Mutations in the rfbT Gene Are Responsible for the Ogawa to Inaba Serotype Conversion in Vibrio cholerae O1

In cultures of Vibrio cholerae strains of Ogawa serotype, variant strains which had undergone serotype conversion from Ogawa to Inaba were identified. The rfbT genes cloned from the parent strains were found to produce a 31-kDa protein in the maxicell system, and to cause serotype conversion when introduced into E. coli cells expressing Inaba serotype specificity. On the other hand, rfbT genes cloned from the variant strains neither produced the 31-kDa protein nor caused serotype conversion. Nucleotide sequence of these rfbT genes as well as those of two clinical Vibrio cholerae strains of Inaba serotype revealed that mutations causing premature termination of their rfbT genes were invariably present in strains expressing Inaba serotype specificity. The result strongly suggested that genetic alteration of the rfbT gene is responsible for serotype conversion of Vibrio cholerae O1.     

Bird carcass detection from integrated trials at multiple wind farms

It is often necessary to estimate the number of wind turbine collision fatalities to assess impacts to birds following construction of wind farms. Detection of bird carcasses at wind turbines in the field is affected by carcass persistence and searcher detection rate. Integrated detection trials, which integrate carcass persistence and searcher detection trials into the periodic fatality search, have been proposed as an effective method for estimating these parameters. The purpose of our study was to test whether and how environmental factors affect integrated detection trial outcomes at multiple wind farms. We conducted this study at 10 wind farms in various environments of Japan. Binary data on trial outcomes in open versus forested areas served as our response variable in a generalized additive mixed model informed by days into trial, carcass body mass, season, whether snow covered the ground, and precipitation. For both ground cover types, days into trial and body mass were included in all the top models, suggesting that these factors most influenced bird carcass detection probability in integrated trials. The best model in open areas included days into trial, body mass, snow, and precipitation, and the best model in forested areas included days into trial, body mass, snow, precipitation, and season. Values of area under the curve indicated high accuracy of the best model for both ground cover types. The survey design needs to be appropriate to the size of the target species and to the environment in which the impacts will occur, such as the site's seasonality, its ground cover, and whether snow will cover the ground. Frequency of post-construction fatality monitoring should also be set cautiously, especially at wind farms located on small-bird migration routes, at wind farms in open areas, in areas with snow-covered ground in winter, or in forested areas during spring and summer because detection probabilities decline fastest under such conditions.     

Changes in food intake and growth rate in mice under hypergravity.

Mice exposed to hypergravity, especially soon after start of exposure, diminished the body weight and the food intake. The amount of food intake was kept less than that of the ground control during hypergravity breeding for 2 weeks. Furthermore, the weight of testis relative to the body weight increased compared to that of 1 G control although the relative weights of liver and kidney were not changed. The purpose of this study was whether the low growth rate of the body weight and the increase of the relative testis weight were induced by the decrease of the food intake under hypergravity. We divided 3 weeks old male mice to 3 groups; the 1 G (ground control), the food restricted (FR) under 1 G, and the 3 G group. The 3 G group bred for 2 weeks under the centrifuge at 3 G. The FR group was given the same amount of food as the group ate. The changes in the body weight and the relative weights of testis, spleen and seminal vesicle of the FR group were similar to those of the 3 G group. The hunger test revealed that only the FR group was hunger. Our results suggested that the decrease of the food intake both in response to hypergravity and the food restriction induced the decrease of the body weight but the increase of the relative testis weight.     

FAB CID-MS/MS characterization of tetrasaccharide tri- and tetrasulfate derived from the antigenic determinant recognized by the anti-chondroitin sulfate monoclonal antibody MO-225

Kinetics and collagenolytic role of eosinophils in chronic gastric ulcer in the rat

 An association between eosinophils and tissue damage has been observed in numerous disorders. However, few reports have addressed the role of infiltrating eosinophils in gastric ulcer healing. The aim of this study was to investigate the kinetics and role of eosinophils infiltrating experimental chronic gastric ulcers in the rat. We developed a monoclonal antibody against human matrix metalloproteinase 1 (MMP1) purified from conditioned culture medium of human skin fibroblasts. Acetic acid-induced gastric ulcers were resected from rats on days 1, 3, 5, 10, 20, 40, and 180 after the days of induction (day 0). Tissue specimens were immunostained with this antibody and examined with an electron microscope. Few eosinophils were observed in the granulation tissue until day 20. By days 40 and 180, MMP1-positive eosinophils had increased in the granulation tissue of open ulcers. Azan staining revealed dispersed collagen fibers around infiltrating eosinophils. In contrast, scars demonstrated few eosinophils in fibrous tissue on days 40 and 180. Eosinophils which express MMP1 infiltrate granulation tissue at the chronic stage of gastric ulceration. The results suggest that eosinophils may play a role in tissue remodeling and deterioration of ulceration. Accepted: 18 March 1997     

An immunohistochemical study of epithelial cells in the posterior lobe and pars tuberalis of the human adult pituitary gland

Summary Pituitary glands were examined using reference staining (hematoxylin and eosin, periodic acid-Schiff and alcian blue) and the peroxidase-labeled antibody method, for 1) invading anterior cells in the posterior lobe, 2) intermediate colloid forming follicles, and 3) pars tuberalis cells.The results showed: 1) that the majority of cases possessed invading anterior cells of various amount. Most of these cells were positive for ACTH^1–18, ACTH^17–39 and -MSH. However, on a few occasions, scattered GH, PRL, FSH, FSH, LH and even TSH cells were also present. 2) Colloid forming follicular cells were mostly ACTH cells, but also contained occasional other hormone-secreting cells. Hormone negative cells were correlated with salivary type epithelium. Well established acinic type salivary glands and ciliated epithelium were negative for any hormones immunohistochemically. 3) Pars tuberalis cells were predominantly gonadotrophs but also included TSH and ACTH cells. Some cells appeared to contain both FSH and LH. When these cells underwent squamous metaplasia, they seemed to lose their hormone secreting activity.Part of this study was supported by a Grant-in-Aid for Cancer Research from the Ministry of Education, Science and Culture, Japan     

(+)-5,17-dehydromatrine N-oxide,a new alkaloid in Euchresta japonica

A new lupin alkaloid, (+)-5,17-dehydromatrine N-oxide, was isolated from the fresh aerial parts of Euchresta japonica. Its structure was confirmed by spectrometric data and by direct comparison with a synthetic sample, prepared from (+)-sophoranol ((+)-5-hydroxymatrine). It was also concluded that (+)-5,17-dehydromatrine N-oxide and (+)-matrine N-oxide possess the same configuration with respect to the asymmetric nitrogen by NMR spectra.     

Pumpkin (Cucurbita sp.) seed globulin III. Comparison of subunit structures among seed globulins of various Cucurbita species and characterization of peptide components

Various Cucurbita seed globulins showed patterns similar toone another on SDS-gel electrophoresis, and ß bandsfor unreduced globulins and , ', and ' bands for reduced ones.On gel electrophoresis in 6 M urea, reduced globulin gave twoacidic and two basic bands. These corresponded to and ' chainsand ₁ and ₂ chains, respectively, identified by two-dimensionalurea-SDS gel electrophoresis. The compositions of the and ßsubunits were proposed.
