Saccharomyces boulardii is gaining in popularity as a treatment for a variety of diarrheal diseases as well as inflammatory bowel disease. This study was designed to examine the effect of this yeast on infection by Shigella flexneri, a highly infectious and human host-adapted enteric pathogen. We investigated key interactions between the bacteria and host cells in the presence of the yeast in addition to a number of host responses including proinflammatory events and markers. Although the presence of the yeast during infection did not alter the number of bacteria that was able to attach or invade human colon cancer-derived T-84 cells, it did positively impact the tight junction protein zonula occluden-2 and significantly increase the barrier integrity of model epithelia. The yeast also decreased ERK, JNK, and NF-kappaB activation in response to S. flexneri, events likely responsible for the observed reductions in IL-8 secretion and the transepithelial migration of polymorphonuclear leukocytes across T-84 monolayers. These results, suggesting that the yeast allowed for a dampened inflammatory response, were confirmed in vivo utilizing a highly relevant model of human fetal colonic tissue transplanted into scid mice. Furthermore, a cell-free S. boulardii culture supernatant was also capable of reducing IL-8 secretion by infected T-84 cells. These data suggest that although the use of S. boulardii during infection with S. flexneri may alleviate symptoms associated with the inflammatory response of the host, it would not prevent infection.  相似文献

The objective of this study was to investigate the influence of dynamic compressive loading on chondrogenesis of mesenchymal stem cells (MSCs) in the presence of TGF-β3. Isolated porcine MSCs were suspended in 2% agarose and subjected to intermittent dynamic compression (10% strain) for a period of 42 days in a dynamic compression bioreactor. After 42 days in culture, the free-swelling specimens exhibited more intense alcian blue staining for proteoglycans, while immunohistochemical analysis revealed increased collagen type II immunoreactivity. Glycosaminoglycan (GAG) content increased with time for both free-swelling and dynamically loaded constructs, and by day 42 it was significantly higher in both the core (2.5 ± 0.21%w/w vs. 0.94 ± 0.03%w/w) and annulus (1.09 ± 0.09%w/w vs. 0.59 ± 0.08%w/w) of free-swelling constructs compared to dynamically loaded constructs. This result suggests that further optimization is required in controlling the biomechanical and/or the biochemical environment if such stimuli are to have beneficial effects in generating functional cartilaginous tissue.  相似文献

Human disturbance from tourism and other non-consumptive activities in protected areas may be stressful to wildlife. Animals may move away in space or time to avoid human interaction. For species of particular conservation concern, such as Baird's tapirs (Tapirus bairdii) and jaguars (Panthera onca), a better understanding of how they respond to different levels and types of disturbance is needed in order to manage human visitation to parks in ways that minimize negative outcomes for wildlife. We describe the overlap in activity patterns of tapirs, jaguars, and humans at logged and unlogged sites and at places with low versus high human visitation using camera survey data from protected areas of NW Belize, 2013–2016. Tapirs were nocturnal in all study sites, with > 80% of all tapir detections occurring between 1900 hr and 0500 hr. Their activity patterns were not different in unlogged versus logged sites and did not change with increased human traffic. Jaguars were cathemeral across sites but had more nocturnal activity at the site with the most human impact. Activity pattern overlap between tapirs and jaguars did not differ significantly between logged and unlogged sites, nor between areas with low and high human activity. Human traffic increased from 2013 to 2016 at most of the study locations. In conclusion, this camera trap dataset suggests that non-consumptive human disturbance does not alter the activity patterns of tapirs and jaguars in protected areas lacking hunting pressure.  相似文献

Interspecific competition between individuals of different species can result in reductions in their fecundity, growth or survival, reflecting differential exploitation of resources that become intensified due to spatial co-occurrence, ecological similarity and increased population densities. As two species cannot occupy the same niche, coexistence is only possible if the available resources are used in non-overlapping manners such as niche partitioning or the use of refuges. Among agricultural insect pests, such as fruit flies of the family Tephritidae, competitive interactions can result in competitive displacement, host changes, or the expansion or restriction of the numbers of hosts utilized that can have negative consequences for human agricultural activities. We evaluated the competitive interactions between two fruit fly species of the genus Anastrepha, Anastrepha obliqua (Macquart, 1835) and Anastrepha fraterculus (Wiedmann, 1830), on their respective preferred hosts (mangoes and guava). Experiments of larval competition and competition for ovipositioning sites by adult females were performed to compare the parameters of larval development time, numbers of pupae and emerged adults and numbers of ovipositions in the presence or absence of interspecific competition. We observed that the interactions between those species were asymmetrical and hierarchical, and our results suggest a competitive displacement of A. fraterculus by A. obliqua when those two species are present on the same fruit, whether mangoes or guavas.  相似文献

In 15 anesthetized apneic, oxygenated rabbits we simultaneously measured pleural liquid and interstitial extrapleural parietal pressures by using catheters and/or cannulas and micropipettes connected to a servonull system. With the animal in lateral posture, at an average recording height of 4.4 +/- 0.9 (SD) cm from the most dependent part of the cavity, the extrapleural catheter and the pleural cannula yielded -2.5 +/- 0.6 and -5.5 +/- 0.2 cmH2O; the corresponding values for micropipette readings in the two compartments were -2.4 +/- 0.6 and -5.4 +/- 0.4 cmH2O, respectively (not significantly different from those measured with catheters and cannulas). In the supine animal, interstitial extrapleural catheter pressure data obtained at recording heights ranging from 15 to 80% of pleural cavity lay on the identity line when plotted vs. the micropipette pressure values simultaneously gathered from the same tissues. We conclude that 1) micropipettes and catheters-cannulas yield similar results when recording from the same compartment and 2) the hydraulic pressure in the parietal extrapleural interstitium is less negative than that in the pleural space.  相似文献

The skin of late embryonic, larval, and young postmetamorphic newts, Taricha torosa, has been examined with particular reference to areas of cellular attachment. Stereo electron microscopic techniques and special staining methods for extracellular materials were utilized in addition to conventional avenues of ultrastructural study to investigate the fine architecture of desmosomes, hemidesmosomes, their associated filament systems, and extracellular materials. No evidence has been found that continuity of tonofilaments between adjacent cells exists at desmosomes. Rather, most of the tonofilaments which approach desmosomes (and perhaps also hemidesmosomes) course toward the "attachment plaque" and then loop, either outside the plaque or within it, and return into the main filament tracts of the cell. These facts suggest that the filamentous framework provides intracellular tensile support while adhesion is a product of extracellular materials which accumulate at attachment sites. Evidence is presented that the extracellular material is arranged as pillars or partitions which are continuous with or layered upon the outer unit cell membrane leaflets and adjoined in a discontinuous dense midline of the desmosome. A similar analysis has been made of extracellular materials associated with hemidesmosomes along the basal surface of epidermal cells. An adepidermal globular zone, separating the basal cell boundary from the underlying basal lamina and collagenous lamellae during larval stages, has been interpreted from enzyme and solvent extraction study as a lipid-mucopolysaccharide complex, the function of which remains obscure. These observations are discussed in relation to prevailing theories of cellular adhesion and epidermal differentiation. They appear consistent with the concept that a wide range of adhesive specializations exists in nature, and that the more highly organized of these, such as large desmosomes and hemidesmosomes, serve as strong, highly supported attachment sites, supplemental in function to a more generalized aggregating mechanism.  相似文献

Post‐translational modifications (PTM) of proteins can control complex and dynamic cellular processes via regulating interactions between key proteins. To understand these regulatory mechanisms, it is critical that we can profile the PTM‐dependent protein–protein interactions. However, identifying these interactions can be very difficult using available approaches, as PTMs can be dynamic and often mediate relatively weak protein–protein interactions. We have recently developed CLASPI (cross‐linking‐assisted and stable isotope labeling in cell culture‐based protein identification), a chemical proteomics approach to examine protein–protein interactions mediated by methylation in human cell lysates. Here, we report three extensions of the CLASPI approach. First, we show that CLASPI can be used to analyze methylation‐dependent protein–protein interactions in lysates of fission yeast, a genetically tractable model organism. For these studies, we examined trimethylated histone H3 lysine‐9 (H3K9Me3)‐dependent protein–protein interactions. Second, we demonstrate that CLASPI can be used to examine phosphorylation‐dependent protein–protein interactions. In particular, we profile proteins recognizing phosphorylated histone H3 threonine‐3 (H3T3‐Phos), a mitotic histone “mark” appearing exclusively during cell division. Our approach identified survivin, the only known H3T3‐Phos‐binding protein, as well as other proteins, such as MCAK and KIF2A, that are likely to be involved in weak but selective interactions with this histone phosphorylation “mark”. Finally, we demonstrate that the CLASPI approach can be used to study the interplay between histone H3T3‐Phos and trimethylation on the adjacent residue lysine 4 (H3K4Me3). Together, our findings indicate the CLASPI approach can be broadly applied to profile protein–protein interactions mediated by PTMs.  相似文献

Analysis of frog virus 3-infected BHK cells by two-dimensional, acidic and basic gel electrophoresis showed that at least 90 infected cell-specific polypeptides could be detected. These polypeptides represent between 70 and 85% of the coding capacity of the viral genome. The polypeptides were sequentially induced in at least three phases. The virus gradually suppressed host cell polypeptide synthesis during infection, although the synthesis of a few cell polypeptides may be “switched off” early in infection.  相似文献

Summary The lamina fusca of the hamster eye contains layers of flattened, slightly overlapping fibroblasts. Thin sections of the overlapping margins reveal punctate, tight-junction-like membrane appositions associated with accumulation of cytoplasmic filaments, 5–7 nm in diameter. Intermediate filaments are present in the surrounding cytoplasm. A diffuse dense substance occurs in adjacent intercellular space. Freeze-fracture replicas show that the membrane appositions are mainly single-stranded tight junctions, each composed of two fibrils (micelles), and each continuous or nearly continuous around the fibroblastic perimeter. Fracturing characteristics of these junctions offer a unique opportunity to gain further insight into tight junctional morphology. When exposed, the fibrils adhere to the P-face, measure 9.2±0.3 nm in diameter, and are accompanied by a narrow band of membrane differing in texture from non-junctional membrane. Characteristically, the junctional fibrils themselves mark the deviation line along which fracture planes pass from one membrane of the junction to the other. This pattern exposes, over long distances, the P-face of one membrane on one side of this line and E-face of the adjacent membrane on the other. Analysis of any single junction over such distances reveals that the juxtaposition of the fibrils may gradually twist or undulate over a range of at least 180° within the two involved membranes. The fracture plane appears preferentially to pass between the two junctional fibrils; association of the cytoskeleton with junctional fibrils may govern this route of fracture. Cytoskeletal attachment appears to be to a single fibril and may alternate from one fibroblast to the next depending on which cytoplasmic leaflet is nearest a given fibril.Parts of this work have been presented at meetings of the Association for Research in Vision and Ophthalmology (Kelly and Hageman 1983) and the American Association of Anatomists (Hageman and Kelly 1984)  相似文献
