Immature rats were made copper deficient by feeding them a low (< 1 p. p. m.) copper diet. During the gestation and lactation periods their dams consumed the same diet. Controls received a dietary supplement of 10 p. p. m. copper. At approx 7 weeks of age, the deficient animals exhibited signs of neurological dysfunction and gross lesions of the brain. Cytochrome oxidase activity and copper content of the liver and brain were used as criteria of copper status and confirmed the existence of severe deficiency. The whole brains minus cerebella of the deficient animals contained approx 30% less dopamine and norepinephrine than those of the controls. The tyrosine hydroxylase activity was depressed more than 25% in the copper deficient brains while the superoxide dismutase activity was lowered more than 35%. There was a high correlation between the chief criterion of copper status, liver cytochrome oxidase activity, and the brain concentrations of dopamine, norepinephrine and tyrosine hydroxylase activity. The decrease in activity of tyrosine hydroxylase was sufficient to account for the lowered concentrations of the catecholamines.  相似文献

The mechanism by which the utilization of transferrin-bound iron is linked with cellular metabolism was investigated using rabbit reticulocytes and bone marrow cells. The rate of metabolism was altered by the use of inhibitors which act at different sites in the metabolic pathway (NaF, sodium fluoroacetate, rotenone, 2,4-dinitrophenol, NaCN) and by the addition of metabolic substrates (inosine, sodium pyruvate, sodium lactate). Measurements were made of the rates of iron and transferrin uptake and, in many of the experiments, of cellular ATP and NADH concentrations. The results showed that there was a significant correlation between the rate of iron uptake and the ATP concentration of the cells, but no correlation was found with the NADH concentration. The rate of transferrin uptake was inhibited to a lesser degree than that of iron uptake, and only when the ATP concentration had fallen below that necessary to inhibit iron uptake. It is concluded that the rate of uptake of transferrin-bound iron by immmature erythroid cells is dependent on the intracellular concentration of ATP but is independent of the NADH concentration.  相似文献

Hyperalphalipoproteinemia, characterized by increased plasma concentrations of apoA-I and of HDL lipid and protein, was observed in rats treated with triiodothyronine (T(3)) for 7 days. The increase in the plasma HDL apoproteins was general for apoC, apoE plus A-IV, and apoA-I, as determined by isoelectric focusing. Hypotriglyceridemia, characterized by decreased concentrations of VLDL and apoB, was also observed in the hyperthyroid state. Although in the mildly hypothyroid animals (propylthiouracil-treated), hepatic metabolism of free fatty acid is shifted toward esterification to triglyceride and VLDL formation, as we reported previously, plasma HDL and apoA-I concentrations were not different from control plasma values, while the d 1.006-1.063 g/ml (IDL + LDL) lipoprotein fraction tended to be increased. In general, the proportion of apoE in the (IDL + LDL) fraction of the hypothyroid rat was greater than in controls and hyperthyroid animals, while the proportion of apoE tended to be lower in VLDL from both hypo- and hyperthyroid rats than in VLDL from controls. An enhanced release of apoA-I by perfused livers isolated from rats treated with T(3) was also observed; this enhanced output of apoA-I may explain, in part, the hyperalphalipoproteinemia observed in these rats. The depressed net output of apoA-I in vitro by perfused livers from rats treated with propylthiouracil (PTU) was not expressed in a statistically significant diminished plasma concentration of HDL or apoA-I in the intact animals. Treatment with T(3) also resulted in modification of the content of essential fatty acids in various lipid classes. Linoleic acid residues were significantly reduced and arachidonic acid content was increased in plasma phospholipids and esterified cholesterol in T(3)-treated rats. However, the relative fatty acid composition of unesterified fatty acids and triglyceride fatty acids was not altered by T(3) treatment. PTU treatment had no effect on fatty acid distribution in any of the plasma lipids. Secretion of biliary lipids was increased in perfused livers from T(3)-treated rats, while treatment with PTU did not affect release of lipids in the bile. Treatment of the whole of aSinapis alba plant with supplementary far-red light (FR), in back-ground white light (WL), induces a rapid increase in stem extension rate. This rapid increase is regulated by the light environment of the stem itself. Supplementary FR to the stem increases extension rate after a lag period of 10–15 min. A lag period of 3–4 h follows FR irradiation of the leaf, before an increase in extension rate is detectable. When the stem is given supplementary FR, the change in extension rate which is induced increases with increasing FR fluence rate, and with decreasing phytochrome photoequilibrium. There is no difference between the effects of supplementary FR max 719 nm and supplementary FR max 739 nm for these relationships. The increase in extension rate induced by supplementary FR is reversed by an increase in the fluence rate of red light (R). The effects of temperature and photosynthetically active radiation levels on photorespiration were investigated in Panicum milioides Nees ex Trin. and Panicum schenckii Hack., species known to have low photorespiration rates and other characteristics intermediate between C3 and C4 species. Comparisons were made with the C3 grass species tall fescue (Festuca arundinacea Schreb.). An increase in temperature from 20 to 35 C raised photorespiration from 7.3 to 10.2 milligrams per square decimeter per hour in tall fescue, but the increase in P. schenckii was less than 1 milligram per square decimeter per hour. Increases in temperature caused much less change in CO2 compensation concentration in P. milioides and P. schenckii than in tall fescue, values of 160 microliters per liter being obtained in tall fescue at 40 C compared to about 40 microliters per liter for P. milioides and P. schenckii. Photorespiration in P. schenckii increased by only about 1 milligram CO2 per square decimeter per hour as the photosynthetically active radiation level was raised from 100 to 2,000 microEinsteins per square meter per second. Loss of CO2 into CO2-free air actually decreased from 2.2 to 1.0 milligrams per square decimeter per hour as the radiation level was raised from 100 to 1,100 microEinsteins per square meter per second but tended to rise again at 2,000 microEinsteins per square meter per second. In contrast, photorespiration in tall fescue tripled with radiation level over the same range, reaching a maximum value of 7.2 milligrams per square decimeter per hour as determined by extrapolation of the CO2 response curves to zero CO2. The CO2 compensation concentration in tall fescue was nearly insensitive to photosynthetically active radiation above 140 microEinsteins per square meter per second but, in P. milioides and P. schenckii, it decreased from values of 69 and 62 microliters per liter, respectively, to values of 21 and 16 as the radiation level was increased from 50 to 1075 microEinsteins per square meter per second. Interpolation of CO2-response curves showed that an increase in photosynthetically active radiation level from 100 to 2,000 microEinsteins per square meter per second reduced the CO2 compensation value of P. schenckii from 38 to 19 microliters per liter. Data from these experiments indicate reduced photorespiration or a CO2-recycling mechanism in P. milioides and P. schenckii which causes apparent photorespiration to be nearly insensitive to temperature in the 20 to 35 C range and to decrease at high radiation intensities.  相似文献

Rat liver tryptophan pyrrolase haem is maximally depleted at 30 min after administration of a 400 mg/kg dose of 2-allyl-2-isopropylacetamide. This depletion lasts for 24 h, by which time 5-aminoleevulinate synthase activity becomes maximally enhanced. 2. though the above maximum depletion of pyrrolase haem (at 0.5h) is also produced by a 100 mg/kg dose of the porphyrogen, this does not enhance synthase activity at 24 h. It and smaller doses, however, cause a smaller but earlier enhancement of synthase activity (maximum at 2 h) and produce a similarly short-lived deplation of pyrrolase haem. 3. The depletion of pyrrolase haem and the enhancement of synthase activity by the porphyrogen are inhibited by compound SKF 525-A and phenazine methosulphate, and are potentiated by nicotinamide but not by phenobarbitone. Phenazine methosulphate and nicotinamide also exert opposite effects on hexobarbital sleeping-time. 4. 2-Allyl-2-isopropylacetamde also the depletes pyrrolase haem in vitro. It does so in liver homogenates of control rats in the presence, and in those of phenobarbitone-treated rats in the absence of added NADPH. 5. A discussion of the present results in relation to previous work with other haemoproteins suggests that, whereas cytochrome P-450 (haem) is primarily involved in the production of the active (porphyrogenic) metabolite(s) of 2-allyl-2-isopropylacetamide, the haem pool used by tryptophan pyrrolase may play an important role in the effects of this compound on haem biosynthesis.  相似文献

The concept of molecular complementarity is defined, and a theorem given which describes the symmetry required of self-complementary surfaces. Helicotylenchus oleae n. sp. and H. neopaxilli n. sp., from olive roots and soil in Italy, are described and illustrated. Helicotylenchus oleae can be distinguished from the related species H. canadensis and H. tunisiensis especially by the smaller styler, its distinctive tail shape, and a tail longer than one anal body width. Helicotylenchus neopaxilli differs from the close species H. paxilli by having a conical, anteriorly truncated labial region, shorter stylet, and phasmids always anterior to level of anus. Also illustrated and discussed are histopathological changes within feeder roots of olive caused by the feeding activity of the semi-endoparasitic H. oleae.  相似文献
