黄足黑守瓜是普通丝瓜生长过程中危害叶片的主要害虫.本文对普通丝瓜不同部位叶片挥发性成分、营养成分和物理结构对黄足黑守瓜取食和定位的影响进行了系统研究.本实验采用顶空固相微萃取-气相色谱-质谱法分析了普通丝瓜不同部位叶片挥发性成分,研究结果表明,嫩叶主要挥发性成分有(Z)-3-己烯-1-醇(27.65%)和植醇(45.77%),成熟叶挥发性成分主要有(Z)-3-己烯-1-醇(49.34%),老叶挥发性成分主要有植醇(63.25%),且挥发性成分对黄足黑守瓜远距离定位起到了引导作用.同时,本研究还采用5点取样法、3,5-二硝基水杨酸法和考马斯亮蓝法分别研究了普通丝瓜不同部位叶片营养成分含量与黄足黑守瓜取食之间的关系,实验结果表明,黄足黑守瓜喜食植物叶片嫩叶部位,喜食部位叶片的总糖与还原糖含量、蛋白质含量都最高,且总糖含量与蛋白质含量比值最高.叶片物理结构调查采用石蜡切片法,发现叶片的物理结构对于黄足黑守瓜取食很少造成影响.通过研究,使我们充分地了解影响黄足黑守瓜取食不同部位叶片的因素,便于今后的育种和栽培过程中加强不利于黄足黑守瓜取食的因素,以达到安全、环保地进行农业生产的目的.  相似文献

The sweet potato whitefly, Bemisia tabaci (Gennadius) biotype B was first found in southern China in 1994 and has since then become a pest of increasing importance throughout the nation. The population dynamics of this invasive species and the effect of natural enemies on its population dynamics were studied in commercial cotton fields during the summers of 2003 through 2005 in Langfang, Hebei (northern China). The abundance of Bemisia tabaci was found to increase rapidly in late July, and reached its first peak in mid-August, then followed by the second peak in early September. Whitefly populations subsequently declined slowly to attain a plateau after mid-September. A total of 13 natural enemy species were discovered in association with B. tabaci in cotton fields, including dominant species such as Chrysopa sinica Tjeder, Leis axyridis Pallas, Propylaea japonica Thunberg, Orius similis Zheng, and Eretmocerus sp. Caged experiments on velvetleaf, Abutilon theophrasti Medic, one of the preferred host plants of the insect, indicated higher B. tabaci population densities in small-mesh cages with exclusion of all natural enemies versus those in medium- and large-mesh cages, and the uncaged plants retained the lowest population densities of B. tabaci. While no natural enemies were observed in the small-mesh cages, natural enemies like Misumenopos tricuspiaata, Eretmocerus sp., Encarsia formosa Orius similis, Deraeocoris punctulatus and Scolothrips takahashii were found in medium- and/or large-mesh cages. Findings from this study suggest that under open-field conditions, various indigenous natural enemies of different sizes may act in concert in the suppression of B. tabaci populations.  相似文献

The active vitamin D(3)-metabolite 1,25(OH)(2)D(3) inhibits the interleukin 4/granulocyte-macrophage colony-stimulating factor (IL-4/GM-CSF)-induced differentiation of human monocytes into dendritic cells without altering survival. Colony-stimulating factor 1 (CSF-1) is an important survival factor for cells of the monocytic lineage. We therefore investigated whether the inhibitory activity of 1,25(OH)(2)D(3) is paralleled by a regulation of CSF-1 and its receptor. Purified human monocytes were cultured together with IL-4/GM-CSF in the presence of 1,25(OH)(2)D(3), its analogue tacalcitol, the low-affinity vitamin D receptor ligand 24,25(OH)(2)D(3), or the solvent ethanol for up to 5 days. Expression of CSF-1, CSF-1R, and GM-CSF mRNA was measured by RT-PCR. Protein secretion for CSF-1 was measured by ELISA, expression of CSF-1R by flow cytometry. The results showed that 1,25(OH)(2)D(3) and tacalcitol significantly up-regulated CSF-1 mRNA-expression and protein secretion in a dose-dependent manner. The effect of 1,25(OH)(2)D(3) occurred already after 1h of pre-treatment. In contrast, CSF-1R mRNA- and cell surface-expression was down-regulated simultaneously. The solvent ethanol and 24,25(OH)(2)D(3) were without effect. GM-CSF mRNA expression was not modulated in 1,25(OH)(2)D(3)-treated cells. These data point towards a distinct and specific regulation of CSF-1 and its receptor by 1,25(OH)(2)D(3) and its analogue tacalcitol in human monocytes which parallels the inhibition of differentiation into dendritic cells without altering survival.  相似文献

Interannual climate variation alters functional diversity through intraspecific trait variability and species turnover. We examined these diversity elements in three types of grasslands in northern China, including two temperate steppes and an alpine meadow. We evaluated the differences in community‐weighted means (CWM) of plant traits and functional dispersion (FDis) between 2 years with contrasting aridity in the growing season. Four traits were measured: specific leaf area (SLA), leaf dry matter content (LDMC), leaf nitrogen concentration (LNC), and the maximum plant height (H). CWM for SLA of the alpine meadow increased in the dry year while that of the temperate steppe in Qinghai showed opposing trends. CWM of LDMC in two temperate steppes became higher and CWM of LNC in all grasslands became lower in the dry year. Compared with the wet year, FDis of LDMC in the alpine meadow and FDis of LNC in the temperate steppe in Qinghai decreased in the dry year. FDis of H was higher in the dry year for two temperate steppes. Only in the temperate steppe in Qinghai did the multi‐FDis of all traits experience a significant increase in the dry year. Most of the changes in CWM and FDis between 2 years were explained by intraspecific trait variation rather than shifts in species composition. This study highlights that temporal intraspecific trait variation contributes to functional responses to environmental changes. Our results also suggest it would be necessary to consider habitat types when modeling ecosystem responses to climate changes, as different grasslands showed different response patterns.  相似文献

The effects and mechanisms of mitochondrial DNA (mtDNA) in the development of sepsis-induced lung injury is not well understood. In our present study, we studied the mtDNA effects in sepsis-induced lung injury model, in vitro and in vivo. Compared with the Normal group, the lung histopathological score, the number of positive apoptosis cell, wet/dry (W/D) ratio and TNF-α, IL-1β, and IL-6 concentrations of lipopolysaccharides (LPSs) and mtDNA groups were significantly increased (P < 0.001, respectively). Meanwhile, the lung histopathological score, positive W/D ratio, number of apoptosis cell and tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, and IL-6 concentrations of LPS + mtDNA and small interfering RNA (siRNA)-NC + LPS + mtDNA groups were significantly upregulated compared with those of LPS group (P < 0.05, respectively). However, the lung histopathological score, the number of positive apoptosis cell, W/D ratio and TNF-α, IL-1β, and IL-6 concentrations were significantly improved within the toll-like receptor (TLR9)siRNA + LPS + mtDNA group compared with the LPS group (P < 0.01, respectively). The TLR9, MyD88, and NF-κB proteins or gene expressions of the LPS group and mtDNA group were significantly upregulated compared with those of Normal group by Western blot analysis or immunohistochemistry assay (P < 0.01, respectively), and the TLR9, MyD88, and NF-κB proteins or gene expressions of LPS + mtDNA and siRNA-NC + LPS + mtDNA groups were significantly enhanced compared with those of LPS group (P < 0.05, respectively). However, the TLR9, MyD88, and NF-κB proteins or gene expressions of TLR9siRNA + LPS + mtDNA group were significantly suppressed compared with those of the LPS group (P < 0.01, respectively). In conclusion, mtDNA could provoke lung injury induced by sepsis via regulation of TLR9/MyD88/NF-κB pathway in vitro and in vivo.  相似文献

High-quality and disease-resistant male sterile lines have great potential for applications in hybrid rice breeding. We introduced specific mutations into the TMS5, Pi21, and Xa13 genes in Pinzhan intermediate breeding material using the CRISPR/Cas9 multiplex genome editing system. We found that the transgenefree homozygous triple tms5/pi21/xa13 mutants obtained in the T1 generation displayed characteristics of thermosensitive genic male sterility(TGMS) with enhancedresistance to rice blast and bacterial blight. Our study provides a convenient and effective way of converting breeding intermediate material into TGMS lines through multiplex gene editing, which could significantly accelerate the breeding of sterile lines.  相似文献

目的总结和分析心脏外科手术中应用冠状动脉顺行灌注联合冠状静脉窦逆行灌注和冠状动脉桥灌注技术进行心肌保护。方法30例患者分为2组:A组(顺灌联合逆灌技术)20例和B组(顺逆灌结合桥灌技术)10例,疾病种类有:冠心病合并瓣膜病、冠心病合并室壁瘤、升主动脉病变合并主动脉瓣病变和单纯瓣膜病变。结果术中转流平稳,血流动力学稳定,监测指标均在正常范围,无手术死亡和围手术期并发症。结论采用冠状动脉顺行灌注联合冠状静脉窦逆行灌注或结合冠状动脉桥灌注心肌停搏液进行心肌保护,取得良好效果。  相似文献
