The motor, sensory, and postganglionic sympathetic neurons forming the left ulnar and right radial nerves of long-tailed macaques (Macaca fascicularis) and pigtailed macaques (Macaca nemestrina) were localized by the horseradish peroxidase method of tracing neuronal connections. The ulnar and radial motoneurons formed a longitudinal column of variable extent in the lateral part of the ventral horn. In most animals, the ulnar motoneurons extended between the caudal ends of the C7 and T1 segments; the radial motoneurons extended between the rostral level of the C4 and the middle part of T1 segments. Although there were areas of overlap in the spinal distribution of ulnar and radial motoneurons, the ulnar motoneurons were located more dorsally and dorsolaterally than were the radial motoneurons. In most animals, labelled sensory neurons whose axons run with the ulnar nerve occurred in the C8–T4 dorsal root ganglia, and those whose axons run with the radial nerve occurred in the C5–T3 ganglia. The radial sympathetic neurons were distributed in stellate through T7 paravertebral sympathetic ganglia, and the ulnar sympathetic neurons were distributed in stellate through T4 paravertebral sympathetic ganglia. Though the motor, sensory, and sympathetic neurons forming the ulnar and radial nerves had wide segmental distributions, all showed peak frequencies in two segments. The cross-sectional areas of the motor, sensory, and postganglionic sympathetic neurons forming the radial and ulnar nerves were measured in the animal that showed the greatest amount of labelling for each nerve. The ulnar and radial motoneurons had a similar range of sizes, with cross-sectional areas between 120 and 2,160 μm2. Most were smaller than 900 μm2. The sensory neurons forming the ulnar and radial nerves also displayed a similar range of sizes, measuring between 120 and 3,360 μm2 in cross-sectional area. Most neurons measured between 201 and 800 μm2. The ulnar sympathetic neurons measured between 120 and 840 μm2, and the radial neurons between 120 and 2,120 μm2. In both cases, most neurons measured between 120 and 600 μm2. The mean cross-sectional area for the radial sympathetic neurons was, however, larger than that for the ulnar sympathetic neurons.  相似文献

To assess the accuracy of species delimitation and phylogenetic relationships of the Aleochara fucicola species complex, we performed molecular phylogenetic analyses. Detailed micromorphological characters were also examined using scanning electron microscopy (SEM). The molecular data set included two mitochondrial (COI and COII) and three nuclear protein‐coding genes (CAD, EF1α and wg) for 19 populations of five species. Significant discordance was found between mitochondrial and nuclear gene trees. Two species, A. puetzi (Assing) and A. segregata Yamamoto and Maruyama, were not separated in the mitochondrial gene trees, but clearly were differentiated in the nuclear and concatenated gene trees. The SEM data also supported the separation of these two species, but an analysis of genetic divergence data did not. Adaptation to extremely harsh environments might reduce morphological variation in the A. fucicola species complex during the colonization of seashores. We present a multilocus phylogeny of the species complex. It suggests that the ancestor of the A. fucicola species complex occurred along the southern coasts of Northeast Asia, followed by dispersals to northern coasts.  相似文献

The DNA polymerase gene of Thermococcus marinus ( Tma ) contains an intein inserted at the pol-b site that possesses a 1611-bp ORF encoding a 537-amino acid residue. The LAGLIDADG motif, often found in site-specific DNA endonucleases, was detected within the amino acid sequence of the intein. The intein endonuclease, denoted as PI- Tma , was purified as a naturally spliced product from the expression of the complete DNA polymerase gene in Escherichia coli . PI- Tma cleaved intein-less DNA sequences, leaving four-base-long, 3'-hydroxyl overhangs with 5'-phosphate. Nonpalindromic recognition sequences 19 bp long were also identified using partially complementary oligonucleotide pair sequences inserted into the plasmid pET-22b(+). Cleavage by PI- Tma was optimal when present in 50 mM glycine–NaOH (pH 10.5), 150 mM KCl and 12 mM MgCl2 at 70 °C.  相似文献

A phylogenetic analysis of the tribe Liparocephalini Fenyes is presented based on morphological and molecular characters. The data set comprised 50 adult morphological characters, partial COI (907 bp), COII (366 bp) and 12S rDNA (325–355 bp), and nearly complete sequences of 18S rDNA (1768–1902 bp) for 21 species. Eighteen species of liparocephaline beetles from all eight genera and three outgroups, are included. The sequences were analysed separately and simultaneously with morphological characters by direct optimization in the program POY4 and by partitioned Bayesian analysis for the combined data. The direct optimization (DO) tree for the combined data under equal weighting, which also shows a minimum incongruence length difference value, resulted in a monophyletic Liparocephalini with the following patterns of phylogenetic relationships (outgroup ((Baeostethus, Ianmoorea) (Paramblopusa ((Amblopusa, Halorhadinus) (Liparocephalus, Diaulota))))). A sensitivity analysis using 16 different parameter sets for the combined data shows the monophyly of the liparocephalines and all its genera under all parameter sets. Bayesian analysis resulted in topological differences in comparison with the DO tree under equal weighting only in the position of the genus Paramblopusa and clade (Amblopusa + Halorhadinus), which were reversed. Historical biogeography and the stepwise evolutionary colonization of intertidal habitat in the Liparocephalini are discussed. Based on the biogeographical analyses, we hypothesize that the ancestor of the Liparocephalini occurred along the Panthallassan Ocean, the direct antecedent of the Pacific Ocean, followed by repeated dispersals to the Nearctic from the Palearctic. We also hypothesize that ancestors of the Liparocephalini appear to have arisen in the littoral zone of beaches and then colonized rocky reef areas in the low tidal zone later through high‐ to mid‐tide zones. © The Willi Hennig Society 2009.  相似文献

Red cell carbonic anhydrase types in Korean cattle were investigated by means of starch gel electrophoresis.
A carbonic anhydrase zone (X) located ahead zone F was found in 12 out of 502 Korean cattle.
The distribution of CA phenotypes in 490 Korean cattle was CA FS type 92 and CA SS type 398. The CA FF type has not yet been recognized in Korean cattle. The gene frequencies were CAF– 0.094 and CAF = 0.906. No statistically significant differences were observed between gene frequencies in cattle from six different Korea provinces.  相似文献

Cell proliferation is a delicately regulated process that couples growth signals and metabolic demands to produce daughter cells. Interestingly, the proliferation of tumor cells immensely depends on glycolysis, the Warburg effect, to ensure a sufficient amount of metabolic flux and bioenergetics for macromolecule synthesis and cell division. This unique metabolic derangement ould provide an opportunity for developing cancer therapeutic strategy, particularly when other diverse anti-cancer treatments have been proved ineffective in achieving durable response, largely due to the emergence of resistance. Recent advances in deeper understanding of cancer metabolism usher in new horizons of the next generation strategy for cancer therapy. Here, we discuss the focused review of cancer energy metabolism, and the therapeutic exploitation of glycolysis and OXPHOS as a novel anti-cancer strategy, with particular emphasis on the promise of this approach, among other cancer metabolism targeted therapies that reveal unexpected complexity and context-dependent metabolic adaptability, complicating the development of effective strategies. Reactive oxygen species are produced during anaerobic exercise mostly by Fe ions released into plasma and endothelial/muscle xanthine oxidase activation that generates uric acid (UA) as the endpoint metabolite. Paradoxically, UA is considered a major antioxidant by virtue of being able to chelate pro-oxidative iron ions. This work aimed to evaluate the relationship between UA and plasma markers of oxidative stress following the exhaustive Wingate test. Plasma samples of 17 male undergraduate students were collected before, 5 and 60 min after maximal anaerobic effort for the measurement of total iron, haem iron, UA, ferric-reducing antioxidant activity in plasma (FRAP), and malondialdehyde (MDA, biomarker of lipoperoxidation). Iron and FRAP showed similar kinetics in plasma, demonstrating an adequate pro-/antioxidant balance immediately after exercise and during the recovery period (5–60 min). Slight variations of haem iron concentrations did not support a relevant contribution of rhabdomyolysis or haemolysis for iron overload following exercise. UA concentration did not vary immediately after exercise but rather increased 29% during the recovery period. Unaltered MDA levels were concomitantly measured. We propose that delayed UA accumulation in plasma is an auxiliary antioxidant response to post-exercise (iron-mediated) oxidative stress, and the high correlation between total UA and FRAP in plasma (R-Square = 0.636; p = 0.00582) supports this hypothesis.  相似文献

The macrolide antibiotic bafilomycin and the related synthetic compound SB 242784 are potent inhibitors of the vacuolar H+-ATPases (V-ATPase). It is currently believed that the site of action of these inhibitors is located on the membrane bound c-subunits of V-ATPases. To address the identification of the critical inhibitors binding domain, their specific binding to a synthetic peptide corresponding to the putative 4th transmembrane segment of the c-subunit was investigated using fluorescence resonance energy transfer (FRET), and for this purpose a specific formalism was derived. Another peptide of the corresponding domain of the c′ isoform, was checked for binding of bafilomycin, since it is not clear if V-ATPase inhibition can also be achieved by interaction of the inhibitor with the c′-subunit. It was concluded that bafilomycin binds to the selected peptides, whereas SB 242784 was unable to interact, and in addition for bafilomycin, its interaction with the peptides either corresponding to the c- or the c′-subunit isoforms is identical. Since the observed interactions are however much weaker as compared to the very efficient binding of both bafilomycin and SB 242784 to the whole protein, it can be concluded that assembly of all V-ATPase transmembrane segments is required for an efficient interaction.  相似文献
