Polycomb Group (PcG) genes encode proteins that form large multimeric and chromatin-associated complexes implicated in the stable repression of developmentally essential genes. HPH2, the Homo sapiens polyhomeotic homologue 2, functions as one of the subunits of PcG complex 1. In our study, SIAH-1, an E3 ligase, could directly associate with HPH2 both in vitro and in vivo. Both the cysteine-rich region of SIAH-1 and the PxVxAxP motif of HPH2 were essential for the interaction. HPH2 was co-localized with SIAH-1 in nuclei. Furthermore, SIAH-1 was able to facilitate the ubiquitination and degradation of HPH2 via ubiquitin-proteasome pathway in vivo. The ubiquitination activity was severely impaired in the SIAH-1 mutant that either lost E3 ligase activity or had weakened binding ability with HPH2, strongly suggesting that SIAH-1 was the direct E3 ligase of HPH2. Thus, our results propose a novel role of SIAH-1 in regulating the expression level of HPH2 through the ubiquitin-proteasome pathway.  相似文献

Background

A single nucleotide polymorphism (SNP) at locus 11q23.3 (rs498872) in the near 5′-UTR of the PHLDB1 gene was recently implicated as a risk factor for gliomas in a genome-wide association study, and this involvement was confirmed in three additional studies.

Methodology/Principal Findings

To identify possible causal variants in the region, the authors genotyped 15 tagging SNPs in the 200 kb genomic region at 11q23.3 locus in a Chinese Han population-based case-control study with 983 cases and 1024 controls. We found evidence for an association between two independent loci (both the PHLDB1 and the ACRN1 genes) and a predisposition for gliomas. Among the multiple significant SNPs in the PHLDB1 gene region, the rs17749 SNP was the most significant [P = 1.31×10−6 in a recessive genetic model]. Additionally, two novel SNPs (rs2236661 and rs494560) that were independent of rs17749 were significantly associated with glioma risk in a recessive genetic model [P = 1.31×10−5 and P = 3.32×10−5, respectively]. The second novel locus was within the ARCN1 gene, and it was associated with a significantly reduced risk for glioma.

Conclusions/Significance

Our data strongly support PHLDB1 as a susceptibility gene for glioma, also shedding light on a new potentially candidate gene, ARCN1.  相似文献

Previously, we showed that OsGLU3, a β-1,4-endoglucanase, can affect the cellulose synthesis for root elongation in rice. And the phosphate starvation induced root elongation in rice depends on the function of OsGLU3. Understanding the emergence of cooperation in spatial public goods game remains a grand challenge across disciplines. In most previous studies, it is assumed that the investments of all the cooperators are identical, and often equal to 1. However, it is worth mentioning that players are diverse and heterogeneous when choosing actions in the rapidly developing modern society and researchers have shown more interest to the heterogeneity of players recently. For modeling the heterogeneous players without loss of generality, it is assumed in this work that the investment of a cooperator is a random variable with uniform distribution, the mean value of which is equal to 1. The results of extensive numerical simulations convincingly indicate that heterogeneous investments can promote cooperation. Specifically, a large value of the variance of the random variable can decrease the two critical values for the result of behavioral evolution effectively. Moreover, the larger the variance is, the better the promotion effect will be. In addition, this article has discussed the impact of heterogeneous investments when the coevolution of both strategy and investment is taken into account. Comparing the promotion effect of coevolution of strategy and investment with that of strategy imitation only, we can conclude that the coevolution of strategy and investment decreases the asymptotic fraction of cooperators by weakening the heterogeneity of investments, which further demonstrates that heterogeneous investments can promote cooperation in spatial public goods game.  相似文献

Energy homeostasis is an essential characteristic of all organisms, requiring fluctuation in energy accumulation, mobilization, and exchange with the external environment. In insects, energy mobilization is under control of the lipase brummer (bmm), which regulates nutritional status by hydrolyzing the ester bonds in triacylglycerol (TAG). In the present study, we investigated the role of bmm in the lipid mobilization and starvation resistance in the brown planthopper (BPH; Nilaparvata lugens), which is economically one of the most important rice pests in Asia. A severe decrease in TAG and glyceride contents was observed in the starved BPHs, while there was a partial rescue after refeeding. The starvation condition caused a significant increase in the expression levels of Nlbmm, and supplement of food after starvation dramatically reduced the Nlbmm expression. Sucrose rescue after starvation significantly suppressed the expression of Nlbmm, while caused an accumulation of TAG and glyceride. Knockdown of Nlbmm by double‐stranded RNA treatment extended the lifespan to starvation, whereas it increased the level of TAG and glyceride in the BPHs. The decreased lipolysis rate by dsNlbmm‐treated BPHs eventually resulted in increase of starvation resistance. These data demonstrated that the regulation of energy homeostasis by Nlbmm affects starvation resistance, probably through lipid mobilization control in N. lugens.  相似文献

Ecology of plankton in the downstream reaches of Kaduna River, Zungeru, Niger state, Nigeria was investigated between April and August 2015. Plankton and surface water samples were collected monthly from three stations for analysis of plankton diversity and physico-chemical parameters respectively. Station 1 is relatively unperturbed station located at the outskirts of Zungeru town, station 2 with moderate human activity located in Zungeru town while station 3 with minimal level of anthropogenic activities was located in Wushishi town about 7 km from station 2. The result revealed that the minimum and maximum values for pH water temperature, air temperature, conductivity, dissolved oxygen (DO) biochemical oxygen demand (BOD), phosphate, Nitrate, and secchi disc transparency were 5.38–6.79, 25–31 °C, 26–31 °C, 32–72 μs/cm, 3.50–8.20 mg/l, 1.00–5.00 mg/l, 0.06–1.13 mg/l 0.44–1.31 mg/l, 39.140 cm respectively. A total of 26 species of zooplankton and 24 species of phytoplankton were encountered in the study. The highest number of plankton (20 taxa) was recorded at station 2 while station 1 with 18 taxa and station 3 with 13 representative taxa. For zooplankton, the highest number of taxa was recorded at station 1 (20 taxa), followed by station 3 (14 taxa) and 10 taxa in station 2 were identified. Copepoda of the order cyclopidae dominated all the three stations while Bacillariophyta dominated the three stations for phytoplankton diversity. Canonical correspondence analysis (CCA) ordination was used to determine phytoplankton and zooplankton abundance in relation to the physico-chemical parameters. Nitrate, phosphate and dissolved oxygen had over riding influence on the distribution of the plankton in the water body which indicate that the river is still very productive in terms of providing starter food organisms for higher aquatic life. However, care should be taken to protect the river from further deterioration due to various degrees of human activities.  相似文献

A limitation of conventional optical coherence tomography angiography (OCTA) is the limited field of view normally used in data acquisition. As the technology improves, larger fields of view that capture information away from the macular are being explored in order to provide an enhanced ability to detect pathology. However, normative measurements for important OCTA metrics like vessel density and intercapillary distance are not currently well-characterized in the peripheral retina. In this prospective study, we measured vessel density and intercapillary distance of the superficial vascular complex, ganglion cell layer plexus, and deep capillary plexus in montaged macular/temporal scans from 53 (33 men) healthy volunteers. Vessel density and intercapillary distance were also compared across different regions of the retina, including along arcs at separate distance from the fovea. Compared to the central macular region, the temporal retina had significantly lower vessel density, decreased thickness, and greater intercapillary distance in the superficial vascular complex, GCLP ganglion cell layer plexus, and deep capillary plexus (Wilcoxon rank sum test P < 0.001), with each of the plexuses examined here showing a general decrease in vessel density and an increase in intercapillary distance towards the temporal region. No significant difference was noted comparing corresponding vessel density and intercapillary distance regions above and below the macula, and multiple linear regression showed that age and intraocular pressure were not associated with vessel density and intercapillary distance in most models. Repeatability analysis reported as intraclass correlation coefficients demonstrated moderate to excellent reliability of vessel density and intercapillary distance in all OCTA layers. These results should help provide an enhanced baseline to help identify vascular pathology in the peripheral retina.  相似文献

Revealing selectivity mechanism of cyclin-dependent kinases (CDK) and their inhibitors is an important issue to develop potential anticancer drugs. The substituted 4-(Pyrazol-4-yl)-pyrimidines are potent inhibitors of CDK4 but not of the highly homologous CDK2. In order to reveal the inhibitory selectivity of these inhibitors to CDK4 over CDK2, we select one of substituted 4-(Pyrazol-4-yl)-pyrimidines as a representative (marked as A1 hereunder) and perform molecular docking, molecular dynamics simulations and binding free energy analysis for CDK4/A1 and CDK2/A1, respectively. The electrostatic and van der Waals (vdW) interactions of the A1 inhibitor with CDK4/CDK2 are discussed. The computed binding free energies based on the MM-PBSA method are consistent with experimental bioactivity ranking of A1 inhibitor to CDK4/CDK2. On the other hand, the conformational characteristics of CDK2 and CDK4 induced by A1 inhibitor are analysed and revealed. Results demonstrate that the vdW interactions considerably contribute to binding of CDK4/CDK2 with A1 inhibitor and are similar in size. The hydrogen bonding between A1 inhibitor and CDK4/CDK2 is considerably favourable to the binding, in which the hydrogen bond between the NH group of the pyrazole group of A1 and the residue Asp158 of CDK4 plays a crucial role in inhibitory selectivity of A1 inhibitor to CDK4 over CDK2. The electrostatic interaction energy differences between the corresponding residues of CDK4/A1 and CDK2/A1 confirm the above inference. The conformational changes of CDK2 and CDK4 induced by A1 inhibitor influence the selectivity of A1 inhibitor to CDK4/CDK2.  相似文献
