Through the Mectizan® Donation Program, Merck & Co., Inc. has donated Mectizan (ivermectin, MSD) for the treatment of onchocerciasis worldwide since 1987. Mectizan has also been donated for the elimination of lymphatic filariasis (LF) since 1998 in African countries and in Yemen where onchocerciasis and LF are co-endemic; for LF elimination programs, Mectizan is co-administered with albendazole, which is donated by GlaxoSmithKline. The Mectizan Donation Program works in collaboration with the Mectizan Expert Committee/Albendazole Coordination, its scientific advisory committee. In 2005, a total of 62,201,310 treatments of Mectizan for onchocerciasis were approved for delivery via mass treatment programs in Africa, Latin America, and Yemen. Seventy-seven percent and 20% of these treatments for onchocerciasis were for countries included in the African Programme for Onchocerciasis Control (APOC) and the former-Onchocerciasis Control Programme in West Africa (OCP), respectively. The remaining 3% of treatments approved were for the six onchocerciasis endemic countries in Latin America, where mass treatment is carried out twice-yearly with the goal of completely eliminating morbidity and eventually transmission of infection, and for Yemen. All 33 onchocerciasis endemic countries where mass treatment with Mectizan is indicated have ongoing mass treatment programs. In 2005, 42,052,583 treatments of co-administered albendazole and Mectizan were approved for national Programs to Eliminate LF (PELFs) in Africa and Yemen. There are ongoing PELFs using albendazole and Mectizan in nine African countries and Yemen; these represent 35% of the total number of countries expected to require the co-administration of these two chemotherapeutic agents for LF elimination. In Africa, the expansion of existing PELFs and the initiation of new ones have been hampered by lack of resources, technical difficulties with the mapping of LF endemicity, and the co-endemicity of LF and loiasis. Included in this review are recommendations recently put forward for the co-administration of albendazole and Mectizan in areas endemic for LF, loiasis, and onchocerciasis.  相似文献

Copy number variations (CNVs) are large insertions, deletions or duplications in the genome that vary between members of a species and are known to affect a wide variety of phenotypic traits. In this study, we identified CNVs in a population of bulls using low coverage next‐generation sequence data. First, in order to determine a suitable strategy for CNV detection in our data, we compared the performance of three distinct CNV detection algorithms on benchmark CNV datasets and concluded that using the multiple sample read depth approach was the best method for identifying CNVs in our sequences. Using this technique, we identified a total of 1341 copy number variable regions (CNVRs) from genome sequences of 154 purebred sires used in Cycle VII of the USMARC Germplasm Evaluation Project. These bulls represented the seven most popular beef breeds in the United States: Hereford, Charolais, Angus, Red Angus, Simmental, Gelbvieh and Limousin. The CNVRs covered 6.7% of the bovine genome and spanned 2465 protein‐coding genes and many known quantitative trait loci (QTL). Genes harbored in the CNVRs were further analyzed to determine their function as well as to find any breed‐specific differences that may shed light on breed differences in adaptation, health and production.  相似文献

Genomic prediction utilizing causal variants could increase selection accuracy above that achieved with SNPs genotyped by currently available arrays used for genomic selection. A number of variants detected from sequencing influential sires are likely to be causal, but noticeable improvements in prediction accuracy using imputed sequence variant genotypes have not been reported. Improvement in accuracy of predicted breeding values may be limited by the accuracy of imputed sequence variants. Using genotypes of SNPs on a high‐density array and non‐synonymous SNPs detected in sequence from influential sires of a multibreed population, results of this examination suggest that linkage disequilibrium between non‐synonymous and array SNPs may be insufficient for accurate imputation from the array to sequence. In contrast to 75% of array SNPs being strongly correlated to another SNP on the array, less than 25% of the non‐synonymous SNPs were strongly correlated to an array SNP. When correlations between non‐synonymous and array SNPs were strong, distances between the SNPs were greater than separation that might be expected based on linkage disequilibrium decay. Consistently near‐perfect whole‐genome linkage disequilibrium between the full array and each non‐synonymous SNP within the sequenced bulls suggests that whole‐genome approaches to infer sequence variants might be more accurate than imputation based on local haplotypes. Opportunity for strong linkage disequilibrium between sequence and array SNPs may be limited by discrepancies in allele frequency distributions, so investigating alternate genotyping approaches and panels providing greater chances of frequency‐matched SNPs strongly correlated to sequence variants is also warranted. Pituitaries were removed from rams, wethers, and wethers that received Silastic implants containing 5 alpha-dihydrotestosterone (DHT), 17 beta-estradiol (E2) or DHT + E2. After homogenization and centrifugation (100,000 X g), aliquots of the supernatants were subjected to analytical gel filtration on Sephadex G-100 Superfine to separate native ovine luteinizing hormone (oLH) from its uncombined subunits. Immunoreactive oLH and oLH subunits were quantified in the elution profiles to examine the effects of castration and gonadal steroid administration on the intracellular levels of uncombined oLH subunits. Pituitaries from rams contained 1.41 +/- 0.26, 0.191 +/- 0.024, and 0.0246 +/- 0.0043 micrograms oLH, oLH alpha and oLH beta per mg tissue, respectively, which translated to oLH alpha/oLH and oLH beta/oLH molar ratios of approximately equal to 0.29 and approximately equal to 0.04. Castration decreased the concentrations of oLH and its subunits by approximately 50%, but did not significantly alter the oLH alpha/oLH and oLH beta/oLH molar ratios. All three steroid treatments further decreased the concentrations of oLH and oLH beta. Pituitaries from DHT-implanted wethers exhibited similar oLH alpha/oLH and oLH beta/oLH molar ratios to rams and unimplanted wethers. However, in E2- or DHT + E2-implanted wethers, there was a greater reduction in the concentration of native oLH than in the uncombined subunits. Thus, both the oLH alpha/oLH and oLH beta/oLH molar ratios were significantly higher in E2- or DHT + E2-implanted wethers than in the other groups. The apparent molecular sizes of oLH or its subunits were not significantly altered by castration or steroid administration. These results suggest that DHT and E2 decrease the concentrations of uncombined oLH beta as well as native oLH in the pituitary, but do not appear to alter the apparent molecular size of either oLH or its uncombined subunits However, because the levels of uncombined subunits were not decreased to the same degree as oLH in E2-implanted wethers, estrogens may affect the process of oLH subunit combination or may result in the production of molecular forms of oLH that are easier to dissociate.  相似文献

Human mammary medullary carcinoma cells (passages 16 to 21) were cultured for 2 days to allow for attachment, followed by 6 days of culture in either fetal calf serum, human cord blood, human amniotic fluid, or growth factors in the presence or absence of purified human alpha-fetoprotein (AFP). When growth factors were tested alone, only platelet-derived growth factor produced a significant increase in cell proliferation. Although up to 40% amniotic fluid had no effect on cell proliferation, human cord blood was two-fold more potent than fetal calf serum at similar concentrations. The addition of 10 ng/ml of platelet-derived growth factor increased the proliferative activity of human cord blood 1.5- to 2.5-fold. Ablation of endogenous AFP by affinity chromatography reduced the proliferative activity of cord blood by 75%. Similarly, the mitogenic activity of cord blood plus platelet-derived growth factor was reduced by 56% when AFP was removed. Purified AFP dose-dependently enhanced the proliferative activity of platelet-derived growth factor. This synergistic effect was specific for platelet-derived growth factor. We conclude that platelet-derived growth factor is a major growth factor controlling the proliferation of these tumor cells and that AFP may enhance growth factor proliferative activity and human mammary tumor growth.  相似文献

Ricinus communis agglutinin, a galactose-binding lectin, agglutinates phospholipid/qlycolipid vesicles, phospholipid/glycolipid/cholesterol vesicles, low density lipoprotein, and the branched polysaccharide gum arabic. The temperature dependence of the initial velocity of agglutination is similar in all cases, with a maximum at ~25°C, and a decreased rate at lower and higher temperatures. The extent of agglutination is temperature-independent for T ≤ 25°C, and decreases with increasing temperature for T > 25°C. After agglutination at 25°C, an increase in temperature results in deagglutination. This effect is reversible upon return to 25°C. Thus RCA undergoes a reversible temperature-dependent transition between an agglutinating form (≤25°C) and a less active form (> 25°C). These observations have important consequences in the interpretation of experiments which attempt to correlate lectin-binding with temperature-dependent properties such as membrane fluidity or receptor lateral organization in natural and model membranes.  相似文献
