The object was to determine the influence of dietary vitamin E, selenium and their combination on the fatty acid con-tent of erythrocytes, bone marrow and spleen lipids of Akkaraman lambs. After supplementation for 15 days, the amount of all fatty acids was slightly higher (p < 0·05) in the vitamin E as compared to the control group, whereas the amount of longer fatty acids was significantly higher (p < 0·01, p < 0·001) in the selenium and combination groups. On the thirtieth day, the amount of all fatty acids was slightly high (p < 0·5) in all the supplemented groups in comparison with the control group. In the bone marrow lipids, the amount of longer fatty acids was decreased (p < 0·05, p < 0·01, p < 0·001) in the vitamin E and combination groups as compared to the control. Although the amount of some fatty acids was high (p < 0·05, p < 0·01) in the selenium group compared to the control, linoleic (18:2), linolenic (18:3) and the polyunsaturated fatty acids (PUFA) were lower (p < 0·05, p < 0·001). In the spleen lipids, the amount of longer fatty acids was slightly decreased (p < 0·05) in the vitamin E group as compared with the control; however the amount of longer fatty acids was significantly higher (p < 0·05, p < 0·01) in the selenium and combination groups in comparison to the control group. Thus dietary supplementation with selenium was more effective than dietary vitamin E supplementation in altering the fatty acid content of the erythrocyte, bone marrow and spleen lipids. © 1997 John Wiley & Sons, Ltd.  相似文献

Myxozoans are spore-forming, metazoan parasites common in cold-blooded aquatic vertebrates, especially fishes, with alternate life cycle stages developing in invertebrates. We report nine cases of infection in free-flying native and captive exotic ducks (Anseriformes: Anatidae) from locations across the United States and describe the first myxozoan in birds, Myxidium anatidum n. sp. We found developmental stages and mature spores in the bile ducts of a Pekin duck (domesticated Anas platyrhynchos). Spores are lens-shaped in sutural view, slightly sigmoidal in valvular view, with two polar capsules, and each valve cell has 14-16 longitudinal surface ridges. Spore dimensions are 23.1 microm x 10.8 microm x 11.2 microm. Phylogenetic analysis of the ssrRNA gene revealed closest affinity with Myxidium species described from chelonids (tortoises). Our novel finding broadens the definition of the Myxozoa to include birds as hosts and has implications for understanding myxozoan evolution, and mechanisms of geographical and host range extension. The number of infection records indicates this is not an incidental occurrence, and the detection of such widely dispersed cases suggests more myxozoans in birds will be encountered with increased surveillance of these hosts for pathogens.  相似文献

The rhizobacterium Pseudomonas fluorescens CHA0 promotes the growth of various crop plants and protects them against root diseases caused by pathogenic fungi. The main mechanism of disease suppression by this strain is the production of the antifungal compounds 2,4-diacetylphloroglucinol (DAPG) and pyoluteorin (PLT). Direct plant growth promotion can be achieved through solubilization of inorganic phosphates by the production of organic acids, mainly gluconic acid, which is one of the principal acids produced by Pseudomonas spp. The aim of this study was to elucidate the role of gluconic acid production in CHA0. Therefore, mutants were created with deletions in the genes encoding glucose dehydrogenase (gcd) and gluconate dehydrogenase (gad), required for the conversion of glucose to gluconic acid and gluconic acid to 2-ketogluconate, respectively. These enzymes should be of predominant importance for rhizosphere-colonizing biocontrol bacteria, as major carbon sources provided by plant root exudates are made up of glucose. Our results show that the ability of strain CHA0 to acidify its environment and to solubilize mineral phosphate is strongly dependent on its ability to produce gluconic acid. Moreover, we provide evidence that the formation of gluconic acid by CHA0 completely inhibits the production of PLT and partially inhibits that of DAPG. In the Δgcd mutant, which does not produce gluconic acid, the enhanced production of antifungal compounds was associated with improved biocontrol activity against take-all disease of wheat, caused by Gaeumannomyces graminis var. tritici. This study provides new evidence for a close association of gluconic acid metabolism with antifungal compound production and biocontrol activity in P. fluorescens CHA0.Plant growth-promoting rhizobacteria (PGPR) (36) are root-colonizing bacteria that enhance the performance of crop plants by several mechanisms. First, they antagonize plant-pathogenic fungi, mainly by the production of antimicrobial metabolites, but also by competition for iron or rhizosphere niches (9, 23, 24, 59). The biocontrol activity of many disease-suppressive microorganisms is also attributed to stimulation of host defense (induced systemic resistance). Other mechanisms by which these rhizobacteria directly promote plant growth are the production of phytohormones and the increase of nutrient, in particular phosphate, availability to plants (18, 37). Certain rhizobacteria are able to solubilize insoluble or poorly soluble mineral phosphates by producing acid phosphatases and organic acids, mainly gluconic acid (2, 34, 60). Some PGPR combine these different plant-beneficial activities and are able to suppress soilborne plant diseases, as well as to increase phosphate availability for plants (72).In fluorescent pseudomonads, gluconic acid production is catalyzed by periplasmic oxidation of glucose by membrane-bound glucose dehydrogenase (Gcd) (Fig. (Fig.1A)1A) (16, 43). In many gram-negative bacteria, the synthesis of gluconic acid has been shown to be dependent on pyrroloquinoline quinone (PQQ) as an enzymatic cofactor of the Gcd (1, 14). A consecutive oxidation reaction is mediated by gluconate dehydrogenase (Gad), which converts gluconic acid to 2-ketogluconate (Fig. (Fig.1A)1A) (11, 12, 44, 50). These enzymes should be of predominant importance for biocontrol soil pseudomonads, as major carbon sources provided by plant root exudates in the rhizosphere are made up of glucose (29, 69, 70). The two enzymes involved in glucose metabolism may have a substantial influence on general nutrient availability in the rhizosphere. First, Gcd and Gad affect glucose levels, and second, they may modulate the availability of soluble phosphates by controlling the amount of gluconic acid released into the rhizosphere. Furthermore, the production of gluconic acid might substantially change the rhizosphere pH. Therefore, Gcd and Gad enzymes produced by fluorescent pseudomonads are likely to be important for soil fertility and to impact the activities of other organisms living in the rhizosphere, e.g., fungal pathogens attacking the roots. Indeed, gluconic acid metabolism has already been linked to antifungal activity. Recently, Kaur et al. To this end, mutants of strain CHA0 carrying deletions in the gcd gene, encoding Gcd, and the gad gene, encoding Gad (Fig. (Fig.1),1), were created. The three in-frame deletion mutants, CHA1196 (Δgcd), CHA1197 (Δgad), and CHA1198 (Δgcd Δgad), were compared with their parental strain for the ability to produce organic acids, to solubilize inorganic phosphate, to produce the antifungal metabolites DAPG and PLT, to inhibit the growth of fungal pathogens, and to suppress different soilborne diseases. Background and Aim

Switzerland has a low post mortem organ donation rate. Here we examine variables that are associated with the consent of the deceased’s next of kin (NOK) for organ donation, which is a prerequisite for donation in Switzerland.

Methods and Analysis

During one year, we registered information from NOK of all deceased patients in Swiss intensive care units, who were approached for consent to organ donation. We collected data on patient demographics, characteristics of NOK, factors related to the request process and to the clinical setting. We analyzed the association of collected predictors with consent rate using univariable logistic regression models; predictors with p-values <0.2 were selected for a multivariable logistic regression.

Results

Of 266 NOK approached for consent, consent was given in 137 (51.5%) cases. In multivariable analysis, we found associations of consent rates with Swiss nationality (OR 3.09, 95% CI: 1.46–6.54) and German language area (OR 0.31, 95% CI: 0.14–0.73). Consent rates tended to be higher if a parent was present during the request (OR 1.76, 95% CI: 0.93–3.33) and if the request was done before brain death was formally declared (OR 1.87, 95% CI: 0.90–3.87).

Conclusion

Establishing an atmosphere of trust between the medical staff putting forward a request and the NOK, allowing sufficient time for the NOK to consider donation, and respecting personal values and cultural differences, could be of importance for increasing donation rates. Additional measures are needed to address the pronounced differences in consent rates between language regions.  相似文献

Chromatofocusing was utilized to separate rat lutropin isohormones. The pH gradients generated were highly reproducible, allowing accurate comparisons of isohormones in different elution profiles. Extracts of anterior pituitaries from intact male rats yielded at least seven species of immunoreactive lutropin after chromatofocusing. Five species exhibited apparent pI's in the range 8.97 to 9.25. Two additional peaks of rat lutropin were also observed: one in the void volume (pI greater than 9.8) and one which bound to the column but could be eluted with 1.0 M NaCl (pI less than 7.0). All seven lutropin isohormones were active in an in vitro bioassay. The biological-to-immunological (B:I) assay ratios were directly related to the apparent pI. The presence of both the basic and the acidic species of biologically active rat lutropin has not been previously observed with isoelectric focusing. Chromatofocusing should prove to be a valuable analytical tool in the isolation and characterization of gonadotropin isohormones.  相似文献

Pseudomonas fluorescens CHA0 suppresses black root rot of tobacco, a disease caused by the fungus Thielaviopsis basicola. Strain CHA0 excretes several metabolites with antifungal properties. The importance of one such metabolite, hydrogen cyanide, was tested in a gnotobiotic system containing an artificial, iron-rich soil. A cyanidenegative (hcn) mutant, CHA5, constructed by a gene replacement technique, protected the tobacco plant less effectively than did the wild-type CHA0. Complementation of strain CHA5 by the cloned wild-type hcn+ genes restored the strain's ability to suppress disease. An artificial transposon carrying the hcn+ genes of strain CHA0 (Tnhcn) was constructed and inserted into the genome of another P.fluorescens strain, P3, which naturally does not produce cyanide and gives poor plant protection. The P3::Tnhcn derivative synthesized cyanide and exhibited an improved ability to suppress disease. All bacterial strains colonized the roots similarly and did not influence significantly the survival of T.basicola in soil. We conclude that bacterial cyanide is an important but not the only factor involved in suppression of black root rot.  相似文献

AIMS: To assess the impact of the biocontrol strain Pseudomonas fluorescens CHA0 on a collection of barley rhizosphere bacteria using an agar plate inhibition assay and a plant microcosm, focusing on a CHA0-sensitive member of the Cytophaga-like bacteria (CLB). METHODS AND RESULTS: The effect of strain CHA0 on a collection of barley rhizosphere bacteria, in particular CLB and fluorescent pseudomonads sampled during a growth season, was assessed by a growth inhibition assay. On average, 85% of the bacteria were sensitive in the May sample, while the effect was reduced to around 68% in the July and August samples. In the May sample, around 95% of the CLB and around 45% of the fluorescent pseudomonads were sensitive to strain CHA0. The proportion of CHA0-sensitive CLB and fluorescent pseudomonad isolates decreased during the plant growth season, i.e. in the July and August samples. A particularly sensitive CLB isolate, CLB23, was selected, exposed to strain CHA0 (wild type) and its genetically modified derivatives in the rhizosphere of barley grown in gnotobiotic soil microcosms. Two dry-stress periods were imposed during the experiment. Derivatives of strain CHA0 included antibiotic or exopolysaccharide (EPS) overproducing strains and a dry-stress-sensitive mutant. Despite their inhibitory activity against CLB23 in vitro, neither wild-type strain CHA0, nor any of its derivatives, had a major effect on culturable and total cell numbers of CLB23 during the 23-day microcosm experiment. Populations of all inoculants declined during the two dry-stress periods, with soil water contents below 5% and plants reaching the wilting point, but they recovered after re-wetting the soil. Survival of the dry-stress-sensitive mutant of CHA0 was most affected by the dry periods; however, this did not result in an increased population density of CLB23. CONCLUSIONS: CLB comprise a large fraction of barley rhizosphere bacteria that are sensitive to the biocontrol pseudomonad CHA0 in vitro. However, in plant microcosm experiments with varying soil humidity conditions, CHA0 or its derivatives had no major impact on the survival of the highly sensitive CLB strain, CLB23, during two dry-stress periods and a re-wetting period; all co-existed well in the rhizosphere of barley plants. SIGNIFICANCE AND IMPACT OF THE STUDY: Results indicate a lack of interaction between the biocontrol pseudomonad CHA0 and a sensitive CLB when the complexity increases from agar plate assays to plant microcosm experiments. This suggests the occurrence of low levels of antibiotic production and/or that the two bacterial genera occupy different niches in the rhizosphere.  相似文献

Rifampin-resistant Pseudomonas fluorescens CHA0-Rif and mutants in which the regulatory gene algU (encoding sigma factor σE) or gacA (encoding a global regulator of secondary metabolism) was inactivated were compared for persistence in three nonsterile soils.
