Summary Clonal stock (grafts of 15-, 50-, and 100-year-old trees) of spruce (Picea abies (L.) Karst.) and fir (Abies alba Mill.) was exposed either to 150 ppb ozone (300 gm-3) during 9 daylight hours or to carbon-filtered air on 35 days between 29 August and 21 October 1983. At the end of the fumigation the trees did not exhibit any visible signs of injury. Transpiration and leaf conductance in light and darkness, measured with a Licor porometer, revealed, however, stomatal sluggishness and — in spruce-increased transpiration.  相似文献

Summary Usin gintracellular microelectrode technique, the response of the voltageV across the plasma membrane of cultured bovine corneal endothelial cells to changes in sodium and bicarbonate concentrations was investigated. (1) The electrical response to changes in [HCO 3 ] o (depolarization upon lowering and hyperpolarization upon raising [HCO 3 ] o ) was dependent on sodium. Lithium could fairly well be substituted for sodium, whereas potassium or choline were much less effective. (2) Removal of external sodium caused a depolarization, while a readdition led to a hyperpolarization, which increased with time of preincubation in the sodium-depleted medium. (3) The response to changes in [Na+] o was dependent on bicarbonate. In a nominally bicarbonate-free medium, its amplitude was decreased or even reversed in sign. (4) Application of SITS or DIDS (10–3 m) had a similar effect on the response to sodium as bicarbonate-depleted medium. (5) At [Na+] o =151mm and [HCO 3 ] o =46mm, the transients ofV depended, with 39.0±9.0 (sd) mV/decade, on bicarbonate and, with 15.3±5.8 (sd) mV/decade, on sodium. (6) After the preincubation of cells with lithium, replacement of Li by choline led to similar effects as the replacement of sodium by choline, though the response ofV was smaller with Li. This response could be reduced or reversed by the removal of bicarbonate or by the application of SITS. (7) Amiloride (10–3 m) caused a reversible hyperpolarization of the steady-state potential by 8.5±2.6 mV (sd). It did not affect the immediate response to changes in [Na+] o or [HCO 3 ] o , but reduced the speed of regaining the steady-state potential after a change in [HCO 3 ] o . (8) Ouabain (10–4 m) caused a fast depolarization of –6.8±1.1 (sd) mV, which was followed by a continuing slower depolarization. The effect was almost identical at 10–5 m. (9) It is suggested, that corneal endothelial cells possess a cotransport for sodium and bicarbonate, which transports net negative charage with these ions. It is inhibitable by stilbenes, but not directly affected by amiloride or ouabain. Lithium is a good substitute for sodium with respect to bicarbonate transport and is transported itself. In addition, the effect of amiloride provides indirect evidence for the existence of a Na+/H+-antiport. A model for the transepithelial transport of bicarbonate across the corneal endothelium is proposed.  相似文献

Human ceroid lipofuscinosis (CL) is an inherited disease marked by cerebromacular degeneration and early death. We have utilized the canine model to investigate the possible role of dolichol and dolichyl phosphate in the developmental pathology of CL. We found that while brain levels of dolichol increase with age in both affected and unaffected dogs, the amount in the diseased animal was similar to that in controls. Brain levels of dolichyl phosphate ranged from 20 to 35 micrograms/g in control dogs at all ages examined, but increased substantially during development in the affected dogs, a value of 113 +/- 24 micrograms/g (mean +/- SD) being obtained in the end-stage animals. In addition to the results obtained in the canine model, dolichyl phosphate levels in human brain tissues from a 5-year-old with late infantile CL and from a 19-year-old with juvenile CL were found to be 153 and 382 micrograms/g, respectively, compared with a control that assayed 26 micrograms/g. The preliminary findings with human tissues provide further evidence for an association of elevated brain dolichyl phosphate levels with CL. Whether the increase is primary or secondary remains to be determined.  相似文献

Progestin receptors present in cytosols of myomal and myometrial origin were analyzed on sucrose density gradients of low ionic strength using a vertical tube rotor. Short term incubations (90 min) contained mainly 8S receptors. Most myometrial but few myomal preparations contained an additional 4S component, the presence of which was hormone dependent. None could be detected if the serum concentration of estradiol was high. Myometrial receptors incubated over night exhibited only a 4S peak, whereas many myomal ones still sedimented as 8S peaks. The 8 to 4S shift represented a transformation of the receptors and not a sequential interaction of the hormone with first an 8S and then a 4S entity. The transforming activity contained in the myometrial cytosol could also attack the myomal receptor as shown by mixing experiments. Only the 8S peak was observed, whenever the cytosol was first fractionated and then incubated with the hormone. Thus the transformation occurred only in presence of [3H]promegestone. It could be suppressed with protease inhibitors. It is concluded that these tissues contain a receptor processing protease which (1) is much less prominent in myomal than myometrial cytosol, which (2) attacks only the occupied receptors, and (3) the activity of which is hormone dependent.  相似文献

The poly(dG-dC) helical duplex forms a modified, B-family structure (B*) at very high hydration and a normal B structure at slightly lower hydration. The B* structure is slightly different in sugar-phosphate and base-stacking conformations than the B structure. Increasing the hydration or decreasing the NaCl content stabilizes B* with respect to B. Poly(dG-dC) forms the Z structure at low NaCl contents when the hydration is sufficiently reduced. At moderate NaCl content, the B to Z transition is sharp and cooperative for hydration with D2O. Hydration with H2O broadens the transition which occurs at lower hydration. This suggests that hydrogen bonding is stronger in the Z structure and helps stabilize Z over B. IR spectra may be used to quantitatively estimate the fractions of B and Z structures present in a sample. Some new indicator bands are described.  相似文献

Species ranges and relative abundances of dominant planktonic foraminifers of eight late Eocene to early Oligocene deep-sea sections are discussed to determine the nature and magnitude of extinctions and to investigate a possible cause-effect relationship between impact events and mass extinctions.Late Eocene extinctions are neither catastrophic nor mass extinctions, but occur stepwise over a period of about 1–2 million years. Four stepwise extinctions are identified at the middle/late Eocene boundary, the upperGlobigerapsis semiinvoluta zone, theG. semiinvoluta/Globorotalia cerroazulensis zone boundary and at the Eocene/Oligocene boundary. Each stepwise extinction event represents a time of accelerated faunal turnover characterized by generally less than 15% species extinct and in itself is not a significant extinction event. Relative species abundance changes at each stepwise extinction event, however, indicate a turnover involving > 60% of the population implying major environmental changes.There microtektite horizons are present in late Eocene sediments; one in the upperG. semiinvoluta zone (38.2 Ma) and two closely spaced layers only a few thousand years apart in the lower part of theGloborotalia cerroazulensis zone (37.2 Ma). Each of the three impact events appears to have had some effect on microplankton communities. However, the overriding factor that led to the stepwise mass extinctions may have been the result of multiple causes as there is no evidence of impacts associated with the step preceding, or the step following the deposition of the presently known microtektite horizons.  相似文献

本实验采用胶原酶消化,Ficoll 密度梯度离心,制备大鼠睾丸 Leydig 细胞悬浮液进行体外培养(每管内含有10~6 细胞),以研究酪氨酸对 Leydig 细胞睾酮和cAMP 生成的影响。实验结果表明,hCG(100mIU)能明显地促进Leydig 细胞睾酮和 cAMP的生成。睾酮从对照组的3.08±0.58ng(X±SD,下同)增加到41.61±1.52ng,cAMP 含量从19.62±2.56pmol增加到153.24±5.92pmol。若将酪氨酸(60μg)与hCG同时加入到细胞培养液中,则睾酮和cAMP 含量分别下降到 19.22±.0.52ng(P<0.01)和92.63±6.02pmol(P<0.05)。但是,酪氨酸羟化酶抑制剂(α-甲基酪氨酸)对酪氨酸抗hCG致睾酮生成作用无阻断效应,而酪氨酸对外源cAMP(2.5mM)诱导的睾酮生成,则有明显的抑制作用,睾酮含量从27.56±1.53ng降至 19.50±0.47ng(P<0.01)。以上实验结果表明,酪氨酸抗hCG致睾酮生成的作用机理与cAMP有关。  相似文献
