A quantitative autoradiographic technique was combined with immunocytochemical staining to compare 3H-estrogen uptake in individual pituitary cell types 1, 14 or 50 days after castration in both male and female rats. Silver grains were counted over nuclei of immunocytochemically stained cells and means were computed for each cell type. The order of labelling intensity for all groups was gonadotropes greater than or equal to lactotropes = somatotropes greater than thyrotropes = corticotropes. In male rats 3H-estrogen uptake capacity in all of these cell types remained unchanged over the post-castration interval. Only gonadotropes from female rats demonstrated a significant change in estrogen uptake capacity over the intervals examined. Uptake in these cells increased by 137% between 1 and 50 days after ovariectomy. At both 14 and 50 days post-ovariectomy, gonadotropes concentrated significantly more radioactive label than either lactotropes or somatotropes. One day after castration, gonadotropes from females concentrated less 3H-estrogen than males while at 50 days after castration they concentrated significantly more than gonadotropes from male rats.  相似文献

The effects of luteinizing hormone (LH) (0, 100, 10,000 lU/ml) and follicle-stimulating hormone (FSH) (20 μg/ml) supplementation during in vitro maturation of slaughterhouse-derived oocytes on polar body formation and embryo development subsequent to in vitro fertilization and nuclear transfer were evaluated. Go-nadotropin supplementation of maturation medium in the presence of serum neither enhanced the proportion of oocytes forming a polar body nor significantly affected development following in vitro fertilization or nuclear transfer, except at the highest LH concentration. A very high concentration of LH (10,000 lU/ml) significantly decreased polar body formation, initial cleavage, and blastocyst development (P < 0.05). © 1993 Wiley-Liss, Inc.  相似文献

Using the dry-mount autoradiographic technique, a single population of cells within the rat epididymis, the clear cells, have been shown to bind [3H]aldosterone at a nuclear site. Competitive binding experiments demonstrated that aldosterone was more potent than desoxycorticosterone than testosterone in reducing the nuclear uptake of radioactive aldosterone. Furthermore, the other epididymal cells (principal and basal cells) in all regions of the epididymis were not significantly labelled; occasional labelling was noted in some endothelial and stromal cells. It is suggested that aldosterone may play a role in controlling the intracellular and transcellular movement of ions and water necessary for concentrating absorbed macromolecules in the clear cell.  相似文献

Estrogen induces reproductive behavior in lizards by acting on specific areas of the brain. Neural areas that selectively concentrate radioactivity after 3-h-estradiol administration were mapped using thawmount autoradiography. Major accumulations of hormone-concentrating cells occur in the medial preoptic area, basal hypothalamus, amygdala (n. vetromedialis telencephali) and in and around the torus semicircularis. Numerous smaller accumulations are also described and the topographical distribution is compared with that of avian and mammalian species.  相似文献

The damming of rivers has created hotspots for organic carbon sequestration and methane production on a global scale as the reservoirs intercept fluvial suspended and dissolved loads. To better understand how the C-biogeochemistry of a reservoir responds to watershed processes and evolves over time, Lake Decatur, located in the Intensively Managed Landscape Critical Zone Observatory (IML-CZO) was studied. Solid phase analyses (% organic C, C/N, δ13C, δ15N) of soils and sediments sampled from stream bank exposures, river suspensions, and the lake bottom were conducted to characterize organic C (OC) sources throughout the sedimentary system. Agriculturally-driven soil erosion rapidly altered lake bathymetry causing an evolution of sedimentary and OC deposition patterns, which in turn shaped where and when methane production occurred. A positive correlation between OC accumulation rate and porewater dissolved inorganic C (DIC) δ13C profiles indicates that methane generation is strongly influenced by OC burial rate. The sources of the lake bed particulate organic C (POC) have also evolved over time. Drowned vegetation and/or shoreline inputs were dominant initially in areas adjacent to the original river channel but were rapidly overwhelmed by the deposition of sediments derived from eroded agricultural soils. Eutrophication of the lake followed with the onset of heavy fertilizer application post-1960. This succession of sources is expected to be commonplace for reservoirs greater than?~?50–60 years old in agricultural settings because of the relative timing of tillage and fertilizer practices. The 13C/12C ratios of methane from Lake Decatur were more depleted in 13C than what is commonly expected for freshwater sedimentary environments. The 13C-depletion suggests that CO2-reduction is the dominant methanogenic pathway rather than the anticipated acetate dissimilation process. The isotopic observations reveal that commonly held assumptions about methane production and its C-isotopic signature in freshwater systems are over-simplified and not strictly applicable to this system.  相似文献

Michael reaction of the methoxymethyl-protected monodiazeniumdiolate of piperazine (MOM-PIPERAZI/NO) with 4-maleimidobutyric acid followed by its conversion to the N-hydroxy-succinimido ester produces a reagent capable of transferring the nitric oxide (NO)-donating diazeniumdiolate group to the terminal amines of the lysine residues contained in proteins. The reagent has been used to produce diazeniumdiolated bovine serum albumin (D-BSA) and diazeniumdiolated human serum albumin (D-HSA) containing 22 and 19 modified lysyl groups, respectively. Upon dissolution in pH 7.4 phosphate buffer at 37 degrees C, these albumin derivatives gradually released all of their contained NO (approximately 40 mol/mol of protein) with initial rates of about 30-40 pmol/min/mg and half-lives on the order of 3 weeks. This methodology is now available for use in exploiting the unique specific metabolic interactions of proteins to target NO therapy to specific physiological processes in vivo.  相似文献
