Alpha-bag cell peptide [alpha-BCP (Ala-Pro-Arg-Leu-Arg-Phe-Tyr-Ser-Leu)] is a neurotransmitter that mediates bag cell-induced inhibition of left-upper-quadrant (LUQ) neurons L2, L3, L4, and L6 in the abdominal ganglion of Aplysia. Our recent biochemical studies have shown that alpha-BCP[1-9] is cleaved into alpha-BCP[1-2], [3-9], [1-5], [6-9], and [7-9] by a combination of three distinct peptidase activities located within the extracellular spaces of the CNS: A diaminopeptidase-IV (DAP-IV)-like enzyme cleaves alpha-BCP[1-9] at the 2-3 peptide bond; a neutral metalloendopeptidase (NEP)-like enzyme cleaves either alpha-BCP[1-9] or alpha-BCP[3-9] at the 5-6 bond; an aminopeptidase M-II (APM-II)-like enzyme cleaves alpha-BCP[6-9] at the 6-7 bond, but cleaves neither alpha-BCP[1-9], nor the other ganglionic peptidase products. To further understand the manner in which alpha-BCP is inactivated after release, that is loses its electrophysiological activity, we studied its structure-activity relationship by recording intracellularly from LUQ neurons in isolated abdominal ganglia that were arterially perfused with peptides dissolved in artificial sea water. The effects of alpha-BCP[1-9] and 15 of its fragments ([1-8], [1-7], [1-6], [1-5], [2-9], [3-9], [3-8], [6-9], [7-9], [8-9], [6-7], [6-8], [1-2], Phe, Tyr) indicated that the sequence Phe6-Tyr7 was both necessary and sufficient to produce LUQ inhibitory activity.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Evidence is provided that ethylene is a positive regulator of hair cell development in the root epidermis of Arabidopsis thaliana. Treatment of seedlings with increasing concentrations of the ethylene precursor, 1-aminocyclopropane-1-carboxylic acid (ACC) results in progressively more root hair cells developing in positions normally occupied by non-hair cells. Consistent with these findings are observations that treatments that block either ethylene synthesis or its perception reduce the number of root hairs. A model is proposed in which either ethylene or ACC is a signal involved in specifying the pattern of cell differentiation in the Arabidopsis root epidermis. 相似文献

We compare the dynamics of artificial populations of the annual Phlox drummondii introduced along a transect extending from within to beyond the species margin southwest of San Antonio, Texas. The margin is defined by a shift from sandy, nutrient poor soils to clay, nutrient-rich soils. Fourteen sites were chosen along the 6.5-mi transect. In each site seeds were planted into 1.5-m-square cleared plots and uncleared 10-m transects. Transplants also were introduced into cleared plots. Survival, flower production and seed set were monitored over the course of the growing season. Individual fitnesses and the reproductive rates of artificial populations generally declined as the distance from the margin increased. The results indicate soil type per se is not limiting Phlox and that plants can successfully grow and reproduce, albeit at a reduced level, in sites beyond their natural margin. 相似文献

Summary The juvenile hormone esterase (JHE) titer was measured during the last larval instar of 11 species of Lepidoptera (Pieris rapae, Junonia coenia, Danaus plexippus, Hemileuca nevadensis, Pectinophora gossypiella, Spodoptera exigua, Orgyia vetusta, Ephestia elutella, Galleria mellonella, Manduca sexta andEstigmene acrea). All species had a peak of JHE at or near the time of wandering. The peak activity at this time ranged from 0.8 to 388 nmoles JH III cleaved/min·ml. All species exceptJ. coenia had a second peak of JHE during the late prepupal stage. The height of the second peak ranged from 0.4 to 98.4 nmoles/min·ml. However, there was no apparent correlation between size of the first and second JHE activity peaks for the lepidopteran species examined. There was an apparent relationship between the height of the first and second JHE peaks and reports on titer of JH just prior to these peaks. These data support, with some qualifications, the extension of developmental information obtained on several well studied species to a variety of Lepidoptera.Abbreviations

Two cDNAs, GluClα and GluClβ, encoding glutamate-gated chloride channel subunits that represent targets of the avermectin
class of antiparasitic compounds, have recently been cloned from Caenorhabditis elegans (Cully et al., Nature, 371, 707–711, 1994). Expression studies in Xenopus oocytes showed that GluClα and GluClβ have pharmacological profiles distinct from the glutamate-gated cation channels as
well as the γ-aminobutyric acid (GABA)- and glycine-gated chloride channels. Establishing the evolutionary relationship of
related proteins can clarify properties and lead to predictions about their structure and function. We have cloned and determined
the nucleotide sequence of the GluClα and GluClβ genes. In an attempt to understand the evolutionary relationship of these
channels with the members of the ligand-gated ion channel superfamily, we have performed gene structure comparisons and phylogenetic
analyses of their nucleotide and predicted amino acid sequences. Gene structure comparisons reveal the presence of several
intron positions that are not found in the ligand-gated ion channel superfamily, outlining their distinct evolutionary position.
Phylogenetic analyses indicate that GluClα and GluClβ form a monophyletic subbranch in the ligand-gated ion channel superfamily
and are related to vertebrate glycine channels/receptors. Glutamate-gated chloride channels, with electrophysiological properties
similar to GluClα and GluClβ, have been described in insects and crustaceans, suggesting that the glutamate-gated chloride
channel family may be conserved in other invertebrate species. The gene structure and phylogenetic analyses in combination
with the distinct pharmacological properties demonstrate that GluClα and GluClβ belong to a discrete ligand-gated ion channel
family that may represent genes orthologous to the vertebrate glycine channels.

Biometry and phenology of two sibling Phylloscopus warblers on their circum-Mediterranean migrations

The Mediterranean Sea is known as an ecological barrier for numerous migratory birds flying from European breeding grounds to African wintering sites. Birds generally avoid migration over open sea and fly over land. In the Mediterranean Basin, few land bridges or bottlenecks for migratory birds exist. The narrowest are at the western and eastern extremes: the Strait of Gibraltar and Israel. Comparative studies between these locations are extremely rare to date. Therefore, in order to elucidate the differences between the two flyways, we compared data collected simultaneously for two sister leaf warbler species, the Bonelli’s Warbler complex, Phylloscopus
bonelli and Phylloscopus
orientalis, at ringing stations in the western Mediterranean Basin Gibraltar, and the eastern Eilat, Israel. Data on biometrics and passage dates of individuals trapped at Gibraltar and Eilat were used, and it was found that mean arrival date of Western Bonelli’s Warblers at Gibraltar was 15 days later than Eastern Bonelli’s Warblers at Eilat. Furthermore, Western Bonelli’s Warblers had shorter wings than Eastern Bonelli’s Warblers. On the other hand, birds in Eilat were in poorer body condition than individuals in Gibraltar. The comparison between geographically distant stop-over sites contributes to furthering our understanding of the development of migration strategies across ecological barriers in sibling species. Our study showed that populations that breed in southwestern Europe migrate through Gibraltar and winter in West Africa are able to accomplish migration in comparatively good body condition. This is in contrast to those that winter in East Africa, migrate through Israel and have to endure the combined challenge of crossing the Sahel, Sahara and Sinai deserts before reaching their breeding grounds across southeast Europe and southwest Asia. Hence, the discrepancies described between the western and the eastern flyway suggest that individuals in the west, in general, migrate shorter distances, have a physiologically less demanding crossing of the North African deserts and appear to stage before their crossing the Strait of Gibraltar, a privilege unavailable to the migrants of the eastern flyway. 相似文献

Management programs that successfully recovered wild saltwater crocodile (Crocodylus porosus) populations in the Northern Territory of Australia did so with an expanding commitment to maintaining public safety. One aspect of the program is the ongoing removal of resident and immigrant crocodiles within Darwin Harbor (since 1979), the main urban center. We determined the likely sources of crocodiles caught as problem animals between 2015–2017 by comparing recently developed methods for population assignment. Depending on the assignment model used, we estimated that between 30% and 50% of crocodiles in Darwin Harbor originated from the Adelaide and Mary rivers, and the Kakadu region east of Darwin, and between 20% and 30% of crocodiles originated from the Finniss, Reynolds, and Daly rivers southwest of Darwin. Saltwater crocodiles occur at particularly high densities in these catchments. The remainder came from a mixture of different sources across the Northern Territory. The most common animals captured were immature (150–180 cm) males that have traveled 100–200 km. We did not identify any relationships between the distance from the inferred origin to Darwin Harbor and the size and sex of the crocodiles, or the year of capture. The targeted removal of crocodiles from specific sites such as Darwin Harbor, near where most people live, improves public safety in the highest risk areas, without compromising abundant source populations in most areas. 相似文献

Thymus involution is a useful marker of transactivation-mediated side effects in preclinical therapeutic index testing of new anti-inflammatory glucocorticosteroids, and is usually measured post mortem. We have validated the use of MRI for non-invasive in vivo measurement of mouse thymus involution induced by dexamethasone (DEX). Tl-weighted spin echo 7 T images provided satisfactory contrast between thymus and surrounding connective tissue and fat. Increasing doses of DEX caused thymus involution, reflected in MRI volume (87+/-14, 33+/-10, 28+/-6, 16+/-7 microl in dosage groups of Cremophor vehicle, 1, 10 and 30 mg/kg subcutaneous respectively, n=6/group, mean+/-standard deviation) and post mortem wet weight (64+/-12, 33+/-6, 25+/-9, 23+/-8 mg). Correlation between MRI volumes and wet weights was very good (r=0.842). Measuring pre-dose MRI volumes and then assessing DEX effects as post-dose change from baseline produced no statistical advantage relative to considering post-dose MRI thymus volume alone, probably due to variability in pre-dose baseline values compounding post-dose variability. Smaller group sizes were sufficient to achieve a given statistical power using MRI post-dose volume than using wet weight, suggesting a role for MRI in differentiating the effects of compounds which produce similar effects, or in contexts where the use of large groups of animals is impractical or ethically unacceptable. 相似文献

Phytoplankton underpin the marine food web in shelf seas, with some species having properties that are harmful to human health and coastal aquaculture. Pressures such as climate change and anthropogenic nutrient input are hypothesized to influence phytoplankton community composition and distribution. Yet the primary environmental drivers in shelf seas are poorly understood. To begin to address this in North Western European waters, the phytoplankton community composition was assessed in light of measured physical and chemical drivers during the "Ellett Line" cruise of autumn 2001 across the Scottish Continental shelf and into adjacent open Atlantic waters. Spatial variability existed in both phytoplankton and environmental conditions, with clear differences not only between on and off shelf stations but also between different on shelf locations. Temperature/salinity plots demonstrated different water masses existed in the region. In turn, principal component analysis (PCA), of the measured environmental conditions (temperature, salinity, water density and inorganic nutrient concentrations) clearly discriminated between shelf and oceanic stations on the basis of DIN:DSi ratio that was correlated with both salinity and temperature. Discrimination between shelf stations was also related to this ratio, but also the concentration of DIN and DSi. The phytoplankton community was diatom dominated, with multidimensional scaling (MDS) demonstrating spatial variability in its composition. Redundancy analysis (RDA) was used to investigate the link between environment and the phytoplankton community. This demonstrated a significant relationship between community composition and water mass as indexed by salinity (whole community), and both salinity and DIN:DSi (diatoms alone). Diatoms of the Pseudo-nitzschia seriata group occurred at densities potentially harmful to shellfish aquaculture, with the potential for toxicity being elevated by the likelihood of DSi limitation of growth at most stations and depths. 相似文献
