar to the control. The results indicate that prolonged internodal IM activity is maintained by cytokinins and gibberellins coming from the matured upper portions of the culm. Changes in the levels of these regulators during growth presumably result in the histological gradient in the internode.  相似文献

Human embryonic diploid lung fibroblasts grown in Eagle's medium were exposed continually to a variety of environmental conditions over a large number of passages to observe how these conditions affected the growth and longevity of these cells in vitro. The cells grew well at temperatures between 34 and 37 C and some cells could be adapted to grow at 40 C. Very limited growth occurred at 30 to 31 C; however, confluent monolayers of cells could be maintained for months at 30 C and still give rise to actively growing cultures. Increasing the amino acid concentration in Eagle's medium or the calf serum concentration above 10% had no effect on the growth rate or longevity. One per cent calf serum could not support prolonged active growth. Trypsin concentrations between 1 and 0.1% and crystalline trypsin at 50 μg/ml showed no influence on cell growth. Ethylenediaminetetraacetic acid treatment and scraping, however, destroyed many of the cells, and the survivors grew poorly. The clonal morphology varied with age. Young cells frequently gave rise to densely packed clones, whereas older cells gave rise to clones with widely scattered cells. The cloning efficiency was high when the cells were young but decreased rapidly with successive passage. It was relatively constant from the 7th to 20th passage at about 15%.  相似文献

Isolated cortical collecting tubules from rabbit kidney were studied during perfusion with solutions made either isotonic or hypotonic to the external bathing medium. Examination of living tubules revealed a reversible increase in thickness of the cellular layer, prominence of lateral cell membranes, and formation of intracellular vacuoles during periods of vasopressin-induced osmotic water transport. Examination in the electron microscope revealed that vasopressin induced no changes in cell structure in collecting tubules in the absence of an osmotic difference and significant bulk water flow across the tubule wall. In contrast, tubules fixed during vasopressin-induced periods of high osmotic water transport showed prominent dilatation of lateral intercellular spaces, bulging of apical cell membranes into the tubular lumen, and formation of intracellular vacuoles. It is concluded that the ultrastructural changes are secondary to transepithelial bulk water flow and not to a direct effect of vasopressin on the cells, and that vasopressin induces osmotic flow by increasing water permeability of the luminal cell membrane. The lateral intercellular spaces may be part of the pathway for osmotically induced transepithelial bulk water flow.  相似文献

Introgression between two allopatric species,Quercus macrocarpa andQ. gambelii, has been observed in two separate locations: northeastern New Mexico and the Black Hills of western South Dakota and adjacent Wyoming. The probability that this introgression is the result of long-range pollination appears remote. Presumably these two species hybridized during a period of past sympatric association. Further indication of past sympatry in the Black Hills is the presence of a common species of obligate parasite, i.e., wasp(Cynips insulensis) onQ. gambelii in the Rocky Mountains and onQ. macrocarpa of the Black Hills. The hybrid oaks in New Mexico probably reflect a westward migration ofQ. macrocarpa during pluvial periods of the Pleistocene.Quercus gambelii most likely reached the Black Hills during the warmer postglacial hypsithermal era. The hybridization reported here may reflect secondary sympatry, i.e., sympatric occurrence after the species, or their ancestors, became geographically separated.  相似文献

A lettuce floc was prepared and used for recovering enterovirus from an aqueous suspension. The method is simple, and the adsorption of coxsackievirus B5, echovirus 7, and poliovirus 1 is quantitative. The virus-floc complex may be removed from aqueous suspension by low-speed centrifugation and dissolved at an alkaline pH in a small volume of water; virus is then available for assay on cultured cells. Aerobacter aerogenes and Bacillus subtilis accumulated p, p'-DDT and methoxychlor directly from water. Uptake of both (14)C-labeled organochlorine insecticides was rapid; 80 to 90% of the 24-h residues were reached within 30 min. Total cellular residues varied linearly with concentrations of DDT and methoxychlor in water ranging from 0.5 to 5.0 mug/liter. The residue magnification factors from water were between 1,400- to 4,300-fold, but were independent of insecticide concentrations in water. When the insecticide-exposed microbial cells were washed with pesticide-free water, DDT residues were 45% in A. aerogenes and 30% in B. subtilis, whereas the methoxychlor level decreased nearly 75% in both organisms. Subsequent washing did not further reduce the insecticide residue. Autoclave-killed bacteria also rapidly adsorbed DDT and methoxychlor from water and, in some instances, residues were higher than in the living cells. Molecular polarity and lipid solubility appear to influence the retention of the organochlorine insecticides by bacterial cells.  相似文献

A pour-plate method is described for the detection of coagulase production by Staphylococcus aureus. Either Brain Heart Infusion agar or yeast extract-Trypticase soy agar containing swine plasma was the best medium for the detection of coagulase by this method. The advantages of this method and its potential utilization in the clinical laboratory, in food microbiology, and in specialized studies with S. aureus are discussed.  相似文献

Electrokinetic properties of isolated cerebral-cortex synaptic vesicles   总被引:1,自引:0,他引:1  
Synaptic vesicles isolated from guinea-pig cerebral cortex had an electrophoretic mobility of -3.55mum.s(-1).V(-1).cm in saline-sorbitol, pH7.2, at 25 degrees C (ionic strength 0.015g-ions/1). The mobility was pH-dependent, varied with ionic strength and indicated that the vesicular surface contained weak acidic functions with a pK(a) in the range 3.0-3.8. Although the vesicular surface was determined to be highly negatively charged, treatment with neuraminidase had no effect on mobility and indicated that the relatively strong carboxyl groups of sialic acid do not contribute significantly to vesicular electrokinetic properties. Treatment of synaptic vesicles with trypsin or trypsinized concanavalin A resulted in increases in mobility, but treatment with ribonuclease, deoxyribonuclease, chrondroitinase ABC or hyaluronidase had no significant effect on mobility. Mn(2+) or Ca(2+) was more effective in decreasing vesicle mobility than was Mg(2+), Sr(2+) or Ba(2+). The electrokinetic properties of the synaptic vesicle surface are discussed and contrasted with the properties of the synaptosomal membrane.  相似文献
