SYNOPSIS. The fine structure of schizonts and free merozoites of the neogregarine Farinocystis tribolii Weiser, and their development in the fat body of larval Tribolium castaneum were studied.
The surface of a multinucleate schizont and that of a uninucleate merozoite is covered by a double-layered membrane. Rhoptries and micronemes are present. The cytoplasm is packed with ribosomes and also contains dark bodies. Mitochondria are of the vesicular type. The spherical nucleus of the schizont and merozoite contains a large nucleolus. The anterior end of the merozoite has a typical conoidal complex composed of a conoid and a polar ring with 22 subpellicular mirotubules projecting from it.
New findings are a membranous septum across the body of the merozoite at 2/3 of its length below the nucleus and a highly osmiophilic spiral structure in the perinuclear space close to the Golgi complex. In addition, we found some "developmental stages" of the latter structure.  相似文献

Repeated epilation (Er) is a radiation-induced, autosomal, incomplete dominant mutation in mice which is expressed in heterozygotes but is lethal in the homozygous condition. Many effects of the mutation occur in skin: the epidermis in Er/Er mice is adhesive (oral and nasal orifices fuse, limbs adhere to the body wall), hyperplastic, and fails to undergo terminal differentiation. Skin from fetal +/+, Er/+ and Er/Er mice at ages pre- and postkeratinization examined by light, scanning, and transmission electron microscopy showed marked abnormalities in tissue architecture, differentiation, and cell structure; light and dark basal epidermal cells were separated by wide intercellular spaces, joined by few desmosomes, and contained phagolysomes. The numbers of spinous, granular, and superficial layers were highly variable within any given region and among various regions of the body. In some areas, 2-8 layers of granular cells, containing large or diminutive keratohyalin granules, extended to the epidermal surface; in others, the granular layers were covered by several layers of partially keratinized or nonkeratinized cells. In rare instances, a single or small group of cornified cells was present among the granular layers but was not associated with the epidermal surface. Both the granular and nonkeratinized/partially keratinized upper epidermal layers Er/Er skin gave positive immunofluorescence with antiserum to the histidine-rich, basic protein, filaggrin. Proteins in epidermal extracts from +/+, Er/+ and Er/Er mice were separated and identified by radio- and immunolabeling techniques. The Er/Er extract was missing a 26.5- kdalton protein and had an altered ratio of bands in the keratin region. The 26.5-kdalton band was histidine-rich and cross-reacted with the antiserum to rat filaggrin. Several high molecular weight bands present in both Er/Er and +/+ extracts also reacted with the antiserum. These are presumed to be the precursors of filaggrin and to account for the immunofluorescence om Er/Er epidermis even though the product protein is absent. The morphologic and biochemical data indicated that the genetic defect has a general and profound influence on epidermal differentiation, including alteration of two proteins (filaggrin and keratin) important in normal terminal differentiation, tissue architecture, and cytology. Identification of epidermal abnormalities at early stages of development (prekeratinization) and defective structure of other tissues and gross anatomy suggest that the mutation is responsible for a defect in same regulatory step important in many processes of differentiation and development.  相似文献

The three dimensional distribution of intercepted radiation, intercellular CO2 concentration (Ci) and late summer needle nitrogen (N) concentration were determined at the tips of all 54 branches in a 6·2-m-tall Pinus radiata D. Don tree growing in a New Zealand plantation. Measurements included above- and below-canopy irradiance, leaf stable carbon isotopic composition (δ13C) and tree canopy architecture. The radiation absorption component of the model, MAESTRO, was tested on site and then used to determine the branch tip distribution of intercepted radiation. We hypothesized that in branch tip needles: (i) the allocation of nitrogen and other nutrients would be closely associated with the distribution of intercepted radiation, reflecting carbon gain optimization theory, and (ii) Ci would predominantly reflect changes in photosynthetic rate (A) rather than stomatal conductance (gs), indicating that the increase in A for a given increase in N concentration was larger than the corresponding increase in gs. Needle nitrogen concentration was poorly related to intercepted radiation, regardless of the period over which the latter was calculated. At a given height, there was a large azimuthal variation in intercepted radiation but N concentration was remarkably uniform around the tree canopy. There was, however, a linear and positive correspondence between N concentration and δ13C and needle height above ground (r2 = 0·73 and 0·68, respectively). The very strong linear correspondence between N concentration and Ci (r2 = 0·71) was interpreted, using gas exchange measurements, as supporting our second hypothesis. Recognizing the strong apical control in P. radiata and possible effects of leaf nitrogen storage in an evergreen species, we propose that the tree leader must have constituted a very strong carbon sink throughout the growing season, and that the proximity of branch tip needles to the leader affected their photosynthetic capacity and nutrient concentration, independent of intercepted radiation. This implies an integrated internal determination of resource allocation within the tree and challenges the current convention that resources are optimally distributed according to the profile of intercepted radiation.  相似文献

The boreal forest, one of the world's larger biomes, is distinct from other biomes because it experiences a short growing season and extremely cold winter temperatures. Despite its size and impact on the earth's climate system, measurements of mass and energy exchange have been rare until the past five years. This paper overviews results of recent and comprehensive field studies conducted in Canada, Siberia and Scandinavia on energy exchanges between boreal forests and the atmosphere. How the boreal biosphere and atmosphere interact to affect the interception of solar energy and how solar energy is used to evaporate water and heat the air and soil is examined in detail. Specifically, we analyse the magnitudes, temporal and spatial patterns and controls of solar energy, moisture and sensible heat fluxes across the land–atmosphere interface. We interpret and synthesize field data with the aid of a soil–vegetation–atmosphere transfer model, which considers the coupling of the energy and carbon fluxes and nutrient status. Low precipitation and low temperatures limit growth of many boreal forests. These factors restrict photosynthetic capacity and lower root hydraulic conductivity and stomatal conductance of the inhabitant forests. In such circumstances, these factors interact to form a canopy that has a low leaf area index and exerts a significant resistance to evaporation. Conifer forests, growing on upland soils, for example, evaporate at rates between 25 and 75% of equilibrium evaporation and lose less than 2.5 mm day?1 of water. The open nature of many boreal conifer forest stands causes a disproportionate amount of energy exchange to occur at the soil surface. The climatic and physiological factors that yield relatively low rates of evaporation over conifer stands also cause high rates of sensible heat exchange and the diurnal development of deep planetary boundary layers. In contrast, evaporation from broad‐leaved aspen stands and fen/wetlands approach equilibrium evaporation rates and lose up to 6 mm day?1.  相似文献
